1.The clinical efficacy of Fuzheng Huayu Capsule in liver cirrhosis decompensated and the impact on quality of life

Objective To investigate the clinical efficacy of Fuzheng Huayu Capsule in the treatment of liver cirrhosis (decompensated) and its impact on quality of life in patients with TCM therapy research.Methods 160 cases of liver cirrhosis (decompensated) patients were chosen as research subjects.They were divided into experimental groupand control group according to random number table.Both groups used routine liver antiviral therapy,the experimental group was given Fuzhenghuayu tablets therapy,the control group only received conventional therapy.After 6 months of treatment,and treatment for the two groups of patients were evaluated quality of life conditions.Results Compared with before treatment,HA,LN,PC Ⅲ,Ⅳ-C of the indicators were significantly decreased,and the difference was statistically significant (t =10.89,15.88,10.42,12.38,4.41,9.27,4.97,6.55,all P < 0.05).After the two groups of patients for the treatment of fibrotic indicators analysis found more significant decline in the observation group,the difference was statistically significant (t =5.32,3.88,5.25,5.01,all P < 0.05).2 Compared with before treatment,ALT,AST levels of the two groups were significantly decreased,the difference had statistically significant (t =2.02,17.87,2.62,7.96,all P <0.05).for two groups of patients was compared with that total efficiency (87.50% vs 66.25 %),the difference was statistically significant (x2 =10.16,P < 0.05).Compared with pre-treatment,two patients in the field of physiology,psychology field,the field of social relations,environmental areas and total score was significantly higher,the difference was statistically significant (t =13.94,12.37,11.12,10.71,16.45,8.60,6.54,8.35,4.86,8.04,all P < 0.05) ; The two groups of patients after treatment compare the scores and scores found in the observation group was significantly more elevated,compared with the control group,the difference was statistically significant (t =4.97,3.81,4.98,2.56,5.05,all P < 0.05).Conclusion In the treatment of hepatitis cirrhosis,Fuzheng Huayu Capsule can significantly improve the degree of liver fibrosis,improve the quality of life of patients and improve clinical outcomes.
2.Preparation and efficacy evaluation of polyclonal antibodies against serum albumin of five species

Objective To prepare polyclonal antibodies against the serum albumin of human,cattle,sheep,pig and horse,and evaluate their efficacy in the identification of human serum albumin(HSA). Methods The specific polypeptides of human,cattle,sheep,pig and horse serum albumin were prepared by bioinformatics and polypeptide synthesis method,which were coupled with keyhole limpet hemocyanin(KLH)to prepare the peptide antigen after the purity was identified by high performance liquid chromatography(HPLC). Male New Zealand white rabbits were immunized with polypeptide antigens of five species subcutaneously,with 2 for each kind of antigen. The antiserum was then obtained and purified by Protein A affinity chromatography to prepare the polyclonal antibody. The titers and specificity of the polyclonal antibodies were determined by ELISA and Western blot respectively,and the prepared five species of serum albumin were used to identify HSA products. Results The synthetic peptides of human,cattle,sheep,pig and horse serum albumin had a purity of over 91%,and the corresponding polyclonal antibodies all had the titer of 1∶160 000,which showed specific binding with the corresponding antigens and effectively identified the HSA products. Conclusion The polyclonal antibodies of human cattle,sheep,pig and horse serum albumin prepared in this study have good specificity and the preparation process is simple and rapid,suitable for the mass production,which lays a foundation of the development of HSA rapid identification kit.
3.Clinical Analysis on 38 Senile Cases of Cardiac Carcinoma with Total Gastrectomy

Objective To explore the operative methods for senile patients with cardiac carcinoma.Method 38 senile patients with cardiac carcinoma in developing stage and administered total gastrectomy were analyzed and 1~5 years follow-up were given.Results No operation induced death and no anastomosis fistular occured and the 1,3,and 5-year survival rates were 73.68%,52.63%,and 22.86% respectively.Conclusion Laparotomy might be the optimal treatment to the senile patients with cardiac carcinoma.Reconstruction of digestive tract might be suitable for most of the senile patients with developing cardiac carcinoma.Total gastrectomy may diminish the occurrence of complications and may give a better long-term survival rate.
4.Expression change of nitric oxide synthase in major organs during acute liver failure in rats

Objective:To observe the expression changes of nitric oxide synthase(NOS) in the liver,the lung,the kidney and the intestine of rat during acute liver failure(ALF).Methods:Wistar male rats(n=60) were randomly divided into 6 groups:SO group,ALF 6 h group,ALF 12 h group,L-Arg (300 mg/kg) group,L-NAME (30 mg/kg) group,L-Arg+L-NAME group, each group had 10 rats.The expression of eNOS mRNA and iNOS mRNA in the liver,lung and kidney was determined with in situ hybridization,and in the small and large intestines was determined with immunohistochemical method.Results:The expression of eNOS mRNA in the lung and iNOS mRNA in the liver,lung and kidney increased significantly 6 h after operation,but the expression of eNOS mRNA and iNOS mRNA in the liver,lung and kidney reduced 12 h after operation(P
5.A case of primary cutaneous anaplastic large cell lymphoma at the vaccination site

An unusual case of primary cutaneous anaplastic large cell lymphoma induced by influenza vaccination is reported.A 79-year-old man developed an erythematous nodule at the injection site on the upper left arm 2 months after vaccination against influenza A virus,which gradually grew and ulcerated.Pathological examination of the lesion revealed epidermal infiltrate of lymphocytes and numerous clumps of densely distributed lymphocytes with lightly stained cytoplasm,nuclear atypia and karyokinesis.Immunohistochemistry showed that the tumor cells stained positive for CD3 (90%),CD5 (90%),LCA (90%),CD30 (90%),CD10 (a few),Ki-67 (80%),but negative for CD20,CD56,cytokeratin 1-3,epithelial membrane antigen,synaptophysin,and anaplastic lymphoma kinase.The patient was diagnosed with primary cutaneous anaplastic large cell lymphoma at the injection site following vaccination against influenza A virus.
6.IDENTIFICATION OF DEATH CAUSED BY SNAKE BITING BY ABC-IMMUNOCYTOCHEMICAL TECHNIQUE

Varying doses of cobra crude venom were injected subcutaneously into rats.Formaking frozen and methylaldehyde fixed,paraffin embeded sections,tissues blockswere taken from the injected site muscles,hearts,livers,spleens,lungs,kidneysand diaphragms either immediately,or 6,12,24hrs after death.Tissue sectionswere stained with and ABC-immunocytochemical techniques.On HE stained sections,edema,heamorrhage,congestion were observed.Coludyswelling,necrosis,and breakdown of cells were seen in most of the organs.Nospecific morphological changes has been found.On the frozen sections of the musc-les taken from the injected site,kidneys and livers,the venom has been identifiedby ABC-immunocytochemical staining.Although as little as 2LD_(50) (2mg/kg) venomwas given,it could be demonstrated on the membrane of the muscle even 24 hrs afterdeath.In case of large amount of venom (11LD_(50) ) poisoning,the venom was alsofound in nucleus of liver cells and in the endothelial cells of capillaries of renalglomeruli.The pathogenesie of localization of snake venom in endothelial cells ofrenal glomerular capillaries was discussed,there is No report about the localizationof snake venom in nucleus of liver cells It is a very interesting phenomenonIt is usggested that the ABC-immunocytochemical stain is a sensitive and specificmethodforid entifying sanke venom in tissues of snake biting victims.
7.Study of the protective effects of hypertonic saline against ischemia-reperfusion injury of the liver

Objective: To investigate the protective effects and the mechanism of the preconditioning with hypertonic saline against warm hepatic ischemia-reperfusion (I-R) injury. Methods: Sixty patients who underwent partial liver resection were randomly assigned to 3 groups (n =20): group 1 without Pringle maneuver(NPR); group 2 with Pringle maneuver (PR), and group 3 with 7.5% hypertonic saline 4ml/kg preconditioning (HS) 5 minutes before Pringle maneuver for resection and in group 2 , 4ml/kg normal saline were given. Blood samples were obtained at 3, 15, and 30 minutes before and after liver resection, and 2, 24, and 48 hours following resection of the liver for polymorphonuclear leukocytes (PMN) count and serum alanine transminase levels test. In addition, small pieces of liver tissue were cut from remaining liver before resection and 30 minutes after resection to determine MPO concentrations. Results: Following partial liver resection via NPR, no changes in the concentration of circulating PMN and liver MPO concentrations were observed. But in the PR and HS group PMN counts significantly decreased and liver MPO concentrations significantly increased. Serum ALT levels increased in all groups significantly 24 and 48 hours after resection. All of these changes were most pronounced in the PR group and significantly reduced by hypertonic saline. Conclusion: Preconditioning with 7.5% hypertonic saline can protect liver from warm ischemia reperfusion injury and the possible mechanism was the inhibition of PMN accumulation in the liver.
8.Determination of augenol of Jingbutong Babugao by gas chromatography

Objective:To establish a GC method for the determination of eugenol in Jingbutong Babugao. Methods: The determination was carried out on a PEG 20M capillary column with temperature at 160 ?C , FID detector with temperature at 230 ?C . Results: The eugenol sample showed a good linear relationship at the range of 0.38~6.14?g and correlation coefficient was 0.9993. The average recovery was 98.60% with a RSD of 2.18%. Conclusion: The method is simple, accurate, reproducible and can be used for determination of eugenol in Jingbutong Babugao.

Conclusions Pancreatoduodenectomy for the treatment of periampullary carcinomas is with high risks. Postoperative mortality and morbidity were in close correlation with the risk factors.
10.Content Determination of Ursolic Acid in Shanzhajing Jiangzhi Tablets by HPLC

Objective To determine ursolic acid in Shanzhajing Jiangzhi Tablets by HPLC method. Method Chromatographic column was Kromasil C18 (4.6 mm?200 mm, 5 ?m). The Mobile phase was methanol- 0.15% phosphoric acid (88∶12), and the flow rate was 1.0 mL/min. Detection wavelength was 215 nm. Column temperature was room temperature. Results Ursolic acid quantity and its peak area showed good linearity relationship in the range of 0.253~5.052 ?g. The correlation coefficient (r ) was 0.9998 (n=6). The average recovery was 100.37% and RSD was 1.88%. Conclusion The method is easy and accurate, with good sensitivity and repeatability. It is a good method to control the quality of Shanzhajing Jiangzhi Tablets.
