1.Morphological study on skeletal satellite cell transplantation applied in therapy of myocardial infarction

Objective To find the features of cryo-injured infarcted model and the therapeutic effects of skeletal satellites cells(SCs) on myocardial infarction(MI).Methods SCs were implanted into the center of cryo-injured myocardium.Immunohistochemistry,confocal immunofluorescence double-labelling staining and transmission electron microscopy were used to study the state of SCs.Results After cryo-injured frontal wall of the left ventricle(LV) for approximately(15 s) by aluminium rod with(5 mm) in diameter,it made 43% weight of LV ischemia and 17% weight of LV infarcted.At 2nd week,injured area occured as transmural infarct.After transplantation,the implanted cells survived in large numbers.The transplanted cells proliferated remarkably at early phase and differentiated into many multinuclear myotubes after 2 weeks.Some of them developed into muscle fibers with better sarcomeres,but the gap junction was not formed between implanted cells and cardiac myocytes.Conclusions Cryo-injured infarcted model is a proper model to study myocardial infarction.Skelatal satellite cell transplantation will repair morphological structure partly of myocardial infarction area.

3.Application of serum procalcitonin and high-sensitivity C-reactive protein in the early evaluation of inflammatory disease

Objective To explore the clinical applications of the PCT and hs-CRP in inflammatory diseases.Methods The sera of 79 infectious patients were randomly collected.54 samples were bacterial infectious patients,25 samples were nonbacterial infectious patients.The serum levels of PCT and hsCRP in 79 infectious cases were detected by immune fluorescence and Immunoturbidimetry respectively.Results The positive rate of PCT in bacterial infectious patients were much higher than that in nonbacterial infectious patients (x2 =48.337,P < 0.01),and the sensitivity was 92.5%,the specificity was 88.0% ;The positive rate of hs-CRP in bacterial infectious patients were much higher than that in nonbacterial infectious patients (x2 =22.347,P < 0.01),and the specificity was 88.9%,the specificity was 56.0%.The positive predictive value (88.9%) and negative predictive value (70.0%) of hs-CRP were lower than those of PCT (positive predictive values of 94.3% and negative predictive value of 84.6%).Conclusion Theserum levels of PCT in inflammatory diseases were significantly increased with an high sensitivity and high specificity.Thus,it may be used as a new marker of inflammatory,which can be helpful in the early evaluation of infectious disease and the observation of curative efficacy.
4.Detection of HBV DNA in kidneys of HBV-associated glomerulonephritis using in situ hybridization and in situ PCR technique

Objective: To detect the localization of HBV DNA in kidneys of HBV-associated glomerulonephritis, and investigate the pathogenesis of HBV-associated glomerulonephritis. Methods: 45 cases of renal biopsy specimens (38 cases had glomerular deposition of HBV antigens, 7 cases were negative for serologic and histologic HBV antigens) were examined for HBV DNA by in situ hybridization (ISH) and in situ PCR (IS-PCR). Results: The positive rate of HBV DNA in renal biopsy was 71% (27/38) in patients with glomerular deposition of HBV antigens. HBV DNA was found in 19 cases (19/26, 73%) of HBV-associated glomerulonephritis; and in 8 cases (8/12, 67%) of IgA nephropathy and lupus nephritis with glomerular deposition of HBV antigens. HBV DNA was detected mainly in the cytoplasm of tubular epithelia with ISH; IS-PCR showed that HBV DNA was localized not only in cytoplasm of tubular epithelia, but also in nuclei of tubular epithelia, in nuclei and cytoplasm of glomerular epithelia and mesangial cells, and in GBM. Conclusion: Our study suggested the presence of HBV infection and replication in glomerular cells and renal tubular epithelia, indicating an etiologic role of HBV in HBV-associated glomerulonephritis.
5.Application of Emotional Problems Factor of Chinese Personality Assessment Inventory II in Anxiety Disorders Patients

Objective:To evaluate the reliability and validity of Emotional Problems Factor of Chinese Personality Assessment Inventory II(CPAI2-E).Methods:Item analysis of the test was carried out;internal consistency reliability, retest reliability and structure validity, differential validity were examined.Results:①Through factor analysis, 20 subordinate factors were got.②The Cronbach ? coefficient of the total was 0.948 and each factor ranged from 0.755 to 0.896;re-test reliability of the total test was 0.718.Conclusion:CPAI2-E can be used as an effective and reliable tool for assessing anxiety disorders.

The stability of cell genetic material is influenced by a variety of factors, both internal and external, which can cause various types of DNA damage, such as DNA alkylation, oxidation, mismatching, loop structure, atypical DNA structure, single-strand break, and double-strand break.These DNA damages disrupt cellular homeostasis and dynamic equilibrium, which cause gene mutations, chromosomal abnormalities, and even degradation, aging, and death at different biological levels.By searching and identifying DNA damage sites, the cell activates a series of biochemical pathways, coordinates the progress of DNA replication and transcription, and then repairs the DNA damage.In this way, the cell maintains its independence and stability.While radiotherapy plays a role in eliminating tumors by DNA damages, it also initiates DNA damage responses.Among the responses, base excision repair, nucleotide excision repair, mismatch repair, double-strand break repair, and post-translesion synthesis repair play a key role in repairing the damages.The dysfunction of these repair pathways will cause differences in tumor radiation sensitivity.This paper summarizes recent research results in DNA damage repair, and focuses on the types of DNA damage and their repair mechanisms, so as to promote the understanding of the great significance of this field and to provide a theoretical basis for exploring the application of DNA damage repair pathways in tumor therapy.
8.The therapeutic effects of oxygen inhalation combined with relaxation training on generalized anxiety disorder

Objective To observe the therapeutic effects of oxygen inhalation combined with relaxation training on patients with generalized anxiety disorder (GAD).Methods A total of 188 GAD patients were divided into the study group and the control group randomly.The two groups were treated with conventional antianxiety drugs (paroxetine),in addition to conventional medical therapy.The study group was aided with oxygen inhalation and relaxation training,the control group was given benzodiazepine.The scores of Hamilton anxiety rating scale (HAMA),global improvement(GI) [ a part of clinical global impression (CGI) ],and treatment emergent symptom scale (TESS) were used to evaluate clinical effects and adverse reactions before and after 4-week treatment.The indexes of breathing,heart rate and blood pressure were evaluated before and after treatment.Results The scores of HAMA scale and GI in the two groups were significantly different after treatment( P < 0.05 ),and the study group changed significantly( P < 0.05 or P < 0.01 ). The adverse reactions of the two groups were slight,there was no significant difference of TESS before and after treatment ( P > 0.05 ). There was no significant difference in terms of breathing,heart rate and blood pressure between the two groups before treatment( P > 0.05 ),but were all lower after treatment the study group changed more apparently and significantly(P < 0.01 ).Conclusions Based on conventional drugs treatment oxygen inhalation combined with relaxation training could improve anxiety and somatization rapidly,there was no obvious adverse reaction and the patients compliance was good,so the treatment was worth to use in clinic.
9.The clinical-pathological characteristics and prognosis of IgA nephropathy patients with moth-eaten GBM lesions

Objective To investigate the clinical and pathological characteristics of IgAN with moth-eaten lesions of GBM.Methods Seven hundred and fifty-six primary IgAN patients(from 1997.1 to 2003.12)were divided into two groups according to the ultrastructural changes of GBM:the IgAN with moth-eaten GBM lesions group(24 cases)and the IgAN without moth-eaten GBM lesions group(101 cases randomly selected).The moth-eaten GBM lesions revealed a local dilatation of irregular outline.Small fragments of the splitting or branching lamina densa were occasionally seen in the swollen GBM.Results The incidence of moth-eaten GBM lesions in IgAN were 3.1%.There were more severe hematuria and proteinuria[(3.5?2.5)g/d vs(2.1?2.4)g/d]and higher percentage of glomerular sclerosis(62.5% vs 49.5%)in moth-eaten GBM lesions group than in the non-moth-eaten GBM lesions group(P﹤0.05).During mean 27 months'follow-up in the 17 patients,none of them showed worse outcome.Conclusion This result suggests that moth-eaten GBM lesions relate to more severe clinical manifestations and pathological changes but further studies are required to clarify the influence of moth-eaten GBM lesions on the prognosis of IgAN.
10.Study on length of preserved ileocecum by subtotal colectomy with antiperistaltic cecorectal anastomosis

Objective To investigate the length of preserved ileocecum in surgical treatment of slow transit constipation ( STC) by sub-total colectomy with antiperistaltic cecorectal anastomosis .Methods A total of 82 patients with STC were divided into two groups according to the random number table method ,with 41 cases in each group ,all the patients of the two groups underwent subtotal colectomy ,intraopera-tive ileocecum was preserved length of group A was 10~15 cm,group B was 2~3 cm.The operation time,intraoperative bleeding volume, postoperative exhaust time and length of hospital stay were compared .Wexner constipation score and gastrointestinal quality of life index ,ab-dominal pain ,frequency score and emptying time of ileocecus before and after 6 months and 12 months between the two groups were com-pared.Results There was no statistically significant difference in the operation time ,intraoperative bleeding volume ,postoperative exhaust time and hospitalization time of two groups (P>0.05).Wexner constipation score,abdominal pain,frequency score of the two groups after 6 months and 12 months decreased significantly (P<0.05,P<0.01),of which the group B was lower than that of the group A (P<0.05,P<0.01). The gastrointestinal quality of life index after operation significantly increased (P<0.01),of which the group B was higher than that of the group A (P<0.01).Ileocecal emptying time of the group B 12 months after operation was shorter than that of the group A (P<0.01),the differences were statistically significant .Conclusion Subtotal colectomy with antiperistaltic cecorectal anastomosis is an effective method to treat STC,which can reduce the length of preserved ileocecum and improve the prognosis of patients .
