Objective To investigate the changes of Claudin-3 and Claudin-11,two key components of blood-testis barrier (BTB) on male infertility induced by γ-ray irradiation.Methods Fortytwo KunMing male mice (20-25 g) were divided into one control group,three γ-ray irradiation groups and three estrodiol (E2) intervention groups randomly:Group A,sham controlled; the lower abnominal and scrotal area of the mice in Group B,C,D were irradiated with single dose of 2,6 or 10 Gy 60Co γ-ray after anaesthetizd; 17β-estradiol intervention were initiated in Group E,F,G after 6 Gy γ-ray irradiation via hypodermic injection for 4w at the dose of 1,2,4 μg/d,respectively.Mice were sacrificed 2 w after the last E2 administration.The tubule differentiation index (TDI) was counted in testis sections.InhibinβB,Claudin-3 and Claudin-11 transcription levels were assayed with semiquantitative real time PCR.Claudin-11 protein levels in testis were generated by western blot.Results Compared with sham control group,TDI in three γ-ray irradiation groups were markedly reduced (68.5 ± 6.4,35.0± 6.1,16.3 ± 5.7 vs 100.0,all P＜0.05).InhibinββB mRNA expression level in testis of gourp D was markedly decreased (0.5±0.2 vs 1.0±0.1,P＜0.05).Claudin-3 mRNA levels of group C and D were up-regulated to 2.17 and 3.49 times,respectively.Claudin-11 protein levels were significantly increased to 2.18 and 2.23 times.Compared with group C,TDI in three E2 intervention groups were improved,which were obvious in group F and G (61.7±7.2,55.8±11.9 vs 35.0±6.1,P＜0.05).The InhibinβB mRNA levels were increased,though there were no significant differences (all P ＞0.05).Claudin-3 mRNA levels in group F and G were down-regulated (1.3± 0.2,1.6±0.3 vs.2.2 ± 0.2,all P＜0.05).In group F significantly reduced mRNA level and protein level of Claudin-11 were observed (mRNA:1.2±0.2 vs.1.8±0.2,P＜0.05; Protein:1.5±0.5 vs.2.2±0.3,P＜0.05).It was negatively correlated TDI with mRNA expression levels of Claudin-3 and Claudin-11 in the irradiated testis (rs =-0.884,P＜0.05; rs=-0.758,P ＜0.05,respectively).Conelusions Irradiation could elevated the expression of claudin-3 and claudin-11.

Aim To investigate the changes of heme oxygenase-1(HO-1) in cardiomyocytes after acute myocardial infarction and the impact of simvastatin on HO-1 expression in rats.Methods Myocardial infarction models were made by anterior descending coronary artery ligation on male SD rats whereas sham group by spurious ligation.Survivals were randomized into myocardial infarction(MI) group,simvastatin(Sim) group and sham group 24 hours after the operation.The Sim group was treated with simvastatin 40 mg?kg-1?d-1 via gavage till sacrifice.MI and sham groups were gavaged with equal volume of 0.9% NaCl at the same time.Rats were sacrificed at time points of 24 hours,7 days and 28 days after the operation,for the detection of HO-1 mRNA by RT-PCR,HO-1 protein level by Western bolt,activity of superoxide dismutase(SOD) and content of malondialdehyde(MDA) by spectrophotometric method in non-infarcted zone.Results Expression of HO-1 mRNA and protein level increased at 24 hours,peaked at 7 days and decreased to basal levels at 28 days.All the indexes at each time point mentioned above were significantly higher in MI group than those in sham group.At the 7th day and 28 th day after the operation,the indexes were higher in Sim group than those in MI group(P

The validity of (99m)Tc-YIGSR, a novel receptor radio-tracer, in imaging the Ehrlich ascites tumor was evaluated. YIGSR, a pentapeptide of laminin, was labeled with (99m)Tc by using a bifunctional chelator S-Acetly-NH(3)-MAG(3). The MIBI was labeled with (99m)Tc by following the kit instruction. The mice of tumor group were intravenously injected 1-2 mCi of (99m)Tc-YIGSR or (99m)Tc-MIBI via caudal vein, immobilized and imaged under a Gamma camera. The same procedure was performed in mice of blockade group, in which the unlabeled YIGSR was previously injected to block the receptor-recognition sites, and inflammation group serving as control. The reverse-phase Sep-Pak C(18) chromatogram was found to have an essentially complete conjugation between YIGSR and S-Acetly-NH(3)-MAG(3). The conjugated YIGSR could be radio-labeled successfully with (99m)Tc at room temperature and neutral pH, with a radio-labeling yield of 62%. Without the chelator S-Acetly-NH(3)-MAG(3), the YIGSR was labeled with (99m)Tc at an efficiency of 4%. The imagological study revealed obvious tumor accumulation of (99m)Tc-YIGSR 15 min after the injection, and the uptake peaked after 3 h with a tumor-to-muscle ratio (T/M) of 11.36. The radio-tracer was slowly cleared up and resulted in a T/M of 3.01 at the 8th h after the injection. As for blocked group, the tumor uptake of radiotracer was significantly lower, with the highest T/M being 4.61 after 3 h and 0.89 after 8 h. The T/M was 3.72 at the 3rd h and 1.29 at the 8th h after the (99m)Tc-YIGSR injection in the inflammatory group. The T/M was significantly higher in tumor group than in inflammatory group or control group (P<0.001). In the 99mTc-MIBI group, the T/M was 1.40 at the 3rd h and 0.55 at the 8th h after the injection, which showed a significant difference as compared with (99m)Tc-YIGSR (P<0.001). It is concluded that YIGSR can be successfully radiolabelled by using S-Acetly-NH(3)-MAG(3). (99m)Tc-YIGSR has many advantages in tumor imaging, such as quick and clear visualization, high sensitivity and specificity, and satisfactory target/non-target ratio (N/NT). It promises to be tumor radio-tracer.

OBJECTIVETo develop determination of dimethyl ether in workplace air by GC.

METHODSDimethyl ether were sampled with solvent desorption active carbon tube. The sampled carbon tube were desorbed by carbon tetrachloride, Elite-WAX column was used with temperature programmed from 40 to 90 degrees C, and the carrier gas programmed from 1.4 - 2.5 ml/min.

RESULTSThere was a good linearity (r = 0.9999) over the concentration of 0-63.91 microg/L, detection limit can reach 0.75 microg/ml.

CONCLUSIONThis method proved to be accurate and sensitive, to meet the determination of the dimethyl ether in workplace air.

Objective To observe the relationship between serum level of H-FABP before percutaneous coronary intervention (PCI) and prognosis in patients with ST-segment elevation myocardial infarction (STEMI). Methods A total of 104 cases were recruited in this study. PCI was performed 12 h after onset. The concentration of H-FABP was detected before operation. Gensini score system was used to calculate the integral of all patients after coronary angiography. According to Gensini score, patients were divided into three groups, 34 cases in group A (8≤Gensini scores<41), 34 cases in group B (41≤Gensini scores<64) and 36 cases in group C (Gensini scores≥64). Indicators were analyzed statistically including systolic blood pressure (SBP), diastolic blood pressure (DBP), smoking history, H-FABP, left ventricular ejection fraction (LVEF), total cholesterol (TC), triglycerides (TG), high density lipoprotein (HDL-C), low density lipoprotein (LDL-C), fasting blood sugar, diabetes (DM) and hypertension. The patients were followed up for 12 months after operation. According to the occurrence of major adverse cardiovascular events (MACE), patients were divided into MACE group and non-MACE group. The related factors of two groups were statistically analyzed. The multivariate Logistic regression analysis was used to screen the risk factors of MACE for patients with STEMI after emergency PCI. Results There is a gradually increasing tendency in total cholesterol levels among three A, B and C groups (P<0.05). LVEF values were lower in B group and C group than that of A group (P<0.05). There were no significant differences in levels of SBP, DBP, H-FABP, TG, HDL-C, LDL-C, fasting plasma glucose and smoking history, DM, and hypertension between three groups (P>0.05). In 12-month follow-up, the proportion of STEM combined with DM, the time from onset to PCI, the level of TC, the level of H-FABP before operation and Gensini score were significantly higher in MACE group than those of non-MACE group (P<0.05). Logistic regression analysis showed that STEM combined with DM, higher serum level of H-FABP before operation were risk factors of MACE in 12-month after operation (P<0.05). Conclusion For patients with acute STEM combined with diabetes and elevated preoperative serum level of H-FABP, be alert to the occurrence of MACE in 12-month after PCI.

Objective To investigate the efficacy and complications of therapeutic ERCP for patients over 80 years. Methods Data of 80 patients aging over 80 years, who tnderwent therapeutic ERCP from August 2007 to August 2009 were retrospectively studied. The etiology, complications and therapeutic effects were evaluated. Results A total of 51 patients (63. 8% ) were accompanied by cardiovascular diseases, and 43 patients were diagnosed as having malignant bile duct stricture (53. 8% ). The overail therapeutic efficacy was 96. 3%. The incidence of post-operative complications was 12. 5% (10/80). Occurrence of complications in patients with malignant tumors ( 18. 6% ) was significantly higher than that in patients with bile duct stones (5.6% ,P ＜0. 05). Incidence of complications in those with cardiopulmonary diseases was similar with that in patients without (11.8% vs. 13.8%,P＞0.05). Conclusion Age more than 80should not be considered as contraindication of ERCP. Therapeutic ERCP is effective in senior patients. Patients with advanced ages shows a relatively high incidence of complication after ERCP, which may not be related to other accompanying diseases.

The clinical prescriptions of hemoptysis that built by Menghe physician Ma Peizhi were collected, and were analyzed by the method of unsupervised data mining, such as association rules and clustering algorithm. From the prescriptions, we got the frequency of drugs, the association rules among drugs, 10 core drug combinations and 5 new prescriptions. Accordingly, after the analysis, Menghe physician Ma Peizhi had rich experience in the treatment of hemoptysis. His therapy for hemoptysis was clearing heat and moistening lung, dispel heat from blood to stop bleeding, and enriching yin and blood. And we can also make a conclusion that the TCM inheritance support system is of great practical value for minning the old doctors' clinical experiences.

Objective To investigate the drug combination of mutual restraint which is one of the application of seven different combination in drug in traditional Chinese medicine. Methods With the methods of association rules and complex system entropy clustering, we analyzed the drug combinations of mutual restraint that were collected from Compendium of Material Medica, to obtain the frequence of the single drug and drug combination of mutual restraint and the association rules between them. Results The single drugs with higher frequency are Rhizoma Coptidis, Fructus Crotonis, Rhei Radix Et Rhizoma, Azurite, Rhizoma Acori Tatarinowii, etc. The drug combinations with higher frequency are Azurite and Artemisia capillaries, Fructus Crotonis and Rhizoma Coptidis, Fructus Crotonis and Soybean etc. The associative drug combinations which confidence is 1 include “Indigo juice → Fructus Crotonis”,“Indigo juice → Rhizoma Coptidis”, etc. Conclusion The drug combinations of mutual restraint mainly include mineral drugs, antipyretics, purgatives, and toxic drugs.
