1.Detection of cardiac development gene after RNA interference for histone acetylation to MSCs induced by 5-azacytidine

Objective To construct 7 shRNA plasmids targeted to Gcn5 and to explore their role on histone acetylation in regulating MSCs differentiation into cardiomyocytes.Methods MSCs were isolated and passaged.Seven shRNA plasmids were constructed by pGenesil-1(4.9 kb) vector with GFP gene.MSCs were treated with 10 ?mol/L 5-azacytidine respectively for 24 h,2,4 and 7 d and transfected with the plasmids via lipofectamine~(TM)2000.Gcn5,as the marker of the acetylation,and GATA4,as the marker of cardiomyocytes,were detected in MSCs during differentiation with or without shRNA plasmids transfection by RT-PCR.Results MSCs were fibroblast-like but changed morphologically after induction.Seven Gcn5 shRNA plasmids were constructed and identified by restriction endonucleases PstⅠ/SalⅠand DNA sequencing.The expression of Gcn5 and GATA4 could be detected in MSCs without shRNA plasmids tranfection,but no expression after shRNA plasmids transfection.Conclusion Gcn5 plays a very important role in MSCs differentiation into cardiomyocytes,which lays the basis for further research of the importance of acetylation in the regulation of cell differentiation.
2.Discussion on radiation-resistant PP in single-use syringe

It is a mature technology producing single-use syringes by polypropylene(PP). But the traditional sterilization process for single-use syringe pollutes the environment, and the residues endanger the health of users. Irradiation sterilization, instead of traditional sterilization process, is the developing trend. However, the traditional PP does not have the ability of radiation resistance. These methods can improve the radiation resistance ability of PP effectively by adding the stabilizer or floating agent. Toughening or temperature treatment can have the same function. Radiation-resistant PP is introduced in this paper. Furthermore, the developing trends for the materials are discussed.
3.Effects of artemisinin on early - phase response of allergic conjunctivitis in a murine model induced by pollen

? AIM: To investigate the immunosuppression of artemisinin in the early-phase response( EPR) of allergic conjunctivitis in a murine model induced by pollen.?METHODS:Twenty-five Balb/c mice were divided into five groups at random: A: model group, B: artemisinin topical treatment group, C: oral artemisinin group, D:dexamethasone topical treatment group, E: negative control group. Balb/c mice were first sensitized with mixture of ragweed crude extract and complete Freund’s adjuvant by left footpad and root of tails injection at days 0. At days 7 and 14, mice were strengthen sensitized by intraperitoneal injection. Negative control group replacd with a mixture of equal amounts of Freund’s adjuvant and PBS mixture. Treatment time was between 21-27d:group B received 1% topical artemisinin eye drops 4 times per day; group C received artemisinin at doses of 600mg/kg orally once per day; group D received topical dexamethasone eye drops 4 times per day. The model and naive groups replaced with solvent for control. After treatment, mice were excited by ragweed pollen crude infusion drops. Within 1h after the excitation, the eyes were taken for histopathology testing, the serum for detection of specific IgE for ragweed pollen.?RESULTS:After excitation, allergy symptoms of model group were more significantly compared with negative control group and the treatment groups; allergy symptom score, mast cell degranulation ratios and the ragweed pollen specific IgE in serum in the topical treatment groups and oral group were higher than that in the negative control group, but significantly lower than that in model group.?CONCLUSION: Artemisinin topical treatment and oral treatment for mice could inhibit the symptoms of pollen allergic conjunctivitis, early-phase response of mast cell degranulation ratio and specific IgE generation for ragweed pollen, suggesting that artemisinin has some therapeutic effect for pollen allergic conjunctivitis.
4.Excimer laser in situ keratomileusis for treatment of high myopia

Objective To evaluate the effects of the excimer laser in situ keratomileusis (LASIK) for treatment of high myopia.Methods LASIK was performed on 308 eyes of 186 patients who were followed up for one year. The patients were divided into two groups (group A and group B) according to preoperative diopters.Results One year after surgery, in group A(-6.0~-10.0D) the uncorrected visual acuities of 0.5 and 1.0 were in 95.76% and 77.58% of the eyes, the mean residual refraction was -0.32D±0.46D. In group B(-10.25~-20.0D) the uncorrected visual acuities of 0.5 and 1.0 were in 62.24% and 37.06% of the eyes, the mean residual refraction was -0.92D±1.13D.Conclusion It is an effective and safe refraction surgery to correct high myopia with LASIK.
5.Evaluation on the Effect of Serum CEA,Ferritin and Plasma D-D on Lung Cancer Chemotherapy

Objective To investigate changes in the level of serum CEA,Ferritin and plasma D-D of the lung cancer patients suffering before and after the chemotherapy,and evaluate its application value in judging the curative effect of chemotherapy on the lung cancer patients.Methods Chose 120 lung cancer patients diagnosed from September 2011 to September 2014, and determined the curative effect in accordance with the standard for evaluating the curative effect of solid tumors set down by WHO.Chose another group of 120 healthy physical examinees as its control group.Tested the contents of serum CEA, Ferritin and plasma D-D of both groups respectively and analyzed the rules of change in the results of the patients before and after chemotherapy and in different chemotherapeutic effect stages.Results The contents of serum CEA,Ferritin and plas-ma D-D of the lung cancer patients were obviously higher than those of such patients after chemotherapy and those of the control group,and the difference was of statistical significance (t=4.123~14.224,P <0.05).The contents of serum CEA, Ferritin and plasma D-D of the patient at complete remission (CR)stage were lower than those at partial remission (PR) stage,stable disease (SD)stage and progress disease (PD)stage,and the difference was of statistical significance (t=4.230~12.336,P <0.05).Conclusion The test on the serum CEA,Ferritin and plasma D-D of the lung cancer patients before and after chemotherapy is of certain clinical value and the change in their content levels can be taken as a reference index for the chemotherapeutic effect of lung cancer patients.
6.Application of NSE in Diabetic Peripheral Neuropathy

Objective To investigate application value of changes in serum neuron-specific enolase (NSE)in early diagnosis of diabetic peripheral neuropathy (DPN).Methods 160 cases of diabetes were recruited from January 2013 to December 2014, 120 of which were combined with DPN according to the 2009 “DPN Diagnostic & Treatment Guidelines”(the DPN group), and the other 40 were not (the DM group).Patients were further stratified into mild,moderate,and severe DPNs according to the Toronto Clinical Scoring System (TCSS).Another 160 healthy subjects were recruited as control.Serum NSE level was determined by chemiluminescence immunoassay,and was compared among the DPN group,the DM group,and the con-trol group.Serum NSE levels were also compared among DPN patients with different serum glucose levels and different TC-SS grading.Results Serum NSE levels (μg/L)of the DPN group,DM group,and control group were 23.5±3.0,12.4±2.3 and 11.3±1.6 respectively,and the difference between DPN and the other two groups was significant (t=12.312,14.523, P <0.05).DPN patients with serum glucose levels of 3.9~6.1,6.1 ~8.0,and ≥8.0 mmol/L had NSE levels of 22.1 ± 1.6,23.6±1.2 and 25.0±1.5 respectively,and the difference was significant (t =4.021 ~ 7.534,P <0.05 for all).DPN patients with mild,moderate,and severe DPN had NSE levels of 21.6±1.1,23.1±1.5,25.2±1.3,and the difference was significant (t=3.320~5.526,P <0.05 for all).NSE level was positively correlated with TCSS score (r=0.572,P <0.05). Conclusion Serum NSE level is closely correlated with DPN,and can be used as a key indicator for early diagnosis of DPN.
7.Significance of serum homocysteine in patients with diabetes mellitus complicating proliferative retinopathy

Objective To evaluate the role of serum homocysteine (HCY) in diabetes mellitus(DM ) complicating proliferative diabetic retinopathy (PDR) .Methods 174 patients with DM were selected and divided into PDR ,non‐PDR(NPDR) and non‐dia‐betic retinopathy (NDR) groups .The serum HCY level was detected .Results Serum HCY levels in the PDR ,NPDR and NDR groups were (18 .20 ± 5 .60) ,(14 .40 ± 6 .70) and ,(12 .10 ± 6 .80)μmol/L respectively ,which was highest in the PDR group (P<0 .05) .Conclusion HCY could result in PDR generation possibly by injuring the retinal blood vessels .
8.The correlations between the leptin levelsand the prevalence of cervical cancer

Objective To study the correlations between the leptin levels and the prevalence cervical carcinoma. Methods Blood samples were collected from 79 patients with cervical squamous cell carcinoma, 28 patients with cervical intraepithelial neoplasia and 35 healthy donors. The serum leptin concentrations were measured by enzyme linked immunosorbent assay. Meanwhile, tissue samples were obtained from the patients with cervical cancer and cervical intraepithelial neoplasia, Then the expressions oleptin in these tissues were detected by Western blot. Results The level of serum leptin in the patients with cervical cancer was significantly higher than those both in the patients with cervical epithelial tumor and in the healthy control group. the Western blot results also confirmed that the the expressions of leptin in the tumor tissues were atwas the highest level. Conclusion The elevated levels of leptin in serum and tumor tissues of patients with cervical cancer suggest that leptin may be involved in the occurrence and development of cervical cancer.

Despite the continuous improvement of cardiopulmonary bypass(CPB),renal dysfunction remains a frequent complication of cardiac surgery.Previous studies reported that the incidence of acute renal dysfunction following CPB was 7%-40%,while that of acute renal failure(ARF) was 1%-10%.In those who developed ARF,the prognosis was poor,with mortality of about 40%-80%.The present paper reviews the mechanism,risks and available preventive measures of acute renal injury following CPB.
