Objective To investigate the clinical curative effect of needling miuci fractures after treatment of knee joint dysfunction.Methods 60 patients with fracture fixation of knee joint function disorder were randomly divided into observation group and control group,30 cases in each group.The observation group was treated by needling method with conventional treatment.The control group received routine treatment only.The subjective pain (visual analogue scale)and IKDC score were used to score the patients with knee pain.Results The observation group compared with the control group,the subjective pain score decreased more significantly(t =2.050,P <0.05), while the IKDC score improved more significantly(t =2.185,P <0.05).The treatment effective rate of the observa-tion group was 93.33%,which was higher than 70.00% of the control group (χ2 =80.08,P <0.05).Conclusion The clinical curative effect of dysfunction of miuci combined with routine treatment of knee joint after operation is significant,and it is worthy of promotion.

Objective To evaluate the relationship between Helicobacter pylori (Hp) infection and pathohistological presentation of gastric and deuodenal mucosa.Methods The gastritis biopsies were taken through endoscopy from each gastric body,antrum and duodenum in 60 patients with gastritis and 22 patients with duodenal ulcer respectively for the examination of histology.Result The positive rate of Hp was nearly 100% in active chronic gastritis which was significantly higher than that of active duodenitis.The total detecting rate of Hp infection and incidence rate of active inflammation in antrum were obviously higher than that of gastric body and duodenal bulb.The detecting rate of Hp infection of duodenal bulb in 22 cases of duodenum ulcer was 54% which was significantly lower than that of antrum.Conclusion The Hp infection is a very important pathogenic factor of active inflammation in gastric mucosa on the basis of which the severe inflammation of mucosa and ulcer development.Therefore the early elimination of Hp is very important.

Angiogenesis is an important step during the occuring, development and metabsis of tumor which is regulated by some factors. PLGF is one of the VEGF family. It is homologous with VEGF-A, VEC,-FR-1 is its receptor. Although it expresses weakly during physiology. PLGF is an important pathologic angio-genesis factor at the condition such as anoxia, truma or tumor. Becanse its expression becomes strong at path-ologic cyophoria and many kinds of tumors, PLGF has a good value in clinic reasereh.

[Summary] With an increasing higher failure rate from 50%to 70%after simple sutured surgery for rotator cuff tears , scholars have put forward the patch augmented biomechanics for healing massive tears .Research hotspots lie in the material structural and physiological properties .There are two types of rotator cuff patches , which are synthetic materials ( non-degradable and degradable type ) and biological rotator cuff patch ( autologous or allogeneic tissue , xenogeneic material ) .The synthetic materials rotator cuff patch is able to provide powerful biomechanics but has severe immune rejection , and the biomaterials patch provides excellent histological reconstruction in rotator cuff and accurate regulation of biological activity of metabolic degradation but lower biomechanical maximum tensile strength .This article reviewed the current common problems and progress in the application of the rotator cuff patches .

Objective To explore the effects of arsenite on the expression of estrogen receptor (ER) mRNA,ER protein and the ER signaling pathway in rats.Methods Random grouping experiment design was used to evaluate the effects of arsenite on the expression of ER mRNA,ER protein and the ER signaling pathway (24 rats were divided into 4 groups:control,Aslow,Asmiddle,Ashigh groups;6 rats in each group).Female rats were exposed to NaAsO2 (0.625,2.500,10.000 mg/kg) by oral perfusion at gradient doses for three months respectively.The expression of ER mRNA as effect biomarker was observed through RT-PCR.The concentration of ER,vascular endothelial growth factor (VEGF),cyclin D1 (CYC-D1),and progesterone receptor (PR) in serum was determined as effect biomarkers using enzyme-linked immunosorbent assay (ELISA).Results The levels of ER mRNA,VEGF,PR,CYC-D1,and ER in control,Aslow,Asmiddle,and Ashigh groups were statistically different (F =204.13,227.14,256.13,179.23,167.34,all P ＜ 0.01).With increasing of arsenic,the expression of ER mRNA,VEGF,and PR gradually decreased (all P ＜ 0.01);the dose effect relationship was obvious;the expression of CYC-D1 and ER in Asmiddle [(25.45 ± 9.29) U/L,(22.16 ± 9.16) ng/L] was higher than that in Ashigh [(2.60 ± 0.23) U/L,(9.06 t 2.06) ng,/L,all P ＜ 0.01].Arsenic was negatively associated with ER mRNA,ER,VEGF,CYC-D1 and PR (r =-0.689,-0.515,-0.675,-0.456,-0.397,P ＜ 0.01 or ＜ 0.05).ER mRNA was positively associated with ER,VEGF,CYC-D1 and PR (r =0.894,0.662,0.713,0.699,P ＜ 0.01 or ＜ 0.05).Conclusions The expression of ER mRNA and ER protein are inhibited by arsenic,ER may be served as a sensitive biomarker of reproductive system damage after exposed to arsenic.The ER-VEGF,CYC-D1 and PR pathway participate in the process of reproductive system damage caused by arsenic.Arsenic is probably a environmental estrogen.

ObjectiveTo investigate the encountered life events in patients with tuberculosis within 1 year before the attack.MethodsThe encountered life events within 1 year before the attack were investigated in 85 patients with tuberculosis, the life event frequency (LEF) and life event intensity (LEI) were calculated respectively, and the life events were compared with those within 1 year in 55 healthy subjects.Results71 cases (83.5%) of the 85 patients with tuberculosis had encountered different life events within 1 year before the attack, which was significantly different from that in the control group (9 cases, 16.4%) (χ2=12.351,P<0.01 ). The LEF, negative LEF, average LEI, average negative LEI in the tuberculosis group before the attack were obviously higher than those in the control group (P<0.05～0.01), but there were no significant differences in the positive LEF and average positive LEI between the two groups (P>0.05).ConclusionThe encountered life events are closely correlated with primary tuberculosis, so more mental cares should be paid to the patients.

OBJECTIVEEstablished with amphotericin B perforated patch-clamp technique, to study the electrophysiological properties of calyx synapses.

METHODSIn the present experiments, we studied the application of perforated patch clamp technique on the calyx synapses of mice with Amphotericin B.

RESULTSThe use of Amphotericin B significantly slowed down the decay of channel currents and the optimum concentration was 400 microg/ml.

CONCLUSIONThe syudy developed a stable of perforated patch clamp whole cell recording technique, could be more effecitve, more real reaction neurons electrophysiological characteristics of the channel current. Our work might provide the basic information to future users studying the signal transmission and regulation of auditory system of rodents.

Cholecystolithiasis combined with choledocholithiasis is a common disease.The typical open surgery is challenged by the minimally invasive surgery recently.The minimally invasive surgery combined with laparoscopy and choledochoscopy or duodenoscopy has been accepted widely through analyzing and summarizing comprehensively the current situation of the minimally invasive surgery for cholecystolithiasis combined with choledocholithiasis.Laparoscopic cholecystectomy (LC) +endoscopic sphincterotomy (EST) or endoscopic papillary balloon dilation (EPBD) should be chosen primarily for the patients with cholecystolithiasis combined with choledocholithiasis and without common bile duct dilatation (common bile duct diameter ＜0.8 cm),and LC + laparoscopic transcystic common bile duct exploration and lithotomy are used under favorable conditions.LC + choledocholithotomy or T tube drainage should be chosen primarily for the patients with cholecystolithiasis combined with choledocholithiasis and common bile duct dilatation (common bile duct diameter ＞ 0.8 cm).Primary suture of common bile duct should be used with removal of the common bile duct stones,patency of distal common bile duct and recovery function of sphincter of Oddi.The minimally invasive surgery combined with laparoscopy and choledochoscopy or duodenoscopy which is selected reasonably could improve the treatment of cholecystolithiasis combined with choledocholithiasis and reduce the complications,with a significant clinical efficacy.

Objective To investigate the destructive effect of CSC-DC-CIK who were induced by cytokine induced killer (CIK) cells co-cultured with dendritic cells (DCs) on homologous tumor cells and to explore the possibility of CSC anti-gen involving in killing tumor. Methods Kidney cancer stem cells (KSCs) and lung cancer stem cells (LSCs) were isolated through FACS using CD133 +as a selection marker from cultured kidney cancer cell line A498 and lung cancer cell line A549 respectively. Freeze-thaw method was used to obtain the cancer stem cells(CSCs)antigens. DC cells and CIK cells were collected by in vitro expansion and inducted from the mononuclear cells isolated from human cord blood. The CIK cells were co-cultured with the DCs which were pulsed with the CSCs antigens(CSC-DC-CIK)mentioned above. Immunopheno-types of DC and CIK were analyzed by flow cytometry;cytokines levels were detected by ELISA kits and the destructive ef-fects of two kinds of CSC-DC-CIKs were tested by lactate dehydrogenase (LDH) release assay. Results The expression of phenotypes CD40+, CD80+, CD86+and HLA-DR+were higher in CSC-DC than in CD(P<0.01);the expression of pheno-types CD40+, CD80+, CD86+and HLA-DR+of DC and CSC-DC were higher after co-culture than those before co-culture( P<0.01);the expression of phenotypes CD40+, CD80+, CD86+and HLA-DR+of CSC-DC after been co-cultured with CIK were higher than those of DC after been co-cultured with CIK(P<0.01). The CIK phenotypes:CD3+, CD8+, CD56+were in-creased in CIK co-cultured with both CSC-DC and DC than those before co-culture (P<0.01);the expression of pheno-types CD3+, CD8+, CD56 +were higher in CSC-DC co-cultured with CIK than in DC co-cultured with CIK. DC-CIK and CSC-DC-CIK groups were more capable to express IFN-γ, TNF-α, IL-2 than they were before co-cultured with CIK (P<0.01). CSC-DC-CIK group can secrete more above cytokines than DC-CIK group does(P<0.01). The destructive rates of KSC-DC-CIK and LSC-DC-CIK on target cells were (50.21 ± 4.24)%and (49.32 ± 3.89)%respectively which were much higher than that in DC-CIK(30.25±3.11)%(F=89.157,P<0.01). Conclusion CSC-DC-CIKs have destructive effects on homologous tumor cells. More researches are needed to explore the mechanism and to evaluate the clinical applications.
