1.Process Optimization of PEGylating Fused Protein of LL-37 and Interferon-α2a.

PEGylating is an effective way for prolonging the half-time period and decreasing the immunogenicity of protein drugs. With experiments of single factor, it was proved that the optimal processes for PEGylating the fused protein of LL-37 and interferon (IFN)-α2a were: PEG molecular weight was 5,000, fused protein concentration was 0.6 mg/mL, the mole ratio of protein to mPEG₅₀₀₀-SS was 1:10, the reaction temperature was 4 °C, and the pH was 9.0, respectively. With orthogonal experiments, we proved that the influential order of 3 main factors is: the fused protein concentration > the mole ratio of protein and mPEG₅₀₀₀-SS > pH and the optimal conditions were the fused protein concentration as 0.6 mg/mL, the mole ratio of protein and mPEG₅₀₀₀-SS as 1:10, pH as 8.8. Under these optimal conditions, the average rate of PEGylated protein with 3 times parallel experiments was 86.98%. After PEGylated, the interferon activity and antimicrobial activity of fused protein could be remained higher than 58% and 97%, respectively.

2.Analysis on Patients’Compliance and Its Influential Factors

OBJECTIVE:To study the measures to improve the patients’compliance.METHODS:An investigation was carried out in136patients in a given hospital in Wuhan in the way of questionnaire and interview.RESULTS:There was no significant difference in compliance among the patients with different sex,age,education background and income,but there we_ re significant differences among the patients with different initiatives,conceptions,habits,and knowledge of drug use.CONCL_ USION:It is suggested that IMB model should be adopted to improve patients’compliance in the respect of external informa-tion,internal motivation,and behavioral skills.
3.Next-generation sequencing in study of hepatitis B virus variation

The high mutation rate during hepatitis B virus (HBV)replication leads to HBV quasispecies.The study of HBV quasispecies has an important significance for the hepatitis B pathogenesis,prognosis,and outcome prediction.Recently,the next-generation sequen-cing (NGS)is extensively used in many biological and medical fields due to its high throughput,ultra-deep coverage,and high sensitivity, which also brings new strategies to HBV quasispecies studies.This article describes the working principles and features of several commonly used NGS technologies,reviews the application of NGS technologies in HBV quasispecies detection in recent years,and particularly discus-ses the variations in different regions of HBV genome and the population characteristics of HBV quasispecies.For now,NGS technologies used in HBV variation detection mainly presents the advantages of high sensitivity and high throughput.However,how to lower the cost,in-crease the accuracy of sequencing,and standardize the procedure of massive sequencing data management and personalized analysis are still challenging problems.The issue about NGS that matters at present and urgently needs to be solved is how to overcome its limitations,and then put it into HBV-related studies and ultimately clinical application.
4.Research advances in cellular immunotherapy for primary hepatocellular carcinoma

The present therapy for primary hepatocellular carcinoma (HCC)consists of surgery as well as local radiotherapy and chemother-apy.However,the majority of patients are susceptible to recurrence after comprehensive treatment,and the overall treatment outcome is not ideal due to the lack of effective drugs and strategies.Increasing evidence has demonstrated that the immune system is closely related to the development,progression,metastasis,and recurrence of HCC.Thus,immune therapy,especially cellular immunotherapy,could regulate immune function and induce specific antitumor immunity to achieve the goal of controlling HCC and reducing its recurrence and metastasis, which has become an essential part in the comprehensive treatment of HCC.The findings in preclinical and clinical studies on cellular immu-notherapy for HCC data are reviewed,and the current problems are discussed.
5.Myostatin Gene Expression Level in the Soleus Muscle of Tail-Suspended Rats

Objective To study the effect of tail-suspending on myostatin(MSTN)expression in the soleus muscle of rats.Methods The MSTN mRNA levels in soleus muscle of tail-suspended and normal control rats were measured and compared.Results The MSTN mRNA in soleus muscle of tail-suspended rats was 2.2 times(hind limb unloading for 14 days)and 3.5 times(hind limb unloading for 30 days) higher than that in normal control rats (P<0.05).The relative wet weight of the soleus muscle in tail-suspended rats was decreased by 11%(unloading 14 days)and 1 9%(unloading 30 days),respectively,compared with that in normal control rats.Conclusion The expression of MSTN mRNA is elevated in the soleus muscle of tail-suspended rats,probably due to atrophy of the soleus muscle.

claudin protein is the major component of tight junctions complex between cells,which plays an important role in cells junction and adhesion. Its specific expression in different tissues directly affects the biological behavior of tumor cells. In pancreatic tumors, the claudin's expression provides an important refer-ence for the diagnosis and treatment of tumors. In addition, claudin are involved in the proliferation and ap-optosis of tumor cells. In the treatment of tumors, claudin-mediated targeting therapy has great prospects forthe same.
7.The relationship among big five-personality, resilience and general health of new recruits

Objective To discuss the relationship among Big Five-Personality, Resilience and General Health of the new recruits in a certain troop. Methods 205 new recruits completed the questionnaires, which include NEO Five-Factor Inventory ( NEO-FFI) , Connor-Davidso Resilience Meter (Chinese version) , and General Health Questionnaire (GHQ-12). Results The neuroticism trait of personality positively correlated to General Health( r = 0.50, P<0.001) and significantly predicted mental disease (β =0.4, P<0.01). The self-strengthening dimension of resilience was negatively related to general health ( r = - 0. 51 , P<0. 01 ). Furthermore, self-strengthening played partial mediating role between neuroticism and general health ( Sobel Z =2. 92, P<0. 01). Conclusion The neuroticism trait of personality is a risk factor for new recruits'mental health, while self-strengthening is a protective factor for it.

10.Association study of circulating endothelial cells and clinical outcome of pulmonary hypertension secondary to congenital heart disease

Objective To compare the count of circulating endothelial cells (CECs) between patients with reversible pulmonary hypertension (RPH) and irreversible pulmonary hypertension (iRPH) by flow cytometry in order to find a new biomarker to distinguish RPH from iRPH.Methods Ninety pulmonary hypertension associated with congenital heart disease patients treated in our hospital between September 1,2013 to March 31,2014 were enrolled in this study.According to the mean pulmonary arterial pressure (mPAP) measured by echocardiography in six months after treatment,the patients were separated into 2 groups:iRPH group(mPAP≥50 mmHg),RPH group(mPAP <50 mmHg).Results Totally the clinical data of 87 patients were enrolled in the statistic analysis,86 patients finished the follow-up in 6 months after the surgery,1 patient died from pulmonary hypertensive crisis.The count of CECs was not correlated with the extent of pulmonary hypertension(P =0.925).The number of CECs was higher in iRPH group than that of RPH group(0.46‰ vs.0.09‰,Z =-5.021,P =0.000).And also the age of patients in iRPH group was elder than that of patients in RPH group [(43.1 ± 37.4) months vs.(9.3 ± 12.6) months,t =-5.079,P =0.000].Conclusion The count of CECs is significantly increased in iRPH patients.It could be used as one biomarker to distinguish RPH from iRPH.
