Objective To study the effects of arsenic compounds on metallothionein-3 gene expression in human normal hepatic cells. Methods cDNAs were cloned by SMART method. Bioinformatics was utilized to analyze the homologue, chromosomal localization, structure and encoding protein of the cloned gene and the trans-membrane information of the encoding protein. Metallothionein-3 gene expression level in L-02 cell line treated by arsenite was determined by cDNA microarrays. Results Metallothionein-3 cDNA gene was cloned and located in the chromosome 16q13. The results of bioinformatics analysis showed that the protein encoded by metallothionein-3 gene could cross the biologic membrane. Metallothionein-3 gene expression up-regulation in human normal hepatic L-02 cell line was found by cDNA microarrays in the early stage after the cells being exposed to arsenite. Conclusion The results of this study showed that the human metallothionein-3 gene was arsenic related gene and this gene might play a vital role in the detoxification metabolism of arsenic compounds at early stage.

Objective To explore the clinical features, characteristics of neuroimaging and prognosis of toxic encephalopathy induced by 1,2- Dichloroethane. Methods The clinical features, characteristics of neuroimaging and prognosis in eight cases with toxic encephalopathy induced by 1,2- Dichloroethane were analyzed retrospectively. Results In acute or subacute stage of toxic encephalopathy induced by 1,2- Dichloroethane, the whole brain injury symptoms and severe intracranial hypertension were the main neurological manifestations. CT and MR examination showed diffused encephaledema and brain swelling. DWI test showed the symmetric diffused low density lesions in white matter of cerebral hemisphere, and the average values of ADC decreased significantly. With the disease improving, the brain swelling was alleviated, the brain lesion foci reduced gradually, and the average values of ADC increased. The prognosis will be improved dramatically if sufficient mannitol, hormone and high pressure oxygen were used in the early stage. Conclusion The diagnosis of toxic encephalopathy induced by 1,2- Dichloroethane depends mainly on the clear toxicant contact history, clinical features and characteristics of neuroimaging. Taking effective measures in the early stage to control actively the encephaledema would be beneficial to the prognosis.

Objective To evaluate the effects of the project of ditching for drain on Oncomelania hupensis snail control and flood prevention security and explore the optimal engineering design plan in Dongting Lake region. Methods A retrospective study was performed on the previous studies about the project of ditching for drain. The reference values of project indices were determined. The outside levee of Nanhu New Distinct of Yueyang City in Dongting Lake region was selected as the study area , and the cross section of marshland perpendicular to the center line of the levee was extracted to research. According to the situa?tions of various water levels,a dynamic simulation was performed on the effect and security of the project of ditching for drain through the software FLAC3D. Results The retrospective study showed that the project would be effective when the relatively subsoil water level decreased by 0.35 m,and the soil water content decreased correspondingly. The dynamic simulation by FLAC3D showed that the minimum safe distances between transverse ditch 1,vertical ditch and levee toe should be 25 m and 13 m respectively. The digging depth of transverse ditch and vertical ditch should be 1.2 m and 1.0 m respectively. If the width of marshland in drought period was less than 500 m,one transverse ditch was efficient. Otherwise, more transverse ditches should be set with the intervals of 300 m. Conclusions The project of ditching for drain is an effective ecological snail elimination method. Optimizing the digging depth of ditches and distances between transverse ditches, vertical ditch and levee toe will en?sure the effects and security of the project.

Epidermal growth factor (EGF) is an epithelial cell growth factor that can stimulate intestinal development, repair the damage of epidermal cells as well as reduce the incidence of pathogen infection and diarrhea. In order to produce a recombinant Lactobacillus plantarum (L. plantarum) expressing porcine epidermal growth factor (pEGF), we constructed a recombinant vector stably expressing pEGF in L. plantarum strains. First, L. plantarum strain Lp-1 was isolated from intestinal contents of piglets. Then the functional domain of pEGF, M6 precursor protein signal peptide (SP) and super strong constitutive promoter (SCP) were connected with the backbone plasmid pIAβ8 to construct the recombinant vector that was transformed into Lp-1 by electroporation. Afterwards, pEGF was expressed in Lp-1 and detected by Tricine-SDS-PAGE and ELISA. After orally irrigated early-weaned BALB/c mice with the recombinant L. plantarum every morning and late afternoon for 10 consecutive days, body weight, villous height and crypt depth in the intestine were measured to examine the influence of the recombinant bacteria on the intestinal development of early-weaned mice in vivo. Finally, the results of our experiments demonstrated that pEGF was successfully expressed in Lp-1 and the molecular weight of pEGF was 6 kDa. In addition, the recombinant pEGF can enhanced the daily gain and exerted significance influence (P < 0.05) to the small intestinal morphology of early-weaned BALB/c mice. In conclusion, pEGF could be expressed in L. plantarum and the recombinant pEGF possesses good biological activity.

Objective To compare the curative effect of perindopril and metoprolol in the treatment of diabetic patients with asymptomatic cardiac insufficiency. Methods According to the method of random numbers, 152 diabetic patients with asymptomatic cardiac insufficiency were divided into the observation group and control group, with 76 cases in each group. The observation group was given with perindopril treatment, while control group was given with metoprolol treatment. The cardiac function index, glycosylated hemoglobin,BNP levels, the incidence of cardiac insufficiency and the occurrence of adverse drug reactions before and after treatment were compared. Results There were no significant differences in LVEF,Tei index and E/A between two groups before treatment (all P >0.05 ).Compared with before treatment, the cardiac function indexes of the two groups were significantly improved after treatment(t=5.96,4.83,4.91,10.22,5.68,5.18,all P<0.05),but the observation group were improved more significantly(t=5.64,4.67,5.03,all P<0.05).Before and after treatment, there was no significant difference in the levels of glycosylated hemoglobin(all P>0.05),and the difference between the two groups was not statistically significant(all P>0.05).The BNP levels of the observation group before and after the treatment were (515.43 ± 58.85)pg/mL and (214.43 ±32.42)pg/mL respectively,of which the control group were (513.94 ±59.11)pg/mL and (263.32 ±43.31)pg/mL respectively, and after treatment, the BNP levels of the two groups were significantly decreased(t=23.45,17.68,all P<0.05),of which the observation group decreased more obvious(t=6.97,P<0.05).The cardiac insufficiency incidence of the observation group was 3.95%,which was significantly lower than 14.47% of the control group(χ2 =5.03,P<0.05).The incidence of adverse drug reactions had no obvious differ-ences in both groups(χ2 =0.08,P>0.05).Conclusion Using perindopril in treatment of diabetic patients with asymptomatic heart function can obviously improve cardiac function, reduce the incidence of BNP and cardiac insuffi-ciency.It has the clinical curative effect and high safety.
8. Epidemiological distribution characteristics of 2 342 cases of hip fracture: A single center analysis
Chinese Journal of Tissue Engineering Research 2020;24(32):5085-5091
BACKGROUND: Early prevention of hip fracture can effectively reduce the risk of hip fracture in a high-risk population. Epidemiological studies can provide guidance for the clinical prevention and treatment of such patients. OBJECTIVE: To retrospectively analyze the epidemiological distribution characteristics of patients with hip fracture admitted to the Second Hospital of Shanxi Medical University from January 1, 2016 to December 31, 2018. METHODS: To collect the patient’s age, sex, fracture type, territorial, season for injury, causes, surgical procedure, intraoperative situation, blood clots, length of hospital stay, cost, and admission to operation time, body mass index, complications, SPSS 26.0 software was used for data processing to analyze the epidemiology of hip fracture distribution characteristics in 2342 patients. RESULTS AND CONCLUSION: The average age of patients with hip fracture was (73.90±13.43) years, and hip fracture mostly occurred in the 80-90 group, accounting for 29.97%. Hip fractures were common in winter, and there was a significant difference in the seasonal distribution of femoral neck and intertrochanteric fractures. Falls resulted in the most hip fractures, accounting for 88.13%. Among the 2 108 patients with hip fractures who underwent surgical treatment, hip replacement was the major choice for femoral neck fractures, accounting for 60.03%; intramedullary fixation was the main choice for intertrochanteric fractures, accounting for 92.04%. Among the 2097 patients with hip fracture who underwent color Doppler ultrasonography, 32.38% (679/2 097) of the patients had arteriovenous thrombosis of the lower extremities, and 28.71% (195/679) of the patients had inferior vena cava filter implantation. The average hospitalization time of patients with hip fracture was (10.49±7.06) days, and the average hospitalization cost was (4.63±3.14) thousand yuan. The average hospitalization cost, average blood loss and average operation time of femoral neck fractures were significantly less than those of intertrochanteric fractures. Patients with intertrochanteric fractures were generally thinner than those with femoral neck fractures. The 44.41% of patients with hip fracture had cardiovascular and cerebrovascular diseases. These results indicate that hip fractures are mainly developed in the older adults, especially in elderly women. With the increase of age, the proportion of intertrochanteric fractures increases gradually. Falls are the main cause of hip fractures in the older adults. Surgical treatment is the main choice for hip fracture. Preoperative blood preparation and perioperative thrombosis prevention can improve the prognosis of patients.

Objective To observe osteocyte survival and osteogenesis after nonvascularized free bone graft transplantation for canine mandible mass defect restoration. Methods An experimental canine premolar was extracted. 1 month later, the length of 3.0 cm edentulous mandible was cut off, as the same length as that nonvascularized and soft-free tissue complete ilium block was cut off and fixed into mandible defect in 40, 120 min. The survival osteocyte and osteogenesis of the nonvascularized graft were detected 4 and 8 weeks after transplantation. Results The continuity of canine mandible segmental defect of 3.0 cm length was repaired by free nonvascularized free bone graft, live osteocytes and osteogenesis were observed in non-vascularized free bone graft at off-body 40 min group in 4-8 weeks. But the nonvascularized free bone graft off-body 120 min was a dead bone, no survival osteocytes were observed, some osteoclasts were seen in the bone graft. Conclusion The length of 3.0 cm canine mandible defect can be restored by non-vascularized free bone graft; the osteocyte survival and osteogenesis are related to their off-body time before they are fixed into recipient.
