1.Demethylation of progesterone receptor in leukemia cell line induced by DNMT1 antisense oligonucleotide wrapped in liposome

Objective To investigate the effects of the transfection of DNMT1 antisense oligonucleotide wrapped in liposome on the promotor methylation and expression of progesterone receptor in leukemia cell lines so as to find a new pathway of methylation for gene therapy. Methods Artificially synthesized DNMT1 antisense oligonucleotide MG88 wrapped in liposome was transfected into the leukemia cell line. Changes of DNMT1 & PRB mRNA were detected by RT PCR, and the changes of the methylation status of PRA & PRB were analyzed by MSP. Results After transfection of leukemia cell line with MG88, DNMT1 mRNA decreased significantly. PRA & PRB promotors were demethylated, and the expressions of PRB mRNA increased significantly. Conclusion MG88 can induce the demethylation of PR promoters in leukemia cell line, resulting in the increase of PRB mRNA expression. So MG88 can be used in the experimental study of DNA methylation for gene therapy.
2.Acupuncture Treatment of 51 Cases of Menopausal Perspiration

Fifty-one cases of menopause syndrome were randomly divided into three groups, 21 cases in the acupuncture and cupping group, 10 cases in the acupuncture group, and 20 cases in the ear acupuncture group, and were respectively treated by acupuncture plus cupping therapy, acupuncture,and ear acupuncture. The total effective rate was respectively 80.9%, 20.0% and 80.0%. The therapeutic effect was better in the treatment of menopause syndrome by acupuncture plus cupping therapy than by single acupuncture treatment or ear acupuncture (P<0.05).
3.High-resolution ultrasound in predicting extracapsular extension of papillary thyroid cancer

Objective To evaluate the clinical value of high-resolution ultrasound for predicting the extracapsular extension (ETE) of papillary thyroid cancer (PTC).Methods A total of 151 nodules with PTC in 116 patients confirmed by surgery and pathology were enrolled.The ultrasonographic characteristics were retrospectively analyzed,and the percentage of the perimeter of the nodule that abutted the thyroid capsule (A) were observed.Taking A≥1%,A≥25 % and A≥ 50% as cutoff value respectively,the ETE of PTC was predicted.Results Of the 151 nodules with PTC,ETE presented in 59 (59/151,39.07%),while no ETE (non-ETE) was observed in 92 (92/151,60.93%).Nodules in the thyroid parenchyma (A0) which could be observed with ultrasound was 84 (84/151,55.63%),and 1%≤A<25%,25%≤A<50%andA≥50% was 25 (25/151,16.56%),15 (15/151,9.93%),and 27 (27/151,17.88%),respectively.There were statistically significant differences of nodules between the non-ETE and ETE when A≥1%,A≥25 % and A≥50% (all P <0.01).Taking A=50% as the cutoff value for predicting the presence of ETE,the sensitivity,specificity,accuracy,positive predictive value and negative predictive value was 37.29% (22/59),94.57% (87/92),72.19% (109/151),81.48% (22/27) and 70.16% (87/124),respectively.Conclusion High frequency ultrasonography can effectively predict whether PTC has been invaded,which provides important reference information for preselected surgical approach and prognosis of patients.

6.Effects of Electroacupuncture on Adrenalin and Noradrenalin Levels of Acute Hypertension Rats

Objective To explore the effects and mechanism of electroacupuncture on acute hypertension in rats. Methods 36 male Wistar rats were randomly divided into control group (n=8), model group (n=8), drug group (n=10) and electroacupuncture group (n=10). The acute hypertension rat model was induced with injecting adrenalin into abdomen. Blood pressure was measured and blood adrenalin and noradrenalin levels were tested after electroacupuncture in points Baihui DU-20 and Zusanli ST36. Results The systolic and diastolic pressures were lower in the drug group and the electroacupuncture group than in the model group (P<0.05), and there was no difference between the 2 groups (P>0.05). The adrenalin and noradrenalin levels were higher in the model group than in the control group (P<0.05), but they were lower in the drug group and the electroacupuncture group than in the model group (P<0.05). Conclusion The electroacupuncture could reduce the blood pressure of acute hypertension rats by decreasing blood adrenalin and noradrenalin levels.
7.The reliability and validity of the Chinese version of multidimensional fatigue symptom inventory-short form

Objective To assess the reliability and validity of the Chinese version of Multidimensional fatigue symptom inventory-short form(MFSI-SF).Methods The reliability and validity of the Chinese version MFSI-SF were assessed in a sample of 203 cancer patients.Statistical software was used to perform the analysis.Results The results showed moderate correlation between items and the total scale,the content validity index was 0.82,and exploratory factor analysis indicated five dimensions of the scale,the cumulative variance contribution was 56.65%.Confirmatory factor analysis showed moderate model fitting:x2/df=l.73,GFI=0.83,AGFI=0.79,NNFI=0.94,RMSEA=0.06,criterion validity was 0.585,and the Cronbach α of the total scale was 0.896.Conclusions The results demonstrated good convergent validity,it is suitable to evaluate fatigue status in Chinese cancer patients.
8.Evaluation of predicting the ductus arteriosus closure in preterm infants by echocardiography

Objective To investigate the values of echocardiographic parameters for predicting the spontaneous closure of the ductus arteriosus in preterm infants .Methods A retrospective study of 110 preterm infants from October 2013 to October 2014 in our hospital was done .Examination was done at 1 ,3 ,7 d with echocardiography for those infants .Diameter of left atrium(LA) ,aotic root(AO) and arterial canal‐related parameters (the smallest width of ductal color Doppler flow jet and the maximum velocity at the ductus) at first .Preterm infants in this study were divided into two groups .The early patent ductus arterisus group included ones whoes ductus didn′t closed spontaneous in 7 d ,and the control group included ones whoes ductus closed spontaneous in 7 d ,and then the echocardiographic parameters between the two groups were compared .Results (1)The rates of ductus arteriosus sponta‐neous closure in preterm infants at 3 ,7 d were 70 .9% (78/110) ,78 .2% (86/110) ,respectivly .(2)When compared with the control group ,The rate of LA/AO in patent ductus arterisus group were higher than that of the control group(P<0 .01);The smallest width of ductal color Doppler flow jet in patent ductus arterisus group were bigger than that of the control group(P<0 .01);The maximum velocity at the ductus in patent ductus arterisus group were lower than that of the control group(P<0 .05) .(3)The best critical points of the LA/AO ,the smallest width of ductal color Doppler flow jet and the maximum velocity at the ductus of the spontaneous ductus arteriosus closure in preterm infants were 1 .32 mm ,2 .56 mm and 185 .5 cm/s ,respectivly .Conclusion Echo‐cardiography plays a significant role in prediction of the spontaneous closure of the ductus arteriosus in preterm infants .
9.Emergency Interventional Therapy in Digestive Tract Bleeding

Objective To study the methods and value of emergency interventional therapy in digestive tract bleeding.Methods 61 cases with digestive tract bleeding accepted emergency angiography.According to find out positions and causes of bleeding during angiography,these patients accepted arterial embolization and /or perfusion of vasoconstrictor substance.Results In 29 cases accepted arterial embolization and 32 cases accepted perfusion of vasoconstrictor substance,the stoped bleeding immediately occured in 100% and 82.7% respectively.Bleeding recurrence was 3 cases in the patients with arterial embolization one week later and 25 cases of the patients with perfusion of vasoconstrictor substance forty-eight hours later.Conclusion To treat the digestive tract bleeding by arterial embolization or vasoconstrictor substance perfusion are safe and effective hemostatic ways during emergency angiography.Though the bleeding recurrent rate is high after vasoconstrictor substance perfusion,these ways can race against time for surgical operation.
10.Histomorphological assessment of a rat model of skeletal muscle injury induced by local injection of bupivacaine

BACKGROUND: The neurotoxicity and cardiotoxicity of bupivacaine have been reported frequently. However, the studies on bupivacaine-induced muscle toxicity are few.
OBJECTIVE: To establish and evaluate local intramuscular injection of bupivacaine on the changes in histomorphology and ultrastructure of rat multifidus muscle at various time points.
METHODS: A total of 54 male Sprague-Dawley rats weighing 280-320 g were randomly divided into black group (n=18), model group (n=18) and model control group (n=18). Each group was then equal y subdivided into three subgroups according to time points (4, 7 and 14 days) (n=6). Both sides of multifidus muscle of the rats (L4 and L5) were injected with 0.5% bupivacaine. The morphological and ultrastructural changes of multifidus muscle were observed and analyzed with light microscope and transmission electron microscope at 4, 7 and 14 days after model establishment.
RESULTS AND CONCLUSION: (1) A single intramuscular injection of 0.5% bupivacaine induced muscular damage. (2) Hematoxylin-eosin staining results showed fiber necrosis, inflammatory cel infiltration, and a smal amount of macrophages in local skeletal muscle. (3) Under the transmission electron microscope, the structure of myofibrils was destroyed or disintegrated; kinds of bands and lines were indistinct, disrupted or disappeared; the structure of mitochondria was abnormal, the mitochondrial cristae were reduced or disappeared. In the 7- and 14-day groups, multifidus muscle proliferated and repaired. (4) Ultrastructural change scores in skeletal muscle were significantly higher in the model group than in the blank and model control groups (P < 0.05). Above scores were significantly greater in the 4-day group than in the 7- and 14-day groups (P < 0.05), and higher in the 7-day group than in the 14-day group (P < 0.05). (5) Results suggest that a single intramuscular injection of 0.5% bupivacaine can result in pathological changes of skeletal muscle from morphology and ultrastructure. This method can establish a suitable model of skeletal muscle injury.
