1.An analysis on the diagnosis and treatment of abdomen postsurgical gastroparesis syndrome

Objective To summarize the clinical features,diagnosis and treatment principles of abdomen postsurgical gastroparesis syndrome(PGS).Methods The clinical data of 28 patients with abdomen postsurgical gastroparesis syndrome,collected from our hospital in the past ten years,were analyzed retrospectively.Results All 28 patients were cured by conservation therapy.The average gastric dynamics response time was 20.2 days(12.0~34.0 days),which included 2 cases recovered within 1-2 weeks,17 cases recovered within 2-3 weeks,7 cases recovered within 3-4 weeks,2 cases recovered over 4 weeks.Conclusion PGS is a functional reaction rather than mechanical obstruction disease.The diagnosis mainly depends on the symptoms and sighs combined with gastrointestinal angiography or endoscopy.Most of the patients can be cured by conservative treatment.
2.Dissolvability of thundergodvine extract

AIM:To investigate the dissolvability of thundergodvine extract(Tripterygium wilfordii) and triptolide in different dissolvent in transdermal drug delivery system and to study the relationship among dissolvability of extract,and its active component and mean diameters of suspension. METHODS:To determine dissolvability of extract,dissolvability of active component and mean diameters by precipitation method,dissolvability of active component method and mean diameters method. RESULTS:① solvents in sequence of dissolvability of extract,alcohol,isopropanol,ethyl,IPM,water,glycerol;② solvents in sequence of dissolvability of active component alcohol,isopropanol,ethyl,IPM,water,glycerol;③ mean diameters of suspension in sequence:IPM,ethyl,alcohol,isopropanol. CONCLUSION:① The change in dissolvability between extract and active component is identical;② The relationship between the changes in mean diameters and the dissolvent of active component is negative correlation on the whole.
3.Pharmacokinetics character of propofol during anhepatic period of pediatric liver transplantation

Objective: To investigate pharmacokinetics of propofol during anhepatic period of orthotopic liver transplantation (OLT) in children. Methods: In 5 children undergoing OLT, 2 mg/kg propofol was injected intravenously at the beginning of induction and anhepatic periods respectively. The plasma concentration of propofol was measured by reverse-phase high performance liquid chromatography(RP-HPLC), and the pharmacokinetics character of propofol between anhepatic and inducing periods were compared. Results: Propofol had wide distribution and high clearance, and its plasma concentration-time curves were fitted to a three-compartment open model. Conclusion: There is no difference of propofol metabolism between anhepatic and inducing periods, suggesting that there must be notable extra-hepatic metabolism when propofol used in children undergoing OLT.
4.Matrix metalloproteinase-1 and matrix metalloproteinase-3 polymorphisms in promoter regions and ovarian cancer susceptibility:a Meta analysis

Objective To evaluate the association between matrix metalloproteinase‐1 (MMP‐1)‐1 607 bp 1G/2G and matrix metalloproteinase‐3(MMP‐3)‐1 171 bp 5A/6A polymorphisms in promoter regions and susceptibility to ovarian cancer by Meta‐a‐nalysis .Methods Case‐control studies with regards of relationship between MMP‐1‐1 607 bp 1G/2G ,MMP‐3‐1 171 bp 5A/6A pol‐ymorphisms in promoter regions and susceptibility to ovarian cancer were searched in electronic databases .Odds ratio (OR) and 95% confidence interval (CI) were used to evaluate the association between polymorphisms and susceptibility .RevMan5 .0 software was applied for data analysis .Results Eight studies ,containing 5 studies for MMP‐1 and 3 studies for MMP‐3 ,were selected .For MMP‐1‐1 607 bp 1G/2G ,OR(95% CI)=0 .95 (0 .74-1 .21) ,P=0 .67 under 1G/1G :1G/2G+2G/2G model ,OR(95% CI)=0 .93 (0 .70-1 .23) ,P=0 .60 under 1G/1G :2G/2G model ,OR(95% CI)=0 .99 (0 .87-1 .14) ,P=0 .91 under 1G :2G model;for MMP‐3‐1 171 bp 5A/6A ,OR(95% CI)=0 .97(0 .68-1 .38) ,P=0 .85 under 5A/5A+5A/6A :6A/6A model .Overall ,there was statisti‐cal significance .Conclusion It is still not confirmed that significant association between the MMP‐1‐1 607 bp 1G/2G and MMP‐3‐1 171 bp 5A/6A polymorphisms in promoter regions and susceptibility to ovarian cancer exists in current literature .
5.The clinical significance of early nutritional support in severe head-injured patients

Objectives:To evaluate the clinical significance of different early nutrition support in severe head injured patients. Methods:140 cases(GCS≤8) were randomly divided into 5 groups.From A to E,each one had 28 cases.The patients in group A received early parenteral nutrition(PN) and enteral nutritoin(EN) together;group B received early PN,and EN after 1 week later;group C received PN only for more than 2 weeks;group D received early EN only;group E received traditional delayed EN.The clinical observation and statistical comparison were taken. Results:Severe head injured patients could get nutrition from different ways at early stage.A and B groups had better outcomes,and the blood glucose and nutritional data were superior to those of D,E group( P
6.Preliminary study on preparation of Yuanhuzhitong HBS Tablet

Objective: To study the preparation and quality control methods of Yuanhu Zhitong HBS Tablet. Methods: the suitable excipients and formulation process of Yuanhu Zhitong HBS Tablet is sieved by means of testing quantity of tetrahydropalmatine through TLC UV. Results: the beginning floating time of HBS Tablet is 3min; relative dissolution of main component tetrahydropalmatine is 32% within 3hr, 86% within 9 hr. Conclusions: the study is found to be effective in the process of Yuanhu Zhitong HBS Tablet.
7.Short hairpin RNA-mediated silencing of APRIL gene inhibits proliferation and metastasis of human colon carcinoma SW480 cells in vitro

Objective To study the influence of short hairpin RNA-mediated inhibition of APRIL gene on proliferation,invasion and metastasis of human colon carcinoma SW480 ceils in vitro. Methods After treatment with shRNA, the expression level of APRIL protein in human colon carcinoma SW480 cells was detected by western blotting. And cell adhesion, cell migration and cell invasion of SW480 cells transfected with sh637 were analyzed respectively as compared with non-transfected SW480 cells and mocktransfectant. Results The expression of APRIL protein in SW480 transfected with sh637 were significantly lower than mock-plasmid groups and untransfected ones (F=42.15, P=0.00). Cell proliferation was markedly inhibited, compared with untransfected groups and negative control ones (F=24.76, P<0.05).And the average absorption of cell adhesion in transfected groups, mock-plasmid and non-transfected ones were 0.343, 0.409, 0.412, respectively. Cell adhesion decreased 39% compared with untransfected groups. Similarly, there was significant difference for cell migration (F=65.53, P<0.01) between transfected groups (The average ceil number in exposed area to migrate was 47.89±13.16) and nontransfected (98.78±23.26) as well as mock-plasmid ones (108.01±39.11). Then, in view of the average absorption of cell invasion between transfected groups(0.58±0.10) and non-transfected (0.94±0.23) as well as mock-plasmid ones(1.11±0.21), a prominently difference for cell invasion was also found between them (F=5.771, P<0.05). Conclusion The results suggest APRIL has a proliferation-inducing effect and probably be involved in cell invasion and cell metastasis of colon carcinoma and it could enhance capacity of invasion and metastasis of SW480 cells.
8.Rat intestinal absorption trait of peimine and peiminine in Thunberg fritillary bulb extract.

To study the in situ intestinal absorption kinetics and compatibility influence of peimine and peiminine in rats, the absorption of peimine and peiminine in small intestine (duodenum, jejunum and ileum) and colon of rats was investigated using in situ single-pass perfusion method and the drug content was measured by HPLC-ELSD. Perfusion rate, pH, concentration of drug, gender and bile duct ligation can significantly affect the absorption of peimine and peiminine, the Ka, and Papp values in the condition of pH 6.8 and pH 7.4 had significant difference (P<0.01), as drug concentration irlcreased, the absorption parameters of peimine and peiminine decreased, Ka and Papp between low concentrations and middle concentrations was significant difference (P<0.01). Verapamil can not affect Ka and Papp of peimine and peiminine which are in the extract (P> 0.05). Bitter almonds and licorice can significantly reduce the absorption of peimine and peiminine with the usual dose (P<0.01), extracted separately and together had no significant difference on Ka and Papp (P> 0.05). Experimental results show that the absorption features of peimine and peiminine are basically the same, both of them could be absorbed at all segments of the intestine in rats and had no special absorption window, and with significant differences between male and female individuals. The absorption of peimine and peiminine complies with the active transport and facilitated diffusion in the general intestinal segments. Bitter almond and licorice can reduce the intestinal absorption rate ofpeimine and peiminine.

Objective:To investigate the protective effects of nuclear factor ?B inhibition on liver graft reperfusion injury during transplantation.Methods:Orthotopic liver transplantation using modified cuff technique was established in rats,animals were divided according to the grafts cold storaged in 4℃ Ringer's lactated solution with(PDTC group)or without 0.1 mol/L PDTC(control group)for 6 h.During the early stage of reperfusion,DNA binding activities of NF ?B in liver grafts were analyzed by EMSA(electrophoretic mobility shift assay),mRNA level of TNF ? and ICAM 1 by RT PCR,and activities of ALT and LDH were also detected.Results:NF ?B in liver grafts was activated at early stage of reperfusion during transplantation; PDTC treated liver displayed lower activation of NF ?B 1 h after reperfusion,whereas no difference was shown between 2 groups 6 h after reperfusion.Up regulation of TNF ? and ICAM 1 transcription,high level of ALT and LDH activities were observed in both groups during reperfusion,whereas the transcriptional up regulation and the activities of ALT and LDH in PDTC group were reduced compared with those of control group( P
