Objective To investigate the correlation of the S100B level with cardiac function and chronic heart failure. Methods A total of 80 elderly chronic heart failure (CHF) patients who accepted treatments in our hospital from March 2012 to December 2013 were recruited, and 80 subjects undergoing health examinations served as the control group. The New York Heart Association (NYHA) functional classification in the experimental group was determined.Serum levels of S100B and NT-proBNP,left ventricular end diastolic diameter (LVEDD) and left ventricular ejection fraction (LVEF) were compared between the groups, and the correlation of the S100B level with NT proBNP,LVEF and the NYHA functional classification were analyzed. Results The serum S100B level was increased in the experimental group as compared with the control group [(91.4 ±31.9) pg/L vs.(74.9±26.0) pg/L,t=1.980,P=0.000].The serum NT-proBNP level was higher,LVEDD was longer, and LVEF was lower in the experimental group than in the control group (all P<0.01).The serum S100B level was positively correlated with NT-proBNP (r=0.309,P=0.003) and the NYHA functional classification (r=0.508,P=0.001),and negatively correlated with the LVEF (r=-0.192,P=0.004).Conclusions The serum S100B level is increased in patients with CHF and negatively correlated with cardiac function, suggesting that the S100B protein may serve as a valuable tool for the diagnosis and assessment of CHF.However, more studies are needed to provide more evidence.

Objective To detect mutations in the filaggrin (FLG) gene and expressions of FLG and loricrin in patients with ichthyosis vulgaris (Ⅳ),and to investigate their clinical significance. Methods Tissue specimens were resected from the skin lesions of 10 patients with Ⅳ and normal skin of 14 healthy human controls, and immunohistochemical SP method was used to detect the expressions of filaggrin and loricrin. The expression intensity was determined by the Image-Pro Plus (IPP) software, and expressed as positive units (PU).Blood samples were collected from 10 patients of Han nationality with Ⅳ and 100 healthy human controls followed by DNA extraction.PCR and DNA sequencing were performed to detect the presence of 13 mutations (3321delA,441delA,1249insG,E1795X,S3296X,R501X,2282de14,R2447X,S2889X,7945delA,3702delG,Q2417X,R4307X) in the FLG gene. Results FLG was mainly expressed in the cytoplasm of keratinocytes in the stratum corneum, granular layer, prickle layer and basal layer, and loricrin was observed in the cytoplasm and nuclei of keratinocytes in the granular layer, prickle layer and basal layer,in both the lesional and normal skin.Compared with the normal skin, the lesional skin showed significantly weaker expressions of FLG (0.208 2 ± 0.008 0 vs.0.230 0 ± 0.0228,t =3.30,P < 0.01) and loricrin (0.137 0 ± 0.011 2 vs.0.149 3 ± 0.007 3,t =3.07,P < 0.01).Sequencing analysis identified two mutations, including 3321delA in 7 patients and 441delA in 2 patients. No mutations were detected in the healthy controls. Conclusions The 3321delA and 441delA mutations in the FLG gene may represent the most frequent genetic cause of Ⅳ in patients of Han nationality. The low expressions of FLG and loricrin may be associated with the impairment of skin barrier function in patients with Ⅳ.

Virologica Sinica 2008;23(3):173-182
To prepare HIV-1 Vif and hAPOBEC3G and to produce their antibodies, the full length gene fragment of HIV-1 Vif was amplified by PCR from a plasmid of HIV-1 NL4.3 cDNA, and the APOBEC3G gene was obtained by RT-PCR from the total RNA of H9 cells. The resulting DNA construct was cloned into a prokaryotic expression vector (pET-32a). Recombinant pET-vif and pET-APOBEC3G were expressed respectively in Eserichia coli BL21 (DE3) as an insoluble protein. The vector also contained a six-histidine tag at the C-terminus for convenient purification and detection. To express and purify the HIV-1 Vif and hAPOBEC3G in E. coli cells, the accuracy of inserted gene and specificity of proteins were detected by the two enzyme digestion method, SDS-PAGE, and Western blotting. Rabbits were then immunized by Vif or APOBEC3G protein and serum samples were tested by indirect ELISA to determine the level of antibodies. Immunoenzyme and immunofluorescence assays were performed to identify the specificity of polyclonal antibodies. The titer of the anti-Vif antibodies was 1:204800, and that of the anti-APOBEC3G antibodies was 1:102400. Thus the antibodies could detect the antigen expression in the cells, demonstrating that fusion proteins with high purity and their corresponding polyclonal antibodies with high titer and specificity were achieved.

Objective To study the clinical features, diagnosis and treatment of Dieulafoy′s disease. Methods The clinical characteristics, diagnosis, methods of treatment and efficacy of treatment of 19 cases of (Dieulafoy′s) disease were analyzed retrospectively. Results All cases presented as abrupt upper gastrointestinal bleeding.12 cases(63.2%) were diagnosed at the first, and 4 cases (21.1%) at the second ecdoscopic examination, and 3 cases(15.8%) were diagnosed at exploratory operation.Hemorrhage stopped successfully after one endoscopic treatment in 14 cases and after 2 endoscopic treatments in 1 case.4 cases were cured by operation(including 1 case with re-bleeding 72 hours after endoscopic treatment). Conclusions Emergency endosopic examination is the best way to diagnose Dieulafoy′s disease and endoscopic treatment is the method of choice to treat it.If hemorrhage fails to stop after endoscopic treatments, surgical operation should be (performed) immediately.

Virologica Sinica 2011;26(3):206-213
An optimized recombinant HPV16 E6E7 fusion gene(HPV16 ofE6E7)was constructed according to codon usage for mammalian cell expression, and a mutant of HPV16 ofE6E7 fusion gene(HPV16 omfE6E7)was generated by site-directed mutagenesis at L57G,C113R for the E6 protein and C24G,E26G for the E7 protein for HPV16 ofE6E7 [patent pending(CN 101100672)].The HPV16 omfE6E7 gene constructed in this work not only lost the transformation capability to NIH 3T3 cells and tumorigenicity in SCID mice, but also maintained very good stability and antigenicity. These results suggests that the HPV16 omfE6E7 gene should undergo further study for application as a safe antigen-specific therapeutic vaccine for HPV16-associated tumors.

Objective To observe expression of membrane-type 1 matrix metalloproteinase-1 (MT1-MMP) and matrix metalloproteinase-2 (MMP-2) and its relation to clinicopathological characteristics in cutaneous squamous cell carcinoma (SCC) and basal cell epithelioma (BCE). Methods The expression of MT1-MMP and MMP-2 was determined in situ hybridization and ABC immunohistochemical technique respectively in 48 cutaneous SCCs and 41 BCEs. Results The expression of MT1-MMP and MMP-2 in SCC was significantly higher than that in BCE (P 0.05). Conclusions Overexpression of MT1-MMP and MMP-2 might be related to lymphatic metastasis in SCC. Expression levels of MT1-MMP and MMP-2 may not be responsible for the differentiation in aggressiveness of BCE.
