1.Decline of learning and memory in senility and expression of cyclin-dependent kinase-5 in brain development of rats

BACKGROUND: Cyclin-dependent kinase-5 (CDK-5) is one of the members in cyclin-dependent protein kinase family. The attention has being drawn by researchers on the relationship between the expression and distribution of CDK-5 mRNA and its protein in the brain during brain development and neural degeneration in thought-cognition.OBJECTIVE: To probe into the influence of CDK-5 on neurogeny and neural degeneration during cerebral development.DESIGN: Single factor analysis of variance.SETTING: Histological and Embryological Department and Neurobiological Department in Third Military Medical University of Chinese PLA.MATERIALS: The experiment was performed in Histological and Embryological Department and Neurobiological Department in Third Military Medical University of Chinese PLA. Twenty-five Wistar rats of 5 phases were employed, named embryonicphase (E8-E21), neonatal phase (P0-P15),childhood (P16-2 months), grown-up phase (> 2 months) and senile phase (> 8 months), 5 rats in each group.METHODS: In situ hybridization histochemistry (ISH) and immunohistochemistry (IHC) staining was adopted in brain sections from embryonic phase to senile phase.MAIN OUTCOME MEASURFS: Distribution and expression of positive cells of CDK-5 mRNA and protein in various brain areas.RESULTS: Twenty-five rats entered result analysis for all. ① The expression of CDK-5 mRNA presented in entire development from E14 to P350and was in tendency of stability after growth-up. CDK-5 mRNA localized mainly in neurons and positive regions distributed mainly in cerebral cortex, hippocampus, thalamus, hypothalamus, cerebellum and a part of nerve nuclei. ② The expression of CDK-5 was strong after birth and it was weaker in embryonic and senile rats. Positive regions concentrated mainly in peripheral ventricle, hippocampus, cerebellum and a part of nerve nuclei.The expression only presented in hippocampus and Purkinje cellular layer of cerebellum in senile rats.CONCLUSION: CDK-5 in brain runs through entire phases of neural development, it expresses more significantly in neonatal phase and childhood and declines after growth-up, especially in senile phase. The declined expression of CDK-5 in hippocampus of senile rats is closely associated with decline of learning and memory in senility probably.
2.Expression of signal transducer and activator of transcription 3 protein in development of embryo liver in mice

BACKGROUND: Signal transducer and activator of transcription 3 (STAT3) protein is widely expressed in cells and tissues of different type and. It is the important component of signal transduction and transcription chain that participates in the regulation of various physiological functions of cells like proliferation and differentiation. However, there is no literature reported regarding if there is STAT3 expression and whether it participates in embryo liver development both at home and abroad.OBJECTIVE: To explore the effects and mechanism of STAT3 signal transduction route on development of embryo liver by. Studying the expression of STAT3 protein during embryo liver development in mice.DESIGN: Randomized controlled study based on STAT3 protein.SETTING: Department of histology and embryology in a military medical university of Chinese PLA.MATERIALS: The study was completed in the Department of Histology and Embryology, Third Military Medical University of Chinese PLA during January to June 2004. Kunming mice were provided by Animal Centre of Third Military Medical University of Chinese PLA. Ten male mice and 20 female mice with body mass during 25 to 30 grams were randomly chosen.METHODS: Serial frozen sections of mouse embryo was performed in the 13.5 days ( n = 8), 14.5 days ( n = 9) and 15.5 days ( n = 7) after mating respectively. One of the every 2 slices was used as negative control by replacing first antibody of STAT3 with 0. 01 mol/L PBS. Immunochemical technique and immunofluorescence method were used to display the expression of STAT3 protein in embryo liver development in mice.MAIN OUTCOME MEASURES: Expression of STAT3 protein in serial frozen sections of mouse embryo.RESULTS: There was strong expression of STAT3 protein in liver cells in E13.5 days mouse embryo while the expression was decreased in E14.5 days and E15.5 days mouse embryos.CONCLUSION: STAT3 protein has participated in the development of embryo liver. The expression of STAT3 in liver development can be used to explain the mechanism of liver regeneration and provide experimental data for organ repair.
3.xperimental study on the effect of Naogongfude on the aging of the rat brain

Objective To evaluate the effect of Naogongfude (NGFD, a traditional Chinese medicine orally administrated) on learning and memory and the expressions of synaptophysin (SY) and Tau-protein in cerebral cortex and hippocampus in aged rats and to explore the possible mechanism. Methods Rats were divided into normal and neural disturbance groups according to the outcomes of active avoidance reaction (AAR) test, and then each group was randomly divided into control and NGFD-treated experimental groups. Animals were orally fed with NGFD for 2 months (5 ml/d) in experimental group or routinely fed in control and taken AAR and passive avoidance reaction (PAR) tests. After the rats were sacrificed, the synaptosome count, the expression of SY and Tau-protein, and the neuron apoptosis in cerebrum were examined. Results The rats after 2-month NGFD administration had an increased AAR acquisition, obviously delayed AAR extinction and prolonged step through latency (STL) of PAR. The number of synaptosomes was raised and the immunoreactive intensity of synaptophysin was increased remarkedly, while Tau-protein immunoreactivity and apoptotic cells were decreased in cerebrum. Conclusion NGFD does have the effect of improving brain function and putting off the aging of rat brain according to the results of behavior study and morphological observation.
4.Clinical Observation of Vitamine C Tablets for Local Use in the Treatment of Oral Ulcer after Grinding

OBJECTIVE:To observe the clinical efficacy and safety of Vitamin C tablets after grinding for local use in the treatment of oral ulcer.METHODS:Two hundved and eighty-six cases of oral ulcer in our hospital during Feb.2014-Fed.2016 were selected and divided into treatment group and control group according to random number tabel,with 143 cases in each group.Control group was given Guilin watermellon frost,2-3 times a day;treatment group was given Vitamin C tablets powder for local use,2-3 times a day.Clinical efficacy,the time of ulcer disappearance,the incidence of ADR,recurrence rate of ulcer,treatment satisfactory degree were observed in 2 groups.RESULTS:Total response rate of treatment group was 96.5 %,which was significantly higher than 80.4% of control group,with statistical significance (P<0.05).The time of ulcer disappearance was (2.37 ± 0.89) d in treatment group,which was significantly shorter than (4.12 ± 1.33) d of control group;recurrence rate of ulcer was 3.5%,which was significantly lower than 8.4% of control group;treatment satisfactory degree was 90.9%,which was significantly higher than 79.7% of control group,with statistical significance (P<0.05).There was no statistical significance in the incidence of ADR between 2 groups (P>0.05).CONCLUSIONS:Vitamin C tablets after grinding for local use has obvious effect on oral ulcer,significantly shortens the time of ulcer disappearance,reduces recurrence rate,and improves treatment satisfactory degree with good safety.
5.Benzyl isothiocyanate induces apoptosis of human leukemia cells:molecular mechanism

Objective To explore the apoptotic effect of benzyl isothiocyanate(BITC) on human leukemia cells and investigate its related molecular mechanism.Methods Cells(U937,Jurkat and HL60) were exposed to BITC at various concentrations(0,2,4,6 or 8 ?mol/L) for 6 h or 12 h,or at 8 ?mol/L for different time intervals.The apoptosis was measured using flow cytometry.The apoptotic-related proteins,such as Caspase-3,poly ADP-ribose polymerase(PARP),myeloid cell leukemia-1(Mcl-1) and so on,were determined using Western blot assay.Results BITC induced apoptosis in human leukemia cells in dose-and time-dependent manners.BITC at a dose over 4 ?mol/L began to induce an increased expression of Caspase-3 protein and decreased expression of PARP in U937 cells,and when the dose was 8 ?mol/L,the changes reached their summits respectively.The expression of anti-apoptotic protein Mcl-1 was decreased in U937 cell after exposure of BITC.Conclusion BITC induces apoptosis in human leukemia cells including U937,Jurkat and HL60,and downregulation of Mcl-1 may play an important role in BITC-induced apoptosis.
6.Methods and practice of systematic clinical first-aid skills training for community doctor

Objective To observe the effect of clinical first-aid skills training for community doctor. Methods Effect of systematic clinical first-aid skills training and traditional teaching methods were scored by the questionnaire survey in order to compare their difference. Results Through systematic clinical first-aid skills training, the score on contacting the actual work, improving the diagnosis and treatment, analyzing and problem-solving skills, enhancing learning initiative, participation, scientific learning method and self-learning ability, impro-ving clinical thinking, strengthening team spirit and humane education were significantly improved compared to that of the traditional teaching methods (P<0. 01). Conclusion The systematic clinical first-aid skills training is an effective way for community physicians.
7.Effect evaluation and countermeasures of health education for patients discharged from hospital

Objective To investigate the effect of health education for discharged patients, and to explore the reasons and effective countermeasures on the health education. Methods Through the question-naire survey, convenient sampling method was used to evaluate the effect of the hospital health education. The existing problems and corresponding countermeasures taken for continuous improvement were analyzed. SPSS 17.0 statistical software was applied for data processing, the count data was expressed by rate and Statistical methods using chi-square test. Results The hospital health education satisfaction rate rose to 95.21%from 54.17%after intervention, and differences between the two groups was statistically significant (P<0.05). The satisfaction rate of the department of internal medicine, surgery department, and the department of gynaecology and pediatrics was significantly improved than before. Conclusion By improving health education importance, strengthening learning and training of specialized subject knowledge, rationally allo-cating human resources and establishing health education quality control system, the health education for discharged patients was effectively improved with satisfaction.
8.The application of cardiopulmonary resuscitation skill training in pre-job training for clinical interns and its value of quality control

Objective To explore the application of cardiopulmonary resuscitation skill training in pre-job training for clinical interns and its value of quality control. Methods A total of 136 clinical interns in Chongqing Qianjiang Central Hospital who came from Chongqing Medical University, Jishou University, and Hubei University for nationalities were included in this study from June 2015 to June 2017. The test score, pass rates and make-up pass rates after 3 months of pre-job training of cardiopul-monary resuscitation were compared between training and evaluation group (2016 session) and control group (2015 session). We analyzed the correlation of retraining and evaluation of cardiopulmonary resuscitation and test score of pre-job training and its value of predicting the effectiveness of pre-job training. Results There were 65 clinical interns in 2016 session and 71 clinical interns in 2015 session. After 3 months training, the test score, pass rates and make-up pass rates in retraining and evaluation group were significantly higher than that in the control group, respectively (t=15.594,P=0.000; χ2=22.859, P=0.000; χ2=6.179, P=0.018). Conclusions Car-diopulmonary resuscitation skill training could enhance the effectiveness of pre-job training for clini-cal interns in non-affiliated teaching hospital and increase interns' ability to carry out cardiopulmonary resusci-tation and improve the quality of pre-job training for clinical interns. It could be used as an effective quality control strategy of clinical teaching
