1.IN VITRO STUDY ON EFFECTS OF ZINC ON BONE DEVELOPMENT

Objective: To explore the effects of zinc on bone development in vitro. Methods: Mice of 16 gestation were killed by snapping cervical vertebra, and the front limbs were cut off and cultured in basic medium. Which contained 20 ?mol/L Zn 2+ (control group). In Zn deprived group, the basic medium was added TPEN to final concentration 20 ?mol/L. In other three groups, the basic medium was added ZnSO 4 to final concentration of Zn 2+ at 45, 70, and 120 ?mol/L respectively. Results: 1. Compared with control limb(uncultivated),the test limb(cultivated for 6 d)showed longer bone length and higher bone density.Histological analysis showed active bone cell differentiation, proliferation, increased bone trabecula.2.Compared with control group, the Zn-deprived and Zn 2+ 120 ?mol/L group expressed decreased osteocalcin (OC) and 45 Ca content and lower activity of AKP; Zn 2+ 45 ?mol/L and 70 ?mol/L group presented increased OC and 45 Ca content and higher activity of AKP.3.Compared with the control group, Zn-deprived and Zn 2+ 120?mol/L group showed shorter bone length and lower bone density; Zn 2+ 45 ?mol/L and 70?mol/L group showed longer bone length and higher bone density. Conclusion: Zinc is involved in bone metabolism and growth; both zinc-deficiency and zinc-excess can alter bone growth and normal metabolism.
2.Trans-articular movable external fixation of severely comminuted and open tibial Pilon fractures

Objective To investigate the results of comminuted or open tibi al Pilon fractures treated with a joint bridging movable external fixator. Methods From January 2000 to May 2002, fifteen patients with either severely comminuted or open Pilon fractures were treated with self-designed joint bridging movable external fixators. In some patients, the external fixation was combined with li mited open reduction and Kirschner wire internal fixation. Results All patients were followed up. The average period of following up was 10 months (range 6 to 2 2 months). The fractures were externally fixed for averagely 3 months. The inter val between fracture and bony union was averagely 4.5 months. There was no nonun ion occurred. Two delayed unions were further treated with bone grafting and one of them was stabilized with a plate. All the 9 open fractures were infection fr ee, so were the closed injuries. There was no skin sloughs found in all cases bu t minor pin tract infection was found in two cases. Range of motion at last foll ow-up examination was excellent in five patients, good in seven, fair in two, an d poor in one. One patient required ankle arthrodesis for posttraumatic arthriti s. Conclusion The results of the severely comminuted or open tibial Pilon fractu res treated with joint bridging movable external fixators were fairly satisfying . The fixator can firmly stabilize the fracture while ankle movement can be resu med early. The operation is less-invasive, and is with higher union rate and few er complications.
3.Antiinflammatory Effects of Alcohol Extract of Daoguaniou (Caesalpinia sepiaria var. pubescens)

An tiinflammatory effects of alcohol extract of Caesalpinia sepiarla Roxb. var. pubescent Tang et wang were shown by ip iujection. The extract significantly decreased xylene induced swelling of mice ear; carrageenin produced edema of hind paw in rats, and decreased volume of pleural exudate on carrageenin-induced acute pleurisy of rats. It also inhibited egg white induced arthritis iu rats.LD50 value is 303.7mg/kg by ip in mice, confidence limit of 95% is 296.5 ? 189.4mg/kg.
4.A Comparative Study on Removal Methods of ECG Baseline Wandering

Baseline shift has severe effects on the analysis and processing of ECG. Filter and baseline polynomial-fitting are the main methods for removing baseline interference. This paper discussed the filtering ways of morphology filter,median filter,Linear-phase finite impulse response (FIR) filter,wavelet transform,morphology and wavelet combined method,median filter and wavelet combined method,comparing with the approach of baseline polynomial fitted way which composed of differential method and coordinates method. Experimental results showed that the poly-fitting approach are more efficient,had certain adaptive quality and little distortion,while filter ways were easy to implement but not accurate enough. As for real time and accuracy,polynomial fitted way has an advantage over filtering ways. Under the circumstances of having great difficulty in extracting baseline points,it is more appropriate to use filter ways to eliminate baseline wandering. This paper compared the methods in common use for suppressing baseline wander so as to help users to choose the proper way.
5.Effect of basic fibroblast growth factor on the formation of blast-colony-forming cell derived from embryonic stem cells

BACKGROUND:Some studies show that basic fibroblast growth factor(bFGF)strongly expresses dudng the process of embryonic stem cells differentiation into hematopoietic stem cell.yolk sac blooding.and fetal liver hematopoiesis.OBJECTIVE:To study the regulation of bFGF on the blast-colony-forming cell(BL-CFC)by adding bFGF in the medium of embryoid body generation.METHODS:The third to fifth generations of the pdmaw mouse embryonic fibroblasts were recovered,and then incubated with the DMEM medium containing mitomvcin C for 2.5 hours in order to lose the proliferative capacity.Then cells were suspended into single cell by trypsinization and inoculated in the gelatin-coated bottle at the density of 10×104/cm2.After culturing for 24 hours,mouse embryonic stem cells(mESC)of D3 were recovered and placed on the feeder layer cells.According to the composition of medium in embryoid body generation.mESCs were divided into two groups:group A:standard medium+VEGF+SCF;group B:standard medium+VEGF+SCF+bFGF.Each group was cultured for 3 days and 6 days respectively,and the cloning number of BL-CFC was quantified,as well as Flk-1+ expression was observed by immunofluorescence staining.Positive number and average absorbance were analyzed using IMAGE-PRO PLUS imaging analysis system.RESULTS AND CONCLUSION:Adding bFGF in the course of embryoid body growth could significantly increase the number of BL-CFC(P<0.01).and the positive results of Flk-1 and the average absorbance were also increased significantly(P<0.01).bFGF effectively promoted embryoid body amplification and proliferation of BL-CFC.
6.Signal molecules related to self-renewal of hemopoietic stem cells: Effects and pathways

BACKGROUND: There are small amount of hematopoietic stem cells, which are easy to divide in vitro and show the difficulty in applying to transplantation.OBJECTIVE: This paper has focused on the role and means of the Wnt, Notch, Bmi_1, Shh, HOXB4 signaling molecule in the maintenance of hematopoietic stem cell self-renewal and regulation. METHODS: With the key words of "HSC, Wnt, Notch, Bmi_1, Shh, HOXB4" for the search, we searched PubMed database (2002-01/2008-12) in English. Literatures closely related to the hematopoietic stem cell self-renewal related signaling molecules were included. Repetitive research and Meta analysis were excluded.RESULTS AND CONCLUSION: The computer initially retrieved 216 documents, of which 30 documents for research. Hematopoietic stem cells are self-renewing, have strong differentiation and growth and regeneration capacities, can produce various types of blood cells ancestor cells, are widely used to treat blood diseases, but the hematopoietic stem cell differentiation in vitro demonstrated that the difficulties used in transplantation. How to make hematopoietic stem cells in vitro amplification and processing, while maintaining hematopoietic stem cell self-renewal characteristics is of a key issue. In recent years, different signaling pathways to enhance the ability of hematopoietic stem cell self-renewal signaling molecule have been a research hotspot. The article focused on the role and means of the Wnt, Notch, Bmi_1, Shh, HOXB4 in the maintenance of hematopoietic stem cell self-renewal and regulation and found that both the above-mentioned five signaling molecules can enhance hematopoietic stem cell self-renewal function. There are also a number of factors playing an important role in the maintenance of hematopoietic stem cell self-renewal process, such as endogenous factors, a series of transcription factors Oct-4, Ehox, Nanog, SCL, Runx1 and so on, to explore how their regulatory networks formed by the interaction control self-renewal of hematopoietic stem cells will become a key point in the research of self-renewal of hematopoietic stem cells.
7.EFFECT OF ASCORBIC ACID ON ISCHEMIA-REPERFUSIONARRHYTHMIAS IN ISOLATED GUINEA PIG RIGHT VENTRICULARWALL

Objective To investigate the protective effect of ascorbic acid on myocardium from ischemia and reperfusion injury and its electrophysiological mechanism. Methods Using modified Ferrier's arrhythmia induced by ischemia and reperfusion injury in isolated guinea pig right ventricular wall. Results Ascorbic acid at 5, 10, 50 and 100μmol/L slightly decrease incidence of ischemia arrhythmia (P > 0. 05). In contrast, ascorbic acid at 10, 50 and 100μmol/L significantly reduced the incidence of arrhythmia during early reperfusion(2~12min) (P < 0. 01 or P < 0. 001). Ascorbic acid at 5~ 100μmol/L didn't reverse the abbrevating of APD of endocardium and epicardium and didn't affect the conduction time in endocardium (P > 0. 05). However, ascorbic acid abbreviated epicardial conduc tion time significantly (P < 0. 05 or P < 0. 01 or P < 0. 001 ). Conclusion The study suggests that ascorbic acid might exert antiarrhythmic effect via alleviating conduction abnormity to avoid transmural reentries during ischemia and early reperfusion.
8.EFFECTS OF GENISTEIN ON APOPTOSIS IN HUMAN OSTEOBLAST CELLS

Objective:To investigate the effects of genistein (Gen) on apoptosis in human osteoblast. Methods:Human periosteum was maintained in DMEM medium containing 10% newborn calf serum through four passages to isolate and proliferate osteoblast. Then, cells were maintained in phenol red-free DMEM supplemented with 5% charcoal-stripped FCS for 4 d before addition of treatments. Flow cytometer and DNA fragment were used to explore apoptosis in osteoblast, and immunohistochemistry was used to study protein expression of Bax, Bcl-2 and caspase-3. Results:Estrogen depletion could induce apoptosis in human osteoblast. With the similarity of 10-8 mol/L E2, at the conentrations of 10-6 mol/L and 10-5 mol/L, Gen could inhibit apoptosis induced by estrogen depletion. Compared with vehicle control, Gen significantly decreased protein expression of Bax and caspase-3 and increased protein expression of Bcl-2. Conclusion:Genistein could inhibit apoptosis induced by estrogen depletion in human osteoblast.
