2.Demethylation of progesterone receptor in leukemia cell line induced by DNMT1 antisense oligonucleotide wrapped in liposome

Objective To investigate the effects of the transfection of DNMT1 antisense oligonucleotide wrapped in liposome on the promotor methylation and expression of progesterone receptor in leukemia cell lines so as to find a new pathway of methylation for gene therapy. Methods Artificially synthesized DNMT1 antisense oligonucleotide MG88 wrapped in liposome was transfected into the leukemia cell line. Changes of DNMT1 & PRB mRNA were detected by RT PCR, and the changes of the methylation status of PRA & PRB were analyzed by MSP. Results After transfection of leukemia cell line with MG88, DNMT1 mRNA decreased significantly. PRA & PRB promotors were demethylated, and the expressions of PRB mRNA increased significantly. Conclusion MG88 can induce the demethylation of PR promoters in leukemia cell line, resulting in the increase of PRB mRNA expression. So MG88 can be used in the experimental study of DNA methylation for gene therapy.
3.Changes of Selenium Metabolism,Glutathione Peroxidase Activity and Lipid Peroxides Content in Severely Scalding- injured Rats

The changes of selenium metabolsim, glutathione peroxidase acivity and lipid peroxides content in the tissues of rats suffering from 30% TBSA full thickness scalding were observed in the first 7 days after injury. It was found that selenium content in the rat tissues decreased remarkably after injury,which in turn resulted in serious reduction of glutathione peroxidase activity and significant increase of lipid peroxides in the serum, erythrocytes and liver. However the muscular tissue showed no significant changes.These facts imply that after burn injury, the body is in a state of selenium deficiency, the loss of selenium might be responsible for the reduction of anti-peroxidation ability of glutathione peroxidase, and consequently there is an increase of lipid peroxides in the tissues. Only the muscular tissue is insensitive to lipid peroxiation. It is believed that the reduction of anti-peroxidation ability of glutathione peroxidase after burn injury might be one of the main causes to intensify the injury resulting from free radicals.

5.Comparision of two anesthesia combination regimens for renal transplantation

Objective This research aimed to compare two regimens of anesthesia(vecuronium-fentanyl and cisatracurium-remifentanil)for kidney transplantation patients.Methods 108 patients were studied,45 patients(Group Ⅰ)received balanced anesthesia with vecuronium and fentanyl,to the others 63(Group Ⅱ),cisatracurium and remifentanyl was given.Comparison of the difference between the two groups was made on the extubation time,intraoperative hemodynamics,postoperative urine output,serum creatinine and anesthesia complications.Results Extubation time of the Group Ⅰ was significantly longer than that of Group Ⅱ(33.5min ±17.5min,P<0.05),but there was no difference between two groups on intraoperative hemodynamics,postoperative urine output,serum creatinine and anesthesia complications.Conclusions Both of the two regimens can provide safe anesthesia for kidney transplantation.Patients received regimen of cisatracurium and remifentanil experienced shorter extubation time.This regimen may be a better anaesthesic regimen for renal transplantation.

Treating from liver is an important thought to treat esophageal cancer.Regulating the liver has very important significance in the process of treating esophageal carcinoma,which embodies at regulating liver is the key of soothing emotional injuries of esophageal carcinoma,removing esophageal pathology product and coordinating rising-descending function of spleen and stomach,and improving accompainedg symptoms of esophageal cancer.The thought of treating from liver should be used throughout the whole process of the treatment.So we must pay morn attention to regulate the liver and prevent liver injury during the differentiation and treatment of esophageal carcinoma.
7.Construction and effect identification of RNAi eukaryotic expression vectors of iASPP

Objective To construct the RNAi eukaryotic vector of inhibitory member of the ASPP family(iASPP) gene and observe the interfering effect in Jurkat cell line after the vector transfection.Methods The specific siRNA sequence was designed according to the iASPP sequence in GenBank.The sequence was cloned into PGCsilencer~(TM) H1/Neo/GFP and then sequence analysis was performed.The recombinant plasmid was transfected into Jurkat cell by liposome.The iASPP expression was analyzed by RT-PCR and cell apoptosis was detected by FCM.Results The sequence of templet and specific siRNA was correct by sequence analysis.The iASPP expression in Jurkat cells decreased and the apoptosis rate increased from 11.81% to 33.15% after RNAi transfection.Conclusion The RNAi eukaryotic vector PGCsilencer~(TM)H1/Neo/GFP/RNAi was constructed successfully.The RNAi inhibitory effect on the Jurkat cells is confirmed.The successful construction of iASPP RNAi makes it possible to further study the interaction between iASPP and p53.

Objective To assess the reliability and validity of the Chinese version of Multidimensional fatigue symptom inventory-short form(MFSI-SF).Methods The reliability and validity of the Chinese version MFSI-SF were assessed in a sample of 203 cancer patients.Statistical software was used to perform the analysis.Results The results showed moderate correlation between items and the total scale,the content validity index was 0.82,and exploratory factor analysis indicated five dimensions of the scale,the cumulative variance contribution was 56.65%.Confirmatory factor analysis showed moderate model fitting:x2/df=l.73,GFI=0.83,AGFI=0.79,NNFI=0.94,RMSEA=0.06,criterion validity was 0.585,and the Cronbach α of the total scale was 0.896.Conclusions The results demonstrated good convergent validity,it is suitable to evaluate fatigue status in Chinese cancer patients.
10.Effectiveness of clinical rotation training in pain department for general practitioner and its reflections

Objective To evaluate the effectiveness of clinical rotation training in pain department for general practitioner.Methods Totally 31 general practitioners of 3 years program in the First Affiliated Hospital of Chongqing Medical University were involved in clinical rotation training in pain department form August 2008 to December 2011.Training lasted for one month.Clinical rotation training in pain department was conducted for general practitioners by studying rules and regulations,treatment methods,imaging data and clinical practice.Evaluation was made according to the treatment,theoretical knowledge,interpersonal skill and professional competency of general practitioners.Measurement data were expressed as mean ± standard deviation ((-x) ± s) while enumeration data were expressed as rate.Descriptive analysis of the data was used.Results Average total number of patients managed by 31 general practitioners was (15 ± 2),average total number of disease species was (5 ±2),average number of case discussion and small lecture participated was 1 ~2,average number of nerve block participated as assistant was (40 ± 5) and average number of minimally invasive surgery visited and studied was (2 ± 2).Residency duties were skillfully completed and assessment score was (85 ± 4.50).Conclusions Through clinical rotation training,general practitioners can understand common disease diagnosis and treatment category; familiarize clinical evaluation methods and master standards of diagnosis and treatment in pain department.Meanwhile,they would learn doctor-patient communication to establish a good therapeutic relationship with patients.
