1.Effects of exogenous antisense VEGF-C gene on growth of human gastric carcinoma cell line SGC-7901 in vitro

Objective:To observe the expression of VEGF-C protein in human gastric cancer cells SGC-7901 those transfected by pCI-neo-antisense VEGF-C and investigate whether antisense VEGF-C gene could inhibit the growth of human gastric carcinoma cell line in vitro.Methods:By the liposome infection protocol,We transfected two different plasmids including pCI-neo and pCI-neo antisense VEGF-C into human gastric cancer cell line SGC-7901 cultured in vitro.Then those resistant clones were selected by G418 and expanded in DMEM culture medium.Then we used Immunohistochemical method and Western-blot to detect target genes and its expression.The cell viability was detected by MTT assay.We took the untransfected group as the control.Results:The immunohistochemical method and Western-blot showed that there were no expression of VEGF-C mRNA and its protein in the group that transfected by plasmid pCI-neo antisense VEGF-C.MTT assay showed that the group of transfected with antisense VEGF-C had a lower cell viability than other groups(P
2.Effects of antisense VEGF-C gene on vegetation process of implanted gastric tumor SGC-7901 in nude mice

Objective:To investigate the effects of tumorigenicity and vascular generation of nude mice on antisence vascular endothelial growth factor-C(antisense VEGF-C) gene transinfection of human gastric carcinoma cell by observing the vegetation process of implanted gastric tumor.Methods:30 nude mice were divided into three groups randomly:transgenic anti-sense VEGF-C group,transgenic pure liposome group and non-transgenic group.The cell suspension of gastric carcinoma cells SGC-7901 non-transinfected or transinfected by anti-sense VEGF-C gene or pure liposome were subcutaneously injected into those mice.After injection,the size and growth velocity of those neoplasma were detected.The Microlymphatic vessel density(MLVD) and microvessel density(MVD) of tumor were detected too.Results:The neoplasma grew more slowly and was smaller in the mice in those transinfected by anti-sense VEGF-C gene than in non-transgenic ones(P0.05).Conclusions:The antisense VEGF-C gene suppress the tumorigenicity and lymphangiogenesis in the human SGC-7901 gastric carcinoma cell apparently.but it had little effects on angiogenesis of the tumor.VEGF-C might participate the lymphangiogenesis of the gastric tumor.
3.Histone deacetylase inhibitor affected CD4+CD25+Foxp3+ cells in a mouse model of acute graftversus host disease

BACKGROUND:Prevention and treatment of graftversus host disease and elevation of graft survival rate are core problems needed to be solved in alogenic hematopoietic stem cel transplantation. Thus, it is necessary to find a new immunosuppressant. Recent studies showed that histone deacetylase inhibitor has immunomodulatory effects. OBJECTIVE:To observe the effects of histone deacetylase inhibitor SAHA on acute graftversus host disease in mice and the immunomodulatory effects. METHODS: C57BL/6(H-2b)→BALB/C(H-2d) was selected as donor and recipient of complete alotransplantation. At 3, 5, 7, 9 and 11 days after transplantion, mice in the treatment group were intraperitonealy given SAHA (35 mg/kg) (0.2 mL). Mice in the control group were intraperitonealy given sterile water 0.2 mL/time. Flow cytometry, real-time quantitative PCR and pathology were used to compare the clinical manifestations, survival time and CD4+CD25+Foxp3+ cel percentage of acute graftversus host disease in mice of both groups. RESULTS AND CONCLUSION:In the treatment group, the time of acute graftversus host disease was delayed and the extent was reduced and survival time was longer compared with the control group. Survival rate was significantly higher in the treatment group than in the control group (P < 0.05). After transplantation, the proportions of CD4+CD25+Foxp3+ cels gradualy increased with prolonged time in the treatment group. On the contrary, the proportions of CD4+CD25+Foxp3+ cels gradualy decreased with prolonged time in the control group (P < 0.05). Above data suggested that SAHA delayed the occurrence of acute graftversus host disease and lessened the severity of acute graft versus host disease possibly through elevating the proportion of CD4+CD25+Foxp3+ cels.

6.Progress in quality control of Chinese medicine dispensing granules

To clarify the progress of the study on the quality of Chinese medicine dispensing granules,and to provide reference for its further development and application.Referring to the relevant literature about the quality control of Chinese medicine dispensing granules,the main quality control methods,the quality and efficacy of traditional pieces,and the quality of traditional Chinese medicine extracts were analyzed and summarized.At present,the main methods of quality control ofTCM dispensing granule are high performance liquid chromatography and mass spectrometry,infrared spectroscopy.Compared with the traditional decoction and extract of TCM dispensing granule and traditional Chinese medicine composition,quality and effect of each has advantages and disadvantages,it needs further study.This paper reviewed the progress of quality control of Chinese medicine dispensing granules in recent years,and provided a reference for further research and development.
7.Significance of pancreatic stone protein in pancreatic injury and sepsis

Pancreatic stone protein (PSP) is the product of exocrine pancreas mainly synthesized and secreted by the pancreas acinar cells, a small amount secreted by gastrointestinal cells. PSP is one of stress protein family members secreted in the pancreas. PSP increased in acute and chronic pancreatitis, diabetes, inlfammatory bowel disease and gastrointestinal tumors. In recent years, some studies showed that PSP has potential clinical value in pancreatic injury and sepsis. In this article, we reviewed the progress of pancreatic stone protein in pancreatic injury and sepsis.
8.Inlfuence of positional change speed on the recovery rate and discomfort degree in benign paroxysmal positional vertigo patients

OBJECTIVETo explore the influence of positional change speed on the recovery rate and discomfort degree of the patients with benign paroxysmal positional vertigo.METHODSPatients with detailed records from September 2012 to September 2015 were reviewed. 358 patients were divided into quick maneuver (<2s) and slow maneuver (5-10 s) groups randomly based on the time of changing position. The maneuver was done by the same doctor. The discomfort degree during the maneuver was assessed by the visual analog scale.RESULTSThe effective rate of the quick and slow maneuver group is 97.7% and 97.2% respectively, there is no significant differences between the groups (P>0.05). The VAS is 7.70±3.95 and 5.18±3.56 respectively in the quick and slow maneuver group, there is significant differences between the groups (t=3.38,P<0.05).CONCLUSION Positional changing speed does not influence the recovery rate of the BPPV, while the slow maneuver can reduce the discomfort degree.
9.Expression and function of P2X purine receptors in the enteric nervous system

P2X receptors are ligand gated ion channels, extracellular ATP is their natural extracellular ligand. When any of the subtypes of P2X receptors and extracellular ATP are combined, P2X channel opens, allowing cations (calcium, sodium, potassium, etc. ) pass through. Enteric nervous system is composed of the gut myenteric plexus and submucosal plexus, in both of which P2X receptors exist, mediating different effects under physiological and pathological conditions. In this paper, the enteric nervous system,P2X receptor distribution and related functions are reviewed in order to further explore the P2X receptor in the diagnosis and treatment of intestinal diseases.
