In this study we implanted magnetically labeled neural stem cells (NSCs) in PD rats and then monitored their survival and migration in the host brain by magnetic resonance imaging (MRI). The mesencephalic NSCs were obtained from the brain of SD rats. Superparamagnetic iron oxide (SPIO) was transferred to NSCs by Lipofectamine transfection. Eighteen PD lesioned rats were selected for transplantation by evaluation of their rotational behavior in response to amphetamine and randomly assigned to 3 groups, i.e., sham group, PBS group and NSCs transplanted group, with 6 rats in each group. MR scanning was performed at 1, 2, 4, 6, 8 and 10 week(s) following transplantation. At the meantime, rotational behavior was assessed in each group. Our results showed that SPIO particles were clearly visible with Prissian blue staining in neurospheres and cells derived from NSCs. The rotational behavior of the NSCs transplanted group was remarkably improved compared with that of sham group and PBS group (P < 0.05). In vivo MR tracking of NSCs showed that SPIO labeling led to a strong susceptibility change of signal 1 week after transplantation on T2 weighted images. And a large circular hypointense signal appeared in the transplanted area on T2* gradient echo images. Ten weeks following transplantation, the hypointense signal on T2 weighted and T2* gradient echo images was still displayed. It is concluded that SPIO particles could label NSCs effectively, and MRI detection of SPIO labeled cells is a promising method and novel approach to analyzing the NSCs following transplantation in the treatment of PD.

BACKGROUND: More and more patients with periodontal disease require orthodontic treatments. Thus, the remodeling process and its mechanism of inflammatory periodontal tissues become a hot point during orthodontic tooth movement.OBJECTIVE: To observe the remodeling of inflammatory periodontal tissues during orthodontic tooth movement.METHODS: A total of 50 Sprague Dawley rats were randomly divided into the control and periodontitis groups. In the periodontitis group, rats were established periodontitis models. After that, all rats were prepared for orthodontic tooth movement models. The remodeling of periodontal tissues was observed by hematoxylin-eosin staining at 1, 3, 5, 7 and 14 days after orthodontic tooth movement.RESULTS AND CONCLUSION: The movement distance of the periodontitis group was greater than that of the control group. At 0-7 days after orthodontic force application, there was obviously bone resorption at the pressure side and the bone formation was inhibited at the tension side; at 14 days after force application, the bone resorption was diminished, associated with large numbers of multinucleated osteoclasts at the pressure sides in both groups. The findings showed that rats with periodontitis suffered more periodontal traumatism during orthodontic tooth movement, thus, treatment should be delayed until the inflammatory signs were controlled and the local inflammatory was eliminated.

Chinese Journal of Tissue Engineering Research 2014;(20):3172-3177
BACKGROUND:Various studies confirm that smoking can contribute to the resorption of periodontal alveolar bone.
OBJECTIVE:To observe the effect of smoking in alveolar bone resorption of periodontitis rat models.
METHODS:A total of 24 rats were randomly divided into three groups. Normal group:rats were normal y fed without any other pre-treatment;Control group:experimental periodontitis model was established using wire ligation method in rats;Experimental group:rat models were given passive smoking during experimental
modeling period. Al rats were sacrificed after 8 weeks of modeling, periodontal tissue were removed. Hematoxylin-eosin staining was used for examining pathological changes in periodontal tissue, and immunohistochemical analysis was done for observing receptor activator of nuclear factor-κB ligand and osteoprotegrin expression.
RESULTS AND CONCLUSION:After 8 weeks of modeling, expression of receptor activator of nuclear factor-κB ligand factor was significantly higher in alveolar bone of rats from experimental group in comparison to control group (P<0.05), whereas expression of osteoprotegerin in alveolar bone was significantly greater in rats from control group when compared to experimental group (P<0.05). This finding suggests that smoking can increase the expression of receptor activator of nuclear factor-κB ligand protein and reduce osteoprotegrin expression in periodontal rats, thus increasing the resorption of periodontal alveolar bone.
