Poly(U)-binding-splicing factor 60 KDa (PUF60), a U2-related splicing factor that facilitates 3′splice-site recognition at the early stage of spliceosome assembly.High expression of PUF60 is related to the occurrence of multiple tumors, such as colorectal cancer, ovarian cancer, gastric cancer, liver cancer, et al.PUF60 can regulate the expression of c-myc through the core-human transcription factor Ⅱ basal transcription factor.Anti-PUF60 antibodies are detected in the sera of patients with early-stage and recurrent tumor, and the levels are significantly decreased after the operation.PUF60 can be used as a combined or independent test index for the diagnosis of cancer, which can be a potential new target for gene therapy.

OBJECTIVE:To establish the HPLC method for the content determination of paeoniflorin and liquoritin in Jiawei xiaoyao pills. METHODS:It was performed on Hypersil-ODS C18 with the mobile phase of acetonitrile-water(14∶86,V/V)at the flow rate of 1.0 ml/min,the detection wavelength was 230 nm,the column temperature was room temperature and the volume was 10 μl. RESULTS:The linear range was 16.12-80.6 μg/ml for paeoniflorin(r=0.999 6)and 6.08-30.4 μg/ml for liquoritin(r=0.999 8);the RSDs of precision,repeatability and stability tests were all less than 2.0%;the average recovery was respectively 98.51%(RSD=1.94%,n=6)and 98.08%(RSD=1.29%,n=6). CONCLUSIONS:The method is simple,accurate and reliable, and can be used for the content determination of paeoniflorin and liquoritin in Jiawei xiaoyao pills.

The presence and persistence of systemic and lung inflammation in children with acute respiratory distress syndrome (ARDS) is the basis for the use of corticosteroids as a therapeutic agent.The trial of ARDS treated with high-dose short-course corticosteroids showed no benefit,even increase motality.At present,the results of randomized controlled trial and meta-analysis suggested that low-dose and replacement-dose methylprednisolone [1-2 mg/(kg· d)] or equivalent types of corticosteroids may decrease the fatality rate,reduce staying days in ICU and the duration of mechanical ventilation.Use of corticosteroids for ARDS in children is still lack of multicenter randomized controlled trial.

Objective To establish a method of preparing metoprolol succinate sustained-release tablet and its content determination. Methods The formulation was optimized through the orthogonal design test by using release rate of the drug as an indicator.The different batches of metoprolol succinate sustained-release tablets were determined by HPLC. Results The tablets could release drug steadily and slowly as designed,which was similar to imported tablets. The linear range of metoprolol succinate was 10-70 μg?mL-1( r=0.999 8) . Conclusion The releasing rate of metoprolol succinate sustained-release tablet prepared in optimum condition can meet the requirement. This preparation technology is simple, the assay method is rapid, sensitive and reproducible.

Objective:To provide some suggestions for improving the pharmaceutical advertisement supervision system and standardizing the pharmaceutical advertisement market. Methods: Literature reviews, statistical analysis and interviews were adopted to study the status quo and difficulties of pharmaceutical advertisement supervision and pharmaceutical advertisement trends. Results:The top-level design of pharmaceutical advertisements supervision sys-tem has two defects:the separation between approval and penalty and the obstacles to cooperation at all levels of the drug administration. The worsening status quo of illegal drug advertising has elicited concern from scholars and regu-lators. However, there are still some difficulties in the supervision of drug advertisers, advertising agents, and pub-lishers. Conclusion:It is suggested that the drug advertising regulation system should be perfected by strengthening information supervision, establishing a coordination mechanism and consolidating self-discipline consciousness. Addi-tionally, laws and regulations should be promptly revised as an essential way to improve the effects of supervision.

Objective The purpose of the study was to clarify the reasons that may explain the families' non-disclosure of the truthful information to cancer patients in our medical background.Methods The study included 82 family members of cancer patients who was not told truthful diagnosis by convenience sampling.Data were analyzed by Bernard Berelson's content analysis.Results 13 categories and 7 themes were formulated,including avoiding,concerns,judgment,buffer,cognition,impact and endurance.The agreement rates were 75.2％ and 78.7％.Conclusions The families' non-disclosure of truthful information to cancer patients was a kind of avoiding behavior,the judgment of families impacted their attitude towards telling or not.

Objective To study the expressions and correlation of nuclear factor kappa B ( NF-B) p65 and trefoil factor family-3(TFF3) in gastric cancer,in order to evaluate the roles of NF-Bp65 and TFF3 in the occurrence and development of gastric cancer,and their correlation with Helicobacter pylori (Hp) infection.Methods The expressions of NF-Bp65 and TFF3 were detected by immunohistochemistey in 30 normal gastric mucosa specimens ,32 dysplasia specimens and 70 gastric cancer specimens .At the same time,the infection of Hp was detected .Results In normal gastric mucosa ,dysplasia and gastric cancer ,the expression of NF-Bp65 had a increasing tendency ( compared with the normal gastric mucosa group ,χ2 =18.632,44.291,all P<0.01).The expression of NF-Bp65 in gastric cancer group was higher than that in dysplasia group (χ2 =5.205,P<0.05).The expression of TFF3 in three groups also had a increasing tendency (compared with the normal gastric mucosa group ,χ2 =16.944,22.917,all P <0.01).The expression of TFF3 in gastric cancer group was higher than dysplasia group ,but there was no significant difference between the two groups (χ2 =0.162,P>0.05).In dysplasia group,the expressions of TFF3 and NF-κBp65 in Hp positive patients were significantly higher than those in Hp negative patients (P<0.01 or P<0.05).There was significantly positive correlation between expression of NF-Bp65 and TFF3 in gastric cancer group ( r=0.350,P=0.003).Conclusion High expressions of NF-Bp65 and TFF3 may be involved in an early event of gastric cancer . NF-Bp65 expression is positively correlated with the abnormal expression of TFF 3,both of which may be involved in the development and progression of gastric cancer .

Objective To investigate the nutritional status and glycemic control level in elderly patients with type 2 diabetes in urban community of Yangpu district in Shanghai.Methods The 24-hour dietary recall method for 3 consecutive days was used to investigate the dietary nutrients intake in 400 patients with type 2 diabetes.Results The ratio of patients with blood glucose level less than 6.1mmol/L was 23.3％ (93/400).The energy intake in patients with type 2 diabetes was low,and only reached 62.81％ of the recommended standard of energy intake for adults.The energy ratio of three diet nutrients (protein,fat and carbohydrate) were 14.41％,33.93％,51.66％ respectively,which showed that the energy ratio of fat was relatively high.The energy ratio of breakfast,lunch and supper dinner were 23.37％,38.05％ and 38.57％ respectively.The average intake of vitamin E,iron,niacin,manganese,phosphorus and cholesterol in elderly patients was more than the recommended standard of adult diet nutrients,and reached 149.6％,138.1％,137.1％,132.5％,127.5％ and 122.0％ of the recommended standard respectively.But the average intakes of protein,fat,carbohydrate,dietary fiber,calcium,zinc,copper,magnesium,potassium,vitamin A,vitamin B1,vitamin B2 and vitamin C in elderly patients were lower than the recommended standard,and reached 77.9％,90.3％,57.1％,50.7％,49.3％,81.4％,75.7％,75.7％,77.5％,77.0％,55.1％,58.1％ and 67.6％ of the recommended standard respectively.Conclusions The status of dietary nutrients intake in elderly patients with type 2 diabetes in Yangpu district of Shanghai is unreasonable,the intakes of dietary fiber,calcium,vitamin B1,vitamin B2 etc.were insufficient,and reasonable diet instruction should be provided for the elderly patients.

Objective:Comparative analysis of imaging features of abdominal CT, B ultrasound and magnetic resonance cholangiopancreatography in three different methods for detection of extrahepatic bile duct stones. Methods: A random sample of extrahepatic bile duct stones were a sample of 150 cases as the object of study, patients in the endoscopic retrograde cholangiopancreatography after diagnosis, followed by B ultrasound examination, the implementation of CT scanning and MRI imaging in 142 cases, including 78 cases of B ultrasound detection, CT detection, 118 cases underwent MRI examination. Results: The detection results were recorded and compared the 3 imaging methods. Results: The positive rate of B ultrasound 39.4%, the positive rate of CT was 53.8%, MRI positive rate was 86.4%, the positive rate of MRI was significantly higher than that of B ultrasound and CT (x2=59.725, x2=25.595; P<0.01), the detection rate of CT was significantly higher than that of B ultrasound(x2=4.231, P<0.05). Diameter less than 8 mm of extrahepatic bile duct stones with B ultrasound and positive rate of 25%, the positive rate of CT was 33.3%, MRI positive rate was 60%, the positive rate of MRI was significantly higher than that of B ultrasound and CT(x2=7.789, x2=7.789;P<0.01), the detection rate of CT with no significant differences between the B ultrasound. Conclusion:MRI of extrahepatic bile duct stones with B ultrasound and the positive rate of CT is higher, but the clinical needs according to the comprehensive evaluation of individual patients, economic status, symptoms, diagnosis method for selecting suitable.
