2.cAMP signaling pathway and multiple myeloma
Journal of International Oncology 2014;41(5):368-370
The clonal proliferations of multiple myeloma cells consist of several signaling pathways. It has been shown that cyclic adenosine monophosphate (cAMP) signaling pathway,an important intra-cellular messenger delivery system,is related to abnormal proliferation and apoptosis of malignant lymphoid cells including myeloma cells.Manipulating the cAMP signaling pathway could induce cell cycle arrest and apoptosis of various malignant lymphoid cells including myeloma cells, which involve mitochondrial-dependent apoptosis and cAMP-modulated changes of intra-cellular mediators.In-depth researches and analysis of cAMP-induced apoptosis in myeloma cells would provide potential targets and possible therapeutic means in the treatment of multiple myeloma.

Journal of Zhejiang Chinese Medical University 2014;(2):227-229,232
[Objective] The better optimization of traditional Chinese medicine for the treatment of respiratory syncytial virus, in the past five years is re-viewed in this paper, also the related domestic experimental research results, and according to the pathogenesis of RSV, it further summarizes the current domestic research for the main direction of the disease. [Methods] The article from the two aspects of compound Chinese medicine and traditional Chinese medicine monomer, according to the pathogenesis of respiratory syncytial virus section, discusses the experiment in vivo and in vitro related rich research results. [Results] It is found by inducing and summarizing, whether individual or compound traditional Chinese medicine, for respiratory syncytial virus re-search has made great achievement, but with experimental study limited to suppress the virus more value-added; replication and inhibiting inflammatory infiltrates the two big aspects, lacking of deeper side of molecular biology research, making the traditional Chinese medicine in the treatment of respiratory syncytial virus research appear too single, not a broader recognition by the industry. [Conclusion] We need to be sure the safety of traditional Chinese medicine for the treatment of respiratory syncytial virus and curative effect, but at the same time must recognize that because of the complexity of its com-position and design of experiment of traditional Chinese medicine, the irrationality of the existence of many factors, it makes progress with many problems, so we must be aware of their own insufficiency, more in-depth explore its mechanism of action, with al kinds of research direction in the future for us.

5.THE CLINICAL OBSERVATION OF THE PATHOGENITIC FACTORS OF HYPERTENSION IN PATIENTS WITH NIDDM
Journal of Xi'an Jiaotong University(Medical Sciences) 1981;0(03):-
A part of pathogenetic factors of hypertensionin twenty--seven patients with hypertensive NIDDM (group 1) were observed. The control groupsincluded non-hypertensive NIDDM (group 2, n=46 ), primary hypertension (group 3, n= 15) andhealthy persons (group 4, n= 15). The oral glu-cose tolerance test (OGTT), insulin releasing test(IRT ), atrial natriuretic peptide (ANP), an-giotensin I (ATI), aldosterone (ALD), wholeblood volume and urine protein were measured. The results showed that the plasma insulin andsodium levels and the whole blood volume weresignificantly higher in group 1 than those in con-trol group 2, the ANP level was rather lower than that in control group 4. Although the results werecorresponded with the theory that the circulatinghyperinsulinemia may lead to sodium retention andinhibited an adequate ANP stimulation by sodiumchallenge which turn to hypertension, nonhyper-tensive NIDDM patients (group 2) also have thesame findinge. So the retention of sodium and wa-ter can't be used to explain the mechanism of hy-pertension in NIDDM patients, the other patho-genetic factors especialy the factors related to thehyperinsulinemia such as PGE and PGI need to beinvestigated futher.

China Oncology 2014;(10):755-760
Background and purpose:Despite the high remission rate in patients with multiple myeloma (MM) after the standard regimen, but often relapsed and resistant. It has been shown that modulation of cAMP can induce cell cycle arrest and apoptosis in a variety of tumor cells, which has become an interesting approach to cancer therapy. This study aimed to investigate possible effects of cyclic adenosine monophosphate (cAMP) analogue 8-(4-chlorophenylthio) adenosine 3’, 5’-cyclic monophosphate (8-CPT-cAMP) on multiple myeloma cells, provide direction to develop new drugs for the treatment of MM. Methods:The myeloma cell line U266 cells were treated with 8-CPT-cAMP of different concentrations. The proliferation of U266 cells was evaluated through cell counting kit (CCK-8) assay, lfow cytometry was used to analyze the changes of cell were distribution, apoptosis rate as well as mitochondrial transmem-brane potential (ΔΨm) in U266 cells before and after the treatment. Meanwhile, real-time quantitative polymerase chain reaction (RT-PCR) and Western blot assay were used to detect expression levels of apoptosis regulators including caspase-8, caspase-9, Bcl-2 and Bax genes in U266 cells before and after the treatment. Results:The U266 cells were treated 5 days with 8-CPT-cAMP of different concentration, it was shown that 8-CPT-cAMP could signiifcantly inhibit cell growth of U266 cells in a concentration and time dependent manner, the IC50 of 8-CPT-cAMP was reduced obvious prolonged reaction time, and reached to 58.52μmol/L in the iffth day. The cell cycle of U266 cells was stopped in G0/G1 stage as the progress of concentration. It was showed statistical signiifcant difference associated with the cellular proliferation inhibition rate and apoptosis rate in different concentration and control (P<0.05). Meanwhile, 8-CPT-cAMP could induce mitochondrial transmembrane potential collapse in U266 cells. Compared with control groups, the levels of Bcl-2 mRNA transcripts and protein in U266 cells were reduced in 8-CPT-cAMP treated groups (P<0.05), while the levels of caspase-9, Bax mRNA transcription and the expression of Bax protein were increased in treatment groups, but the caspase-8 mRNA had no statistical signiifcant difference with controls. Conclusion:8-CPT-cAMP can inhibit the proliferation and promote apoptosis of myeloma cells, which might be mediated by caspase via mitochondrial pathway.

Objective The purpose of the present study was to develop an animal model of type II diabetic coro-nary heart disease in Zuker diabetic fatty ( ZDF) rats. Methods The ZDF rat model of type II diabetic coronary heart disease was prepared by high-fat diet feeding combined with continuous injection of isoprenaline hydrochloride (ISO) in a dose of 1 mg· mL-1 for 10 consecutive days.Serum creatine kinase (CK), and creatine kinase izozyme (CK-MB), ST segment in electrocardiogram ( ECG) and myocardial pathological changes were detected to evaluate the rat model. Results CK of both the control and model groups was gradually increased with ISO injections, while CK-MB increased first and then decreased. The ST segment in ECG part II had significant changes. The pathological examination found that about half of the myocardial cross section in the model group was necrotic after injections of ISO for 5 days and more than 3/4 of the my-ocardial cross section was necrotic after injection of ISO for 10 days. The results indicated that ISO caused myocardial inju-ry in ZDF rats. Conclusions The variation of CK-MB, CK, ST segments in ECG and myocardial necrosis indicate that the model is successfully established. The use of high-fat diet combined with continuous injection of isoprenaline hydrochlo-ride in a dose of 1 mg· mL is a simple way to develop a Zuker diabetic fatty rat model of type II diabetic coronary heart dis-ease.
