1.Studies on chemical constituents of Lethariella cladonioides

Object To develop and utilize a folk-medicine of Yunnan Province, Lethariella cladonioides (Nyl.) Krog. Methods The constituents were extracted with 80% ethanol and isolated with silica gel column chromatography. The structures were identified by physicochemical properties and spectral analysis. Results Two unknown constituents were elucidated from the extract as 3-aldehyde-6-methyl-2, 4-dihydroxy-ethyl-benzoate (Ⅰ), and 4-methyl-2, 6-dihydroxy-benzaldehyde (Ⅱ). Conclusion They are both new constituents obtained firstly.
2.Determination of the Content of Total Flavonoid in Fufang Psoralen Corylifolia Sustained-release Tablets by Ultraviolet Spectrophotometry

OBJECTIVE:To establish a method for the determination of the content of total Flavonoid in Fructus Psoraleae by UV spectrophotometry.METHODS:The determination was performed using Bavachin standard as control substance,and sodium hydroxide as color-developing agent,with the wavelength set at 440nm.RESULTS:The absorbency and concentration of Bavachin have good linearity in the range of 4.10~ 12.30? g? mL-1(r=0.999 8).The average recovery rate was 100.5%(RSD=1.57%,n=9).CONCLUSION:The method is easy to operate,highly accurate,reproducible and can be applied to determine the content of total Flavonoid in Fufang Psoralen corylifolia sustained-release tablets.
3.Effect of the pathological changes of the pelvic cavity and fallopian tube on the outcome of in vitro fertilization and embryo transfer

Objective To evaluate the effect of the pathological changes of the pelvic cavity and fallopian tube on the outcome of in vitro fertilization and embryo transfer(IVF-ET).Method One thousand and thirty-two patients who underwent IVF-ET were divided into tubal and pelvic infertile group(605 cases)and non-tubal and pelvic infertile group(427 cases).The tubal and pelvic infertile group was also divided into salpingemphraxis group(243 cases),tubal resection group(104 cases),fallostomy group(149 cases),tubal dropsy group(109 cages)according to the tubal lesion regions,and combined with pelvic group(194 cases),combined without pelvic group(411 cases).The data of clinical pregnancy,ectopic pregnancy,and abortion was analyzed respectively.Results The ectopic pregnancy and abortion rates in tubal and pelvic infertile group[10.63%(27/254)and 9.06%(23/254)]were higher than those in non-tubal and pelvic infertile group [3.27%(5/153)and 4.58%(7/153)](P<0.01 or<0.05).The ectopic pregnancy rate was the lowest in tubal resection group[2.17%(1/46)],the highest in fallostomy group[22.41%(13/58)],there was significant difference among the groups(P<0.01).The abortion rate in fallostomy group and tubal dropsy group[10.34%(6/58)and 15.00%(6/40)]was higher than that in salpingemphraxis group and tubal resection group [7.27%(8/110)and 6.52%(3/46)],there was significant difference among the groups(P<0.05).The abortion rate in combined with pelvic group[11.54%(9/78)]was higher than that in combined without pelvic group[7.95%(14/176)](P<0.05).Conclusions The pathological changes of the pelvic cavity and fallopian tube are higher risk factors of ectopic pregnancy and abortion occurrence.The assessment and treatment of pelvic cavity and fallopian tube before assisted reproductive treatment cycles should be enhanced.
4.Impact of diabetes dietary knowledge on hospital readmission rate among patients with type 2 diabetes mellitus

Objective To explore the effect of dietary knowledge of diabetes on the readmission rate on patients with type 2 diabetes mellitus. Methods Totally 334 patients of type 2 diabetes from Suzhou finished the general information and the questionnaires of knowledge of diabetes diet. The impacts of knowledge of diabetes dietary on their readmission rates were also analyzed. Results Both whether having hypoglycemia or not and the score of diabetes dietary knowledge were associated with readmission rate within half a year. Those having low blood sugar (B=1.276, P<0.01) and higher score of diabetes dietary knowledge (B = 0.049, P = 0.006) had higher readmission rate. Further analysis showed that patients with low blood sugar getting higher score of diabetes dietary knowledge had higher hospital admission rate (P = 0.01). Conclusions The readmission rate of type 2 diabetes mellitus patients was impacted by hypoglycemia occurrence and the familiarity degree of diabetes diet knowledge. So it′s important to further explore the causes and make the reasonable intervention to reduce the readmission rate.
5.Clinical analysis of fundus fluorescein angiography in Takayasu arteritis

Background Takayasu arteritis is a non specificity inflammation of aorta and its branch.The incidence of Takayasu arteritis is low and its ocular secondary disease is rare.The correct diagnosis of Takayasu arteritis is very important for its early treatment in clinic.Objective This study is to analyze the fundus findings and characteristic of fundus fluorescein angiography (FFA) of Takayasu retinopathy.Methods The FFA and clinical data of 12 patients (24 eyes) with Takayasu arteritis were retrospectively reviewed.Written informed consent was obtained from all the patients before and initiation of any study protocol.Results In 12 patients,chronic ischemic retinopathy was found in 15 eyes of 9 patients.The arm to retina circulation time(A RCT) prolonged to (19.20±2.95) s in 5 eyes,and the retinal circulation time (RCT) delayed to 10.62±6.15 s in 5 eyes.Peripapillary arteriovenous anastomosis was found in 2 eyes of 2 patients.Macular arch ring was incomplete in 6 eyes of 4 patients.Eight patients (14 eyes) had telangiectasis and microaneurysm,and 2 eyes of 2 patients presented neovascularization on the disc or elsewhere.In 12 patients,hypertensive retinopathy was found in 4 eyes of 3 patients,showing narrow retinal artery,arteriosclerosis,hemorrhage,cotton wool spots and hard exudates.Ten patients were diagnosed as Takayasu arteritis before FFA examination,and 2 patients were determinedly diagnosed after FFA was performed.Conclusion The main features of Takayasu retinopathy are hypertensive retinopathy and chronic ischemic retinopathy.It is important for ophthalmologist to correctly recognize the clinical features of TA.
6.Anti-tumor Effects of Total Saponins Extract from the Aerial and Underground Parts ofParispolyphyllavar.yunnanensis

Objective To investigate the anti-tumor effects of total saponins,extracted from stems and leaves ofParispolyphyllavar.yunnanensis. Methods Taking mouse stomach carcinoma MFC cell line,human mammary cancer cell line(MCF-7)and human cervical carcinoma cell line(Hela),and their tumor-bearing mice as models,the antitumor activity was carried out in vitro and in vivo. CCK-8 assay was used to determine the inhibitory effect in vitro. The tumor-bearing mice model was induced by tumor cell vaccination in normal mouse forelimb left axillary subcutaneous and these mice were randomized into NS group,cytoxan group(30 mg/kg),saponins high-dose,medium-dose and low-dose groups(60,30 and 15 mg/kg). They were given intraperitoneal injection once a day for consecutive 15 days. The tumor inhibition rate and survival of the tumor-bearing mice were measured. Results Saponins,extracted from both aboveground and underground ofP.polyphyllavar.yunnanensis could significantly inhibit the growth of MFC,MCF-7 and Hela cells in time- and dose-dependent manners. The tumor weight in each drug treated group was significantly lower than that in the control group. Conclusion Saponins,extracted from both aboveground and underground ofP.polyphyllavar.yunnanensis have antitumor activity.
7.Association between Circulating Betatrophin Levels and Metabolic Parameters in Non-alcoholic Fatty Liver Disease Patients

Objective To investigate circulating betatrophin levels in non-alcoholic fatty liver disease patients (NAFLD). To explore its association with various metabolic parameters in Chinese subjects. Methods From August 2014 to April 2015 , 58 patients with NAFLD and 41 non-NAFLD control subjects with age and sex matched were enrolled in the Department of Endocrinology of Zhujiang Hospital. Results In NAFLD group, serum betatrophin concentrations significantly increased (781.96 vs. 431.89 pg/mL, P < 0.05) and associated only with HDL-cholesterol level. In normal group, betetrophin levels are associated with multiple metabolic parameters, such as fasting glucose, insulin, C peptide, HOMA-IR, QUICK index and HDL-C levels. Conclusions Serum betatrophin concentration significantly elevated in non-alcoholic fatty liver disease patients and was negatively correlated with HDL-C level (r = -0.479, P < 0.001).

9.Analysis of Chemical Constituents of the Volatile Oil from Stems and Leaves of Sarcmdra glabra by GC-MS

OBJECTIVE: To analyze the chemical constituents of the volatile oil from the stem and leaves of Sarcandra glabra.METHODS: The volatile oil was extracted from the stem and leaves of Sarcandra glabra by steam distillation.The components of the volatile oil were separated and identified by GC-MS.The relative content of each component was determined by area normalization.RESULTS: 29 components in the volatile oil from the stem of Sarcandra glabra were identified,and their content representing 75.88% of the total volatile oil.64 components in the volatile oil from the leaves of Sarcandra glabra were identified,with their content representing 87.98% of the total volatile oil.CONCLUSION: The volatile from leaves and that from stems of Sarcandra glabra varied in the content of low molecular weight of chemical constituents,almost no low molecular weight of chemical constituent contained in the volatile oil in the stems.These data provide scientific bases for exploitation of Sarcandra glabra.
10.Effect of intermittent high glucose on INS-1 cell proliferation and the expression of skp2-p27

Objective To investigate the effect of intermittent high glucose on proliferation, apoptosis, and cell cycle progression of INS-1 cells, and the possible intracellular pathways activated by intermittent high glucose. Methods Cell viability was evaluated by cell counting kit, the cell cycle was determined by flow cytometry,Annexin-V/PI double-labeled cell apoptosis detection kit was used to monitor cell apoptosis. Cell cycle related protein Skp2 and p27 expressions were detected by Western blot. Results ( 1 ) Both intermittent and constant high glucose significantly inhibited the growth of INS-1 cells, and the former effect was more significant. ( 2 ) Intermittent and constant high glucose levels significantly increased apoptosis in INS-1 cells, and the former effect was more significant. (3) Intermittent and constant high glucose levels significantly inhibited the cell process, the G0/G1 cell cycle arrest also was induced by intermittent high glucose, resulting in lowered proportion of the G2/M phase and S phase of INS-1 cells. (4) Intermittent and constant high glucose significantly decreased the level of protein Skp2 and increased the level of cell cycle related protein p27. Conclusion Intermittent high glucose levels affect INS-1 cell growth and proliferation, as well as induce cell apoptosis, probably by decreasing the level of protein Skp2 and increasing the level of p27 in the cells, resulting in arrest of progression through the G1 phase to the S phase of INS1 cells, and thus impairment of cell proliferation.
