1.Autophagy facilitates breast cancer cells to resist tamoxifen by p38/c-Jun pathway

Objective To investigate the effects of autophagy on breast cancer cells to tamoxifen resist-ance.Methods Immunofluorescence was used to count the proportion of the cells with autophagy in MCF -7 and MCF-7/TAMR,as well as ,the average of autophagosome in each cells .Western blotting was used to detect the different expressions of p38,c-jun and LC3 in MCF-7 and MCF-7/TAMR.Stepwise selection was used to detect the duration of resistance to different doses of tamoxifen when upregulating or downregulating of LC 3.West-ern blot was facilitated to investigate changes of activity of p 38/c-jun.Rusults Compared with MCF -7,the proportion of the cells with autophagosome in MCF -7/TAMR was markedly increased ,the same as the average of autophagosome in each cells .Compared with MCF-7,LC3,especially LC3IIprotein expressive levels in MCF -7/TAMR was significantly increased .The expressive level of LC3 during the induction process of TAM resistance was negative correlated with resistant cell lines .The expressions of phosphorylated p 38 and c-Jun in MCF-7/TAMR were significantly increased .Conclusion Autophagy promotes breast cancer cells to resistant to tamox-ifen,due to downregulation of the expression of autophagy related proteins which may inhibit the generation of re -sistant breast cancer cells .This may be a viable treatment strategy for refractory breast cancer .
2.Epigenetic regulation of estrogen receptor-αin breast cancer

Recently,in the management of estrogen receptor(ER)positive breast canner,endocrine thera-py is the main treatment method;and estrogen receptor -α( ER-α) has obtained popularity as one of the most common targets .There are many known risk factors for ER function in breast canner such as ER gene mutation ;unbalanced microenvironment;signal transduction pathway disorders .While more and more attention is paid to the changes in ER expression and function induced by epigenetic aberrations .ER gene promoter methylation induces down regulation and deletion in ER expression ,which has not only a most highly incidence in epigenetic aberra-tions,but also a widen degree of research .Histone modification could influence ER -αexpression and function , either.However,the special mirco RNAs(miRNAs)are also implicated in the regulation of ER -α,and affect the therapy of tamoxifen in breast tumor .
3.RASSF1A and RASSF1A methylation′s relationship with breast cancer

Breast cancer is one of the most common malignant tumors in women.Early diagnosis plays a very important role in the treatment of breast cancer.Recently,with the development of epigenetics,it gives us a new orientation for early detection for cancers.The most understood mechanism of epigenetics is DNA methyla-tion.Ras associated domain family 1A gene(RASSFlA)promoter methylation occurres in tumor′s early stage,and have a statistically concern with tumor′s TNM stage,invasiveness and outcomes.Moreover,DNA methylation can be reversed,so RASSF1A can be a promising candidate for early breast cancer detection biomarker,and can be a potential orientation for breast cancer treatment.
4.Content Determination of Dextran70 in Compound Dextran70 Eye Drop by HPLC-RID

OBJECTIVE:To establish a method for determination of dextran70 in Compound dextran70 eye drop. METHODS:HPLC-RID was performed on the column of Phenomenex PolySep-GFC-P 4000 with the mobile phase of 0.7% Na2SO4 solution. The flow rate was 0.5 ml/min,column temperature was 35 ℃ and injection volume was 50 μl,and the inner temperature of refrac-tive index detector was 40 ℃. RESULTS:The linear range of dextran70 was 0.1-5.0 mg/ml(r=0.999 5);RSDs of precision,sta-bility and reproducibility tests were lower than 2%;average recovery was 97.45%-101.76%(RSD=0.57%,n=9). CONCLU-SIONS:The method is simple,fast and reliable with high separation and short analysis time,and can be used for the content deter-mination of dextran70 in Compound dextran70 eye drop.
5.Observations on the Efficacy of Auricular-plaster Therapy plus Medicine in Treating Heroin Addiction

Objective:To investigate the curative effect of auricular-plaster therapy on heroin addiction. Methods: Sixty cases of heroin addiction were treated by auricular-plaster therapy in cooperation with methadone and another 60 cases with methadone as a control. The treatment lasted 10 days. Results and conclusion: The results showed that the curative effect of auricular-plaster therapy plus methadone on heroin addiction was significantly better than that of methadone in the control group(P<0.01). The rate of drug relapse was also significantly lower in the former(P<0.01). It is considered that auricular-plaster therapy in cooperation with methadone is now an ideal therapeutic method for drug withdrawal.
6.Treatment of 100 Cases of Children's Nocturnal Enuresis with Acupoint Sticking Therapy

Daiwenjiu Gao plasters were stuck onto the following two groups of acupoints,group 1 including acupoints Guanyuan (CV 4), Shenshu (BL 23) and Zusanli (ST 36) and group2 including acupoints Zhongji (CV 3), Pangguangshu (BL 28) and Sanyinjiao (SP 6), to treat 100 cases of infantile enuresis, the results showed that 92 cases were cured, 6 cases got remarkable effect, 2 cases got effect, with total effective rate being 100%.
7.Clinical Application of CT-guided Percutaneous Pediculoplasty for Treatment of Vertebral Pedicular Osteolytic Metastases

Objective To explore the methods and clinical application of percutaneous pediculoplasty in the treatment of vertebral pedicular osteolytic metastases.Methods Percutaneous pediculoplasty was performed in 18 patients with (23 Vertebral pedicles ) osteolytic metastases,3 cases localized in thoracic vertebral pedicles and 15 cases in lumbar vertebral pedicles,and two vertebral pedicles involved in 5 cases . Under Multislice helical CT guidance ,2.5~5ml of polymethylmethacrylate ( PMMA ) mixture (ratio of polymer,moner,ultrafluid iodinated oil was 20g:10ml:3ml) was injected into pathologic pedicles in step by step after successful puncture using 11-13G needles. All cases had been followed for 1~18 months.Results All cases were successful punctured. Pain relieved in all cases after percutaneous pediculoplasty.7 cases had cement leakage,among them,cement leaked into tumor tissue in 5 cases(2 cases with leakage into intervertebral foremen),nerve radicular pain occurred in one case,the pain disappeared 3 days later after intravenous mannitol and lower dose dexamethasone drips;the cement leaked into vertebral canal a little without clinical symptom in one case.Conclusion CT-guided percutaneous pediculoplasty is a minimally effective and safe method for osteolytic vertebral pedicular metastases in treatment of pain and enhancement of spinal stability.
8.Clinical observation on rehabilitation of extremity motor function in the hemiplegic patients with stroke by acupuncture

ObjectiveTo explore an effective rehabilitation method to treat hemiplegic patients with stroke by acupuncture.Methods62 hemiplegic patients were treated by Opposing needling and Non-opposing needling twice a day in alternate way.The motor function of patients was assessed with Brunnstrom Assessment and activities daily living(ADLs) were assessed with Barthel Index revised.ResultsAt 3 months after the treatment,The rate of motor function improved above Ⅳ-Ⅳ level accounted for 62.90%,that of ADL improved 60 points accounted for 56.45%.There were significant difference in Brunnstrom level among pre-treatment and 1,2 months after the treatment(P<0.01),but no significant difference between 2 and 3 months (P>0.05).ConclusionsOpposing needling and Non-opposing needling used twice a day in alternate way can promote the recovery of motor function and ADL of hemiplegic patients with stroke,especially early (less than 2 months) recovery.
9.Association between single nucleotide polymorphism in IL-23R gene and ankylosing spondylitis in Chinese Han population

Objective To study the single nucleotide polymorphisms (SNPs) in IL-23R gene in Chinese Han population with ankylosing spondylitis (AS). Methods SNPs rs11209026, rs1343151, rs11209032 and another three SNPs near them based on their physical distances were genotyped by PCR-directed sequencing. Hardy-Weinberg equilibrium, genotypes and allele frequency analysis were analyzed by SPSS 13.0. Linkage disequilibrium and haplotype analysis were carried out by SHEsis software. Results The difference of genotypes of rs11209032 and the difference of genotypes and allele frequencies of rs6677188 between patients and controls were statistically significant (P<O.01) ; The two SNPs rs11209032 and rs6677188 had strong linkage disequlibfium (D'=0.925, r2=0.561 ). Haplotype analysis had shown a higher proportion of GAC haplotype in patients and a higher proportion of GTC haplotype in controls. Conclusion These results suggest that IL-23R polymorphisms is associated with susceptibility to AS in Chinese Han population and IL-23R gene may be a susceptible gene of AS.
10.Effects of Chronic Manganism on Hearing and Cochlear Cells in Rats

Objective To study the effects of chronic manganism on hearing and cochlear cells in rats by using animal model of chronic manganism .Methods Sixty adult SD rats were randomly divided into Mn - exposed and controlgroups.RatsweretreatedwithMnCl24H2O(100mg·kg -1·d-1)ordeionizedwaterbygastricperfusion, lasted for 12 weeks .The Mn concentration in peripheral blood was measured respectively at 4 weeks ,8 weeks and 12 weeks after treatment .At 12 weeks after treatment ,the auditory brainstem response was recorded ,the hair cells morphology and counting were examined by stretched preparation of basilar membrane stained with FITC -phalloi-din ,and the spiral ganglion cells morphology and counting were studied by HE staining ,the ultrastructure changes of hair cells and spiral ganglion cells were detected by transmission electron microscopy .Results The blood Mn concentration increased gradually with time after treatment .ABR thresholds at 4 ,8 ,16 ,24 and 32 kHz were sig-nificantly increased at 12 weeks after treatment ,especially in the high-frequency range .Morphological study at 12 weeks after treatment showed loss of outer hair cells ,mainly in the basal turn of the cochlea ,and decreased number of spiral ganglion cells .The ultrastructure changes of outer hair cells and spiral ganglion cells included the break -ups ,disappearance or vacuolar change of mitochondria cristas .Conclusion Our data demonstrate that chronic man-ganism can cause loss of outer hair cells and spiral ganglion cells in cochlear in rats ,leading to hearing loss .
