1.ISOLATION, IDENTIFICATION OF LAS-DEGRADING FUNGI AND THEIR BIODEGRADATIVE CHARACTERS

From the LAS-polluted soil were isolated 12 strains of LAS-degrading fungi. They belong to Penicillium, Aspergillus, Scopulariopsis and Cephalosporium. Conditions for the induced formation of LAS-degrading enzymes and their characters of degradative reaction were studied. The LAS-degraded products by fungi and bacteria were analysed and compared.
2.PRELIMINARY OBSERVATION ON T LYMPHOCYTES IN PERIPHERAL BLOOD OF PATIENTS WITH CHRONIC ATROPHIC GASTRITIS AND GASTRIC CANCER BY DEMONSTRATION OF ACID ?-NAPHTHYL ACETATE ESTERASE

A study of the peripheral blood T lymphocytes of patients with chronic atrophic gastritis (mainly of B type) and gastric cancer was undertaken by demonstrating the acid ?-naphthyl acetate esterase in lymphocytes as a T cell marker. A scoring method was also designed by the authors to estimate the esterase activity according to the size and number of the esterase particles. The T lymphocyte levels, i.e. ANAE%, of gastric cancer (46.2?1.87%) and chronic atrophic gastritis (54.4?1.48%) were significantly lower than that of normal controls (72.7?5.9%). The T lymphocyte levels of gastritis patients, however, were significantly higher than that of cancer patients. Based on the above findings, it is suggested that the changes of cell-mediated immunity may play a role in the etiopathogenesis and progression of B type atrophic gastritis.
3.Differential Identification of Genuine and Spurious Panax notoginseng by Near Infrared Spectra

OBJECTIVE:To establish a method for rapid identification of genuine and spurious Panax notoginseng.METHODS:19 batches of samples including P.notoginseng of different origins and the common spurious P.notoginseng in China were collected.The infrared spectra of the samples were collected using near infrared spectrometer,then the spectra were pretreated by unitary vector plus second derivative method and the identification model was established by factorization method.RESULTS:The verification on 10 batches of commercial P.notoginseng samples showed that the established model can accurately identify the genuine and the spurious P.notoginseng.CONCLUSION:NIR spectrometry can quickly and accurately identify the quality of P.notoginseng,and it can be applied in rapid drug testing vehiches for extensive use.
4.Leukocyte Chemotaxis in Filter Assay and Its Affected Factors

In this paper, polymorphonticlear leukocyte chemotaxis filter assay and its affected factors have been studied in accordance with the practical conditions.The results indicated that leukocyte chemotaxis may be detected by using Boycen chemotaxischamber prepared in our laboratory and filters made of mixed esters of cellulose athome.The appropriate experimental conditions are as follows:Single filter methodselected the filter of 3?m pore size.The cells were suspended in Eagle medium at aconcentration of 1?1.5/ml.The chamber was incubated at 37℃ in humidified airfor 120 min.Optimal concentration of bacterial culture filtration,zamsan activatedserum and lymphocyte derived chemotactic factor had eqeal cytotactic potency. Leukocyte chemotaxis is quantitated by counting the number of migrating cells atvarious depths within the filter.The mean number of migrating cells toward chemo-tactic factor is usually about twice as many as that of medium of control.Themodified method which may be acted as an index for investigating leukocyte functionin the experimental immunology and clinical research.
5.Effects of Astragali Injection and extracts from Atractylodes macrocephala on IEC-6 cell migration

Object To observe the effects of Astragali Injection (Hi) and extracts from Atractylodes macrocephala Koidz. (B1, B4, B9, B13) on the migration of rat small intestinal crypt like cells line (IEC 6). Methods IEC 6 cells were inoculated in 6 well microplates and injured in 72 h, and added with Hi, B1, B4, B9, B13. Then D PBS to negative control in the same amount and EGF to positived control, the cell migration was observed in 24, 48 and 72 h after treatment. Methods:492 community peopl e were sampled from community and tested by Generic Quality of L if e Inventory-74.Results:(1) Generic quality of life of comm unity people was well, and their body and mind were in good health; (2) Effects of mental health、physic health and social function on quality life were over ef fect of material life conditions;(3) Qualities of life of elders、workers、non-p rofession people were worse than qualities of young、profession people and cadre (P<0.01).Conclusion:Quality of life of community peop le of Hangzhou was in good condition, but attentions should be paid to qualities of life of elders and workers.
7.Clinical Observation on the Relationship between Helicobacter Pylori and Peptic Ulcer and Chronic Gastritis

Patients with Helicobacter pylori (Hp) positive peptic ulcer were randomly treated with Hp-killing or Hp-nonkilling anti-ulcer drugs, and 19 patients with healed peptic ulcer were followed up to determine the relapse rate of peptic ulcer. It was found that there was a close relationship between Hp infection and the existence of active chronic gastritis, and the clearance of Hp infection could not alleviate the activity of chronic gastritis or influence the 3-week healing rate of peptic ulcer. The relapse rate of the Hp-positive ulcer patients was higher than those Hp-negative cases.
8.Effects of glucocorticoids on intracellular calcium in microglial cells

Aim To explore the effects of hydrocortisone on intracellular calcium in microglial cells.Methods The intracellular calcium was measured by instantaneous scanning with confocal laser microscope(CLM) in BV-2 cells, and fluo3-AM was used to dye the intracellular calcium.Results Both hydrocortisone and nicotine could obviously increase intracellular calcium in BV-2 cells(P<0.05).It was indicated by instantaneous scanning with CLM that hydrocortisone induced the rising of intracellular calcium immediately, and reached the peak about at the fifteenth second, and sustained for 10 seconds, then declined to baseline at 200th second.The effect of hydrocortisone on intracellular calcium exhibited a highly consistency with nicotine.Antagonist of glucocorticoid receptors RU486 could not abolish the rising of intracellular calcium induced by hydrocortisone(P>0.05);but the blocker of α7 nicotinic acetylcholine receptor(α7nAChR) methyllycaconitine could suppress the rising of intracellular calcium induced by hydrocortisone(P<0.05).Conclusion Hydrocortisone enhances intracellular calcium via α7nAChR in microglial cells, which not only demonstrates the non-genomic effect of glucocorticoid, but also suggests that glucocorticoid could serve as endogenous ligand of α7nAChR.

Donguibogam was introduced from traditional Chinese medicine (TCM) instead of the local representa-tive work of the Democratic People's Republic of Korea (DPRK). It is also the concrete manifestation of theories of different schools of doctors according to Dr. Xu Jun's medical thoughts. In this paper, from Dr. Xu's advocacy to Taoism and the concept of aftercare spirit, the exploration was made on health cultivation for different body constitu-tions. According to the health cultivation effects of the medicinals and edible food, the focus was made on the sys-tematic collation and induction of more than 1 400 kinds of herbs contained in the soup liquid from papers in three volumes. The medicinals were divided into eight categories, which were benefiting qi, nourishing yin, tonifying yang, clearing heat and promoting dieresis, drying dampness and resolving phlegm, promoting blood circulation for remov-ing blood stasis, dispersing stagnated liver qi, nourishing blood for hemostasis. Donguibogam specified the health cultivation concept from the correspondence between the body constitution and the medicinal performance. It has a certain guiding significance to the current health cultivation.
10.Identification and characterization of partner proteins interacting with fatty acid activation enzyme Slr1609 in Synechocystis sp. PCC 6803.

To understand molecular modules related to polyunsaturated fatty acids (PUFA) synthesis and eventually produce PUFA at high efficiency, we developed a protein complex analysis technology in Synechocystis sp. PCC 6803, and applied it to identify possible partner proteins interacting with the key enzymes that catalyze PUFA biosynthesis. We first constructed a recombinant expression of protein of slr1609 encoding the fatty acid activation enzyme, by fusing 3xFLAG tag with the target protein. Then we verified its expression by Western blotting targeting 3xFLAG tag. To maximize purification of Slr1609 protein complex, we optimized the protein expression conditions of Slr1609 in Synechocystis in a 5 L fermenter by monitoring its gene expression using RT-qPCR. The purification of the Slr1609 protein complexes was demonstrated by a Native-PAGE analysis. Finally, LC-MS/MS proteomic analysis allowed identification of the possible partner proteins interacting with Slr1609.
