1.Preventive and Curative effects of Bee Pollen of Codonopsis Pilosula Nannf on Chronis hepatic injury by CCl_4

During the period of the subcutaneous injection of CCl_4 0.5ml/kg, four times at intervals of 5 days, the C57 mice were fed with 6.8% bee pollen of Codonopsis Pilosula Nannf (BPCPN). The results showed that BPCPN coule alleviate effctively the hepatic injury induced by CCl_4, and prevent the decrease of serum albumin and liver protein and glycogen after the liver was injured. The electron microscopis and pathologcal examination also showed that the mice had only slight hepatic necrosis, fat degeneration and fibrosis, but had a com plete repair as compared with the control groups. The above results indicated that BPCPN possessed preventive and curative on the impairment of CCl_4and could protect the hepatic metabolic function from the injury of CCl_4.
2.Studies of Hypolipidemic Effect of Amorphophallue Konjac Powder on Hyperlipidemic Rats

Having fed the rats with high-lipid food added with amorphophallue konjac powder 3.3-6.2g/Kg/d for thirty days,we observed an effective prevention of the significant incerese in serum and liver cholesterol and triglyceride,and a boost of serum HDL-C,with effects more obvious than that of clofibrate.A significant decreas in serum and liver cholesterol and triglyceride could also occur after feeding amorphophallue konjac powder 2.6-5.og/Kg/d to the rats with hyperlipidemia for ten days,the effect being more obvious than that of clofibrate.The result of the pathological examination of the liver also showed that amorphophallue konjac powder had an obvious hypolipidemic effect.

5.Effects of Early Activities on Duration and Expenditure of Hospitalization and Activities of Daily Living of Patients with Myocardial Infarction

ObjectiveTo investigate the effects of early activities after acute myocardial infarction (AMI) on duration and expenditure of hospitalization and activities of daily living (ADL) of patients.Methods68 AMI inpatients without complications were divided into the early activity group (33 cases) and control group (35 cases). Patients of the early activity group started early activity training by the bed from the 2nd day after admission, and those of the control group from the 10th day. One week before discharge, ADL of all patients were assessed with Barthel index (BI), and duration and expenditure of hospitalization of two groups were compared.ResultsThe average duration and expenditure of hospitalization in the early activity group were obviously lower than those in the control group (P<0.05~0.01), but the total scores of ADL and the scores of most items at discharge were markedly higher than those in the control group (P<0.05~0.01).ConclusionEarly activity training by the bed can obviously shorten the duration of hospitalization, reduce the medical cost and improve the ADL of AMI patients.
6. Studies on transdermal delivery of sinomenine hydrochloride through mouse skin treated by microneedle arrays

Objective: To study the characteristics and mechanism of transdermal delivery of sinomenine hydrochloride (SH) through mouse skin treated by solid silicon microneedle arrays. Methods: The amount of SH was determined by HPLC system. Hairless rat skin was pretreated with microneedle arrays. The side-by-side diffusion cell method was used to investigate the effects of needlepoint shape, different insertion forces, retention time, and number of microneedles on transdermal SH delivery. Skin samples treated by microneedles were made into paraffin sections for histological examination and were viewed by brightfield microscopy. Results: The skin pretreated with microneedle arrays had a remarkable enhancement of SH transport compared with passive diffusion group (P<0.01); the flat tipped microneedles were more effective than the sharp tipped microneedles in enhancing the skin permeability. The accumulation of SH increased with the enhancement of insertion force; however, when the insertion force exceeded 5.0 N, the accumulation of SH no longer increased. The skin permeability was enhanced with the increase of retention time; when the retention time exceeded 1.0 min, it no longer increased SH accumulation. Although skin permeability increased with the microneedle number, there was no linear correlation was found. Histological examination showed that microneedle piercing created micro-conduits in skin. Conclusion: Microneedles can create conduits in rat skin and greatly increase the skin permeability of SH; microneedle arrays provide an efficient and promising technology for transdermal drug delivery of SH.
7.Effect of Antisense Oligodeoxynudeotides Targeting Nuclear Factor ?B on Expression of Caspase-3 in Glomerulosclerosis

Objective To investigate the effect of antisense oligodeoxynudeotides(ODN)targeting nuclear factor ?B(NF-?B)on the expressions of Caspase-3 and NF-?B in glomerulosclerosis.Methods Male SD rats were divided into 3 groups:sham operation(group A,n=6),glomerulosclerosis(group B,n=6),intervention(group C,n=9).Group C was divided into 3 groups:sense ODN(group C1,n=3),non-sense ODN(group C2,n=3),antisense ODN(group C3,n=3).Glomerulosclerosis models were made for SD rats by unilateral nephrectomy and being injected with adriamycin into caudal vein.After 8 weeks,various ODN applied to corresponding group for intervention.At the end of the 9th week,kidneys were taken out from all rats for the measurement of expressions of NF-?B p65 and Caspase-3 by immunohistochemistry staining.Results After intervention,on the d7,the rats urine protein,glomerular sclerosis index(GI)and expressions of Caspase-3,NF-?B p65 in group C3 were significantly lower than those in group B,C1,C2(Pa0.05).Conclusion Antisense ODN targeting NF-?B can inhibit the activities of NF-?B,Caspase-3 in rats kidneys and retard glomerulosclerosis and glomerular intrinsic cell apoptosis.
8.Protective effects of ethanol extract of Forsythia suspensa flower on mitochondria and antioxidant activity

AIM:To investigate protective effect of ethanol extract of Forsythia suspensa flower(FFE)on mitochondrial injury induced by free radicals,antioxidation and superoxide anion(O ?-__2)scavenging activities and anti-aging mechanism.METHODS:Thiobarbituric acid(TBA)colorimetric method was used to determine malondialdehyde(MDA).Spectrometric method was used to measure the swelling of mitochondria and activities of scavenging O ?-__2 which was produced by reduced nicotinamide adenine dinucleotide(NADH)-4-Nitrobluetetrazolium chloride(NBT)-N-Methylphenazonium methyl sulfate(PMS)system.The neck back of mice were injected subcutaneously with D-galactose to induce aging model at a dose of 100 mg/(kg?d)for 7 weeks.The effects of FFE on the activities of catlase(CAT),surperoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)and anti-hydroxyl radical were assayed by ammonium molybdate colorimetry,xanthine oxidase method,dithiodinitrobenzoic acid colorimetry and Fenton reaction colorimetry in vivo respectively.RESULTS:FFE could inhibit mitochondrial injury and swelling induced by Fe 2+-L-Cys in a concentration-dependent manner and also had significant O ?-__2 scavenging effect.Moreover,the activities of CAT,SOD,GSH-Px and anti-hydroxyl radical in mice liver homogenate were increased significantly by FFE.CONCLUSION:FFE protects mitochondria by scavenging reactive oxygen species,and increasing the activities of antioxidase.So FFE has the pharmaceutical values of antioxidant and anti-aging.
9.Sacral plexus injury and anteroposterior compression zone-Ⅱsacral fractures:a cadaveric study

Objective To study the mechanism of sacral plexus injury resulting from anteroposteri- or compression zone-Ⅱsacral fractures.Methods Six short-term(within one year)embalmed pelves were obtained from Anatomic Department of Hebei Medical University.The preserved sacral plexus and re- sected pubic symphysis were used to make the models of anteropesterior compression zone-Ⅱsacral frac- tures.Quantitative analysis for fracture displacement was carried out to observe the characteristics and mechanism of sacral plexus injury.The removed sacral plexus was replaced by proportional diameter silica gel pips filled with contrast medium.The experimental procedure was repeated under X-ray.The experi- mental data were analyzed by SPSS 10.0 statistic software to obtain result.Results In anteropostefior compression sacral fracture,sacral nerve canal was gradually opened and its volume raised.No sacral nerve was opressed in nerve canal.However ilium external rotation prolonged sacral nerves,especially S_1,S_4 and the more the pubic symphsis was separated the more the sacral plexus nerves were prolonged.At the same time,the sharp border of fracture segment stabbed the nerves,especially L_5(6 cases),S_1(5 cases)and S_2 (3 eases).Conclsions Sacral plexus is tightly connected with the wall of pelvic cavity and fixed. When zone-Ⅱsacral fractures happen sacral plexus is easily damaged.In anteroposterior compression sacral fracture,nerves S_1 and S_4 are likely to be damaged by dragment,and nerve compression injury is attributed to stabbing by fracture segment border,with L_5 and S_1.more hackneyed.
10.Apoptosis of cervical cancer cell line Hela induced by Livin isoform-specific shRNA

Objective: To transfect a recombinant short hairpin RNA(shRNA)expression vector targeting Livin gene isoform(BIRC71,BIRC72)into the cervical cancer cell line(Hela cell),in an attempt to observe RNAi-mediated silen- cing on Livin gene and the induction of Hela apoptosis.Methods: Hela cells were transfected with the recombinant plas- mid pGenesil-1-BIRC71,pGenesil-1-BIRC72 and pGenesil-1-HK via Lipofectamine~(TM)2000.The expression levels of Livin was determined in Hela cells before and after transfection by fluorescence quantitative real-time PCR and Western blotting.
