1.Expression of aquaporin1 in human amniotic membrane and its significance

Objective:To investigate the expression,location and significance of aquaporin1(AQP1) in the tissue of amniotic membrane.Methods:Immunohistochemical method was used to examine the expression of aquaporin1 in the tissue of amniotic membrane of 12 normal full-term pregnant women,11 full-term pregnamt woman with oligohydramnios and 9 with polyhydramnios.Result:Aquaporin1 was expressed in cuboidal epithelial cells of amniotic membrane,and mainly in cytoplasm.There was but a little expression of aquaporin1 in fibroblast of amniotic membrane tissue.There was statistically significant difference of the AQP1 expression between oligohydramnios,polyhydramnios and normal full-term pregnancy women integrative optical density in amniotic membrane tissue,respectively(P
2.Investigation of the expression of vascular endothelial growth factor-C and high risk human papillomavirus in different stages of human cervical lesions

Objective To study the pathogenetic role of HPV infection and vascular endothelial growth factor-C in the development of cervical carcinomas. Methods HPV16 DNA was tested in cervical intraepithelial neoplasia(CIN) 11I, cervical carcinomas and norma1 cervical tissue by PCR.and the expression of vascular endothelial growth factor-C protein was examined by immunohistochemistry. Results The expression of VEGF-C became stronger and stronger with the progression of cervical diseases from chronical inflammation to CIN and to invasive carcinoma of cervix uteri, but no expression in normal cervical tissue. There was significant correlation between HR-HPV and VEGF-C expression in CIN and cervical carcinomas(odds ratio are 19.12 and 20.49; 95 % confidence interval are 2.31-157.8 and 3.28-226.09). Conclusions The expression of VEGF-C, maybe a sensitive maker, is closely related to HR-HPV infection.
3.Changes of the expressions of endoplasmic reticulum chaperones GRP78 and caspase-9 and-12 in fetal rats with intrauterine distress

Objective:To investigate the role of glucose-regulated protein 78(GRP78) and caspase-9 and-12 in intrauterine distress-induced hypoxic-ischemic cerebral damage in fetal rats.Methods:The fetal rat model of intrauterine distress was constructed and the fetal rats were randomly divided into a normal,a sham operation and an ischemia-reperfusion(IR) group.Neuron apoptosis was analyzed by in situ end-labeled DNA(TUNEL).The expressions of caspase-9 and-12 and GRP78 proteins in the hippocampus CA1 area were detected by immunohistochemical staining.Results:Ischemia-reperfusion damage induced classic neuron apoptosis and the number of the apoptotic neurons in the hippocampus CA1 area increased with the progression of reperfusion.The expressions of GRP78 and caspase-9 and-12 were weak in the normal and sham operation group.In the IR group,the expression of GRP78 reached the peak value 3 hours after the reperfusion and then decreased gradually;the intensity of caspase-12 was increased rapidly while that of caspase-9 elevated very little within 3 hours,but both reached the peak value at 12 hours.Conclusion:Intrauterine distress-induced hypoxic-ischemic cerebral damage in fetal rats may trigger the homeostatic control system in the endoplasmic reticulum through the increased expression of GRP78.The apoptotic pathway mediated by caspase-12 in the endoplasmic reticulum may be one of the mechanisms underlying cerebral ischemic injury.
4.Perinatal outcome analysis of different glycosylated hemoglobin levels in gestational diabetes mellitus

Objective To study and analyze the perinatal outcomes of different glycosylated hemoglobin levels in gestational diabetes mellitus (DM).Methods 53 pregnant women diagnosed gestational diabetes in our hospital were selected as the observation group,and 53 pregnant women without gestational diabetes were selected as the control group.The two groups were followed up to terminate pregnancy.The glycosylated hemoglobin level was detected by HPLC.The postpartum hemorrhage,pregnancy induced hypertension syndrome and urinary tract infection and other complications were compared between the two groups.And the premature delivery,polyhydramnios,fetal macrosomia,intrauterine distress,premature rupture of membranes and other adverse pregnancy outcomes were compared between the two groups.According to the different HbA1 c levels in the observation group,they were divided into group A (HbAlc < =6%),group B (HbAlc between 6.1% ~ 6.5%),group C (HbAlc between 6.6% ~7.0%) and group D (HbAlc > 7%),the incidences of adverse pregnancy outcome were compared among four groups.Results The incidence rates of postpartum hemorrhage,pregnancy induced hypertension and urinary system infection of the observation group (18.86%,13.21%,11.32%) were higher than those of the control group (3.77%,1.89%,3.77%),the differences were statistically significant (x2 =11.356,9.179,4.085,all P < 0.05).The incidence rates of premature delivery,polyhydramnios,fetal macrosomia,intrauterine distress,premature rupture of membranes of the observation group (9.43%,9.43%,11.32%,13.21%,15.09%) were higher than those of the control group (1.89%,1.89%,1.89%,3.77%,5.66%),the differences were significant (x2 =5.323,5.323,7.207,5.735,4.781,all P < 0.05).The higher the level of glycosylated hemoglobin,the higher the incidence rates of adverse pregnancy outcomes,such as premature delivery,polyhydramnios,fetal macrosomia,fetal distress.And the incidence rate of premature rupture of membranes had no significant correlation with glycated hemoglobin levels (r =0.955,P < 0.05).Conclusion Glycosylated hemoglobin as an important index of gestational diabetes,its change has great influence on the outcome of pregnancy,HbAl c level higher,adverse pregnancy outcomes and the high incidence of clinical pregnancy,should strengthen the HbAlc test,in order to take timely and effective measures to reduce the incidence of adverse pregnancy outcomes and ensure the safety of mother and child.
