Chinese Journal of Immunology 2015;(7):912-916
Objective:To investigate the inhibitory effect and its possible molecular mechanisms of MicroRNA-34a(miR-34a) on the human nasopharyngeal carcinoma CNE-2 cell line subcutaneous xenograft tumor in nude mice. Methods: The human nasopharyngeal carcinoma CNE-2 cell line was cultured in vitro.miR-34a and Scrambled miRNA recombinant plasmids were successfully established and stably transfected into CNE-2 cells.Fifteen six-week-old male nude mice were divided randomly into three groups:miR-34a group(5 mice) ,Scrambled miRNA group(5 mice) ,Blank control group(5 mice).Different CNE-2 cells were subcuta-neously injected on the back near right lower limb.Tumor volumes were examined every 7 days.Mice were executed on the 35 days, and the eventual average tumor volumes and weights were examined.Total RNA and protein were isolated from tumors, and the expression of miR-34a,CDK6,and Bcl-2 mRNA and protein were determined by qRT-PCR and western blot, respectively. Results: The relative expressions of miR-34a was significantly up-regulated in miR-34a transfected group compared to Scrambled miRNA transfected group (P<0.05).Eight subcutaneous xenograft tumor models were successfully established. The nude mice of miR-34a transfected group appeared a smaller tumor volume compared to the other two groups at the beginning of 21th day( P<0.05).The eventual average tumor volumes in miR-34a group,Scrambled miRNA group and blank control group were(351.37±98.19)mm3,(798.75±91.04)mm3 and
(849.62±101.32) mm3 ,respectively, and the eventual average tumor weights in miR-34a group,Scrambled miRNA group and blank control group were(0.81±0.13)g,(1.47±0.21)g and(1.58±0.37)g,respectively.Both the eventual average tumor volumes and weights in miR-34a group were lower compared to the other two groups(P<0.05).qRT-PCR results revealed that the expression of miR-34a in miR-34a transfected group was significantly higher than in the other two groups, while the mRNA and protein expression of CDK6 and Bcl-2 were lower than the other two groups ( P<0.05 ) .Conclusion: miR-34a may inhibit the growth of human nasopharyngeal carcinoma CNE-2 cell line subcutaneous xenograft tumor in nude mice by down-regulating CDK6 and Bcl-2.
2.Evaluation and treatment of parkinson disease combined with pain
Journal of Chinese Physician 2017;19(6):952-955
Parkinson disease (PD) is a common neurodegenerative disease,of which clinical symptoms give priority to movement disorders, accompanied by a series of non-motor symptoms, such as pain.Studies suggest that 40% to 85% of PD patients combined with pain,of more nature is related to muscleskeletal and dystonia.PD combined with pain severely reduces the quality of life of patients. However,it is still lack of uniform clinical evaluation and effective treatment. This paper makes a brief overview about the status and research progress of the evaluation and treatment of PD combined with pain.
3.Effect of long-term inhaled glucocorticoids on height in children with asthma
International Journal of Pediatrics 2016;43(12):931-933,934
Asthma is the most common chronic respiratory disease in children. Currently, inhaled corti-costeroids is the most effective drug for asthma, however, there are some adverse effect by long-time using. Ac-cording to recent literature,long-time treatment with inhaled corticosteroids may inhibit height growth. But this suppression only occurs within early stage of asthma treatment, and have no effect or slight effect on adult height. The adverse effects of inhaled corticosteroids for height are also related to its iypes, dose and usage.
4.Change of heart rate turbulence in coronary artery disease patients and its predictive value
Chinese Journal of cardiovascular Rehabilitation Medicine 2015;24(1):1-5
Objective: To explore change of heart rate turbulence (HRT) in patients with coronary artery disease (CAD) and its predictive value. Methods: A total of 58 CAD patients were regard as CAD group. The 50 normal persons with negative CAG were regard as normal control group.
All subjects received 24h dynamic ECG, CAG and echocardiography examination. Left ventricular ejection fraction (LVEF) and HRT indexes were compared between two groups. Results: Compared with normal control group, there was significant rise in turbulence onset [TO, (-1.34±1.09)% vs. (2.32±0.60)%] and significant reduction in turbulence slope [TS, (11.14±2.27) ms/RR vs. (3.44±0.60)ms/RR] in CAD group, P<0.01 both; in CAD group, along with coronary artery lesion aggravated (single-, double-, multi-vessel coronary disease), there was significant rise in TO [(1.35±0.52)% vs. (2.56±0.83)% vs. (3.01±1.62)%] and significant reduction in TS [(6.49±1.79) ms/RR vs. (3.33±1.02) ms/RR vs. (1.66±0.30) ms/RR], the difference was significant in two-two comparison among all groups, P<0.05 or <0.01; compared with CAD without cardiac event group, there was significant rise in TO [(1.68±0.61)% vs. (2.24±0.24)%], and significant reduction in TS [(5.87±1.22) ms/RR vs. (1.65±0.23) ms/RR] and LVEF[(66.18±2.64)% vs. (56.36±3.34)%] in CAD with cardiac event group, P<0.01 all. Conclusion: Weakened HRT phenomenon exists in CAD patients. HRT can be treated as an index evaluating severity of CAD patients’ condition and an effective predicting index for recurrent cardiac events in CAD patients.

Chinese Journal of Orthopaedic Trauma 2002;0(02):-
Objective To study the effect of fetal transplants on the fu nctional recovery after spinal cord transection in new born and adult rats. Meth ods The spinal cord tissue at E14 of a rat fetal was transferred into the hemise ction cavity at lumbar spinal cord in the newborn and adult rats. 4, 8, 12 weeks after the operation, spinal cord tissue morphology, combined behavioral score ( CBS), somatosensory evoked potentials (SEP), and motor evoked potentials (MEP) w ere examined. Results The transplanted tissue survived in the host spinal cord. There was a statistical difference in CBS between the newborn graft group and th e adult graft group. The peak latencies of early waves in SEPs and MEPs of the n ewborn graft group were superior to those of the adult graft group (P
6.Advances in studies on the application of platelet-rich plasma in the field of orthopedicsand the underlying mechanisms
International Journal of Biomedical Engineering 2008;31(5):297-301
Platelet-rich plasma(PRP) is a highly-concentrated plasma enriched in platelets and various growth factors. These growth factors will be released abundantly by platelets when PRP is activated by thrombin, which plays a key role in accelerating wound healing, bone repair, and meanwhile, in controlling of inflammatory process. Biocompatibility and versatility of PRP have been confirmed in clinical studies as well as its excellent effects on tissue reparation. These have lead to the extensive use of PRP in many medical fields as a therapeutic alternative. Mechanism of PRP in bone and tissue repair the application of PRP in the field of orthopedics is reviewed in this article. In addition, attempt is made to discuss about some problems still existing in the clinical application of PRP.
