1.Visualization of fetal cranial sutures and anterior fontanelle: Comparison between two- and three-dimensional ultrasound

Objective To evaluate the feasibility of obtaining good quality ultrasound pictures of sutures and anterior fontanelle with three-dimensional and two-dimensional ultrasound. Methods Eighty fetuses at 16-35 weeks of gestation were evaluated with two-dimensional and three-dimensional ultrasound. The sagittal, coronal, lambdoidal and metopic sutures, as well as anterior fontanelle, were inspected. Results The visualization of the sagittal suture of three-dimensional ultrasound was significantly superior to that of two-dimensional ultrasound, while no significant difference in visualization of the metopic, lambdoidal, coronal sutures and anterior fontanelle was found between the two modalities. The visualization of all sutures and anterior fontanelle was significantly superior with three-dimensional ultrasonography to that of two-dimensional ultrasonography before 30 weeks of gestation, whereas no significant difference was found between the two modalities after 30 weeks of gestation. Conclusion Three-dimensional ultrasound can provide more comprehensive information of fetal cranial sutures and anterior fontanelle than two-dimensional ultrasound does. Three-dimensional ultrasound can also supply high visualization of all sutures and anterior fontanelle before 30 weeks of gestation.
2.Observing fetal cranial sutures and fontanels with three-dimensional ultrasonography

Objective To assess the application value of three-dimensional (3D) ultrasound in visualizing fetal cranial sutures and fontanels. Methods One hundred women at different pregnant ages (16-19~+ weeks, 20-24~+ weeks, 25-29~+ weeks, 30-34~+ weeks, respectively) underwent 3D ultrasound, in order to observe the cranial sutures and fontanels of fetuses. Results ①Most cranial sutures and fontanels (70.00%-98.00%) could be visualized with 3D ultrasound. However, the sagittal suture revealed a lower displaying rate (53.00%) than others. ②At different pregnant ages, there was no significant difference of displaying rate of the metopic, coronal, squamoal sutures and the sphenoid, mastoid fontanels between fetuses and there was significant difference of displaying rate of the anterior and post fontanels, sagittal and lambdoid sutures. Conclusion 3D ultrasound can be used as an effective method for visualizing fetal cranial sutures and fontanels.
5.Detection of mitochondrial membrane potential changes in Myelodysplastic syndrome by fluorescent probe JC-1

Objective To explore the function of fluorescent probe JC-1 in detecting the changes of mitochondrial membrane potential(△?m)in early apoptotic cells.Methods After 2-ME was used to induce MUTZ-1 cell apoptosis,cells were dyed with fluorescent probe JC-1,and then the changes of △?m in the early stage of apoptotic cells were analyzed by flow cytometry or detected under fluorescent microscope. Results The control cells with high △?m are those forming JC-1 aggregates in the inner membrane of mitochondria,thus showing orange-red fluorescence.2-ME caused decrease of △?m in MUTZ-1 cells,in which JC-1 maintains monomeric form,thus showing only green fluorescence.The decreases of △?m were in a time-dependent manner,which were significantly higher than those in control group(P
6.Analysis on genetic diversity and genetic relationship of medicinal species in Dipsacus from China by SRAP.

The author detected the genetic diversity and genetic relationship within and among eight medicinal species of Dipsacus by the approach of sequence-related amplified polymorphism (SRAP). The associated genetic parameters were calculated by POPGENE 1.31. The Genetic distance was calculated by TREECONW and the systematic diagrams of genetic relationship were clustered by UPG-MA. The results showed that, using 26 primers, a total of 558 bands were produced, of which 539 were polymorphic loci. There was a high level of genetic diversity among species (PPB = 96.59%, Na = 1.9659, Na = 1.3375, H = 0.2143, I = 0.3423). However, genetic diversity was lower within species, the average of genetic parameters was PPB = 6.97%, Na = 1.0697, Na = 1.0311, H = 0.0187, I = 0.0291. The Nei's genetic differentiation coefficient was 0.9126, indicated that most of the genetic variation existed among species. By clustering analysis, different individuals gathered in the same group and the classified result of SRAP marker between traditional modal characters was almost same. The results confirmed that SRAP marker can be used as one of the effective methods to reveal the genetic diversity and relationship among medicinal species of Dipsacus.

7.Analysis on genetic diversity among wild Dipsacus asperoides by SCoT.

To investigate the genetic diversity among wild Dipsacus asperoides in China, 66 germplasmic resources of D. asperoides were analyzed by Start Codon Targeted Polymorphism (SCoT) molecular markers. Genetic distance was calculated by TREECONW software and the systematic diagram of genetic relationship was clustered by UPGMA method. The results showed that the totals of 181 bands were detected using 20 primers , among which 109 were polymorphic bands. The average percentage of polymorphic bands was 60.13%. Genetic distance changed from 0.030 6 to 0.181 4. The clustering results showed that there was no significant correlation between the classification of the wild D. asperoides and their geographical origin. The relatively high genetic diversity of D. asperoides provides the basis for breeding new varieties.

10.Expression of Matrix Metalloproteinase-9 and Its Tissue Inhibitor in Cutaneous Squamous Cell Carcinoma

Objectives To investigate the expression of matrix metalloproteinase-9 (MMP-9) and its tissue inhibitor (TIMP-1) in cutaneous squamous cell carcinomas (SCC) and the relationship between the expression and invasive growth and metastasis of SCC. Methods Immunohistochemical method of streptavidin-peroxidase (SP) was used to detect the expression of MMP-9 and TIMP-1 proteins on the paraffin embedded sections of 65 patients with cutaneous SCC and 5 cases of normal epidermis. The immunohistochemical results were analyzed quantitatively using an image analysis system. Results MMP-9 and TIMP-1 proteins were diffusely expressed on the tumor nests and the mesenchymal cells around the nests, while in normal epidermis MMP-9 and TIMP-1 were weakly positive.
