1.Research of platelets activation during Inflammation

Platelets is the important hemostatic component in the blood and the critical participants in inflammation. It is an important promoting factor during inflammation and can recruit leukocytes and aggregate in sepsis. Decreasing plate?let count was correlated with reverse clinical outcome. Therefore, the value of the platelets examination in clinical monitor was studied by many researchers. Under circumstance of fungal infections, platelets mediate antimicrobial activity and assist dissemination of the fungi synchronously. Regulating interaction between platelets and fungi is difficult. In allergic inflamma?tion patients, the excessive activating of platelets aggravates airway obstruction and worsen pulmonary function. We reviewed current research in activating platelets during inflammation.
2.Construction of LuxS gene deletion of Streptococcus mutans and evaluation of the acid tolerance of the mutant

Objective To construct Streptococcus mutans UA159 mutants with deletion of LuxS gene related to quorum-sensing pathway and evaluate the aciduricity of the mutants. Methods Using S. mutans UA159 as materials, the PCR fragments of the upstream and downstream regions of LuxS and erythromycin resistance(Eymr) gene of PJT10 were cloned into plasmid PUC19. The resulting constructs were integrated into the chromosome of S. mutans. LuxS gene deletion mutant was then constructed in S. mutans by means of allelic exchange and selected for resistance to erythromycin. The aciduric ability of the mutant under different pH was measured and S. mutans UA159 was used as control. Results The LuxS-deleted status of S. mutans mutants were confirmed by various PCR and DNA sequencing. The results showed that Eymr gene take the place of LuxS gene, while the mutant can not induce bioluminescenece. The LuxS mutant strain displayed a decreased growth ability with the decreasing pH values compared to those of the wild-type strain UA159. Conclusion A LuxS-negative mutants of S. mutans is constructed. The LuxS quorum sensing system is involved in the regulation of aciduricity of S. mutans UA159.
3.Usefulness of Utilin S in Treatment of COPD

OBJECTIVE:To observe thc usefulness of Utilin S in treatment of chronic obstructive pulmonary disease(COP- D). METHODS: In a random control design, trial group was given Utilin S 1. 72ug im, qw. on the basis of conventional treat- ment. Control group was given conventional treatment alone. RESLLTS: The clinical effect. symptom and immune indices of trial group were all better then those of control group. CONCLUSION: Utilin S, as a immunostimulant. has a good accessory therapcutic effect on COPD.
4.Effects of Atorvastatin with Different Doses on Related Indicators and MACE in Patients with Acute Myocar-dial Infarction after PCI

OBJECTIVE:To investigate the effects of atorvastatin with different doses on related indicators and major adverse cardiovascular events(MACE)in patients with acute myocardial infarction(AMI)after percutaneous coronary intervention(PCI). METHODS:120 patients with AMI after PCI were randomly divided into conventional dose group(60 cases)and high dose group (60 cases). All patients were immediately given low molecular weight heparin,Aspirin enteric-coated tablet,Clopidogrel bisulfate tablet,Angiotensin-converting enzyme inhibitor,β-blockers,nitrates and other conventional treatment;based on it,conventional dose group was given 20 mg Atorvastatin calcium tablet,once every day before bedtime;high dose group was given 40 mg Atorv-astatin calcium tablet,once every day before bedtime. The treatment course for both groups was 1 month. Total cholesterol(TC), triglyceride(TG),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol(HDL-C),C-reactive protein (CRP),OX40L,interleukin (IL)-6 levels in 2 groups before and after treatment were observed,the correlation among OX40L with CRP and IL-6 were analyzed,and the incidence of MACE in 2 groups was recorded after 1 month follow-up. RESULTS:Be-fore treatment,there were no significant differences in the TG,TC,LDL-C,HDL-C,OX40L,CRP and IL-6 between 2 groups (P>0.05). After treatment, TG, TC, LDL-C, OX40L, CRP and IL-6 were aignificantly lower than before, TC, LDL-C, OX40L,CRP and IL-6 in high dose group were lower than conventional dose group,HDL-C was significantly higher than before, the differences were statistically significant(P<0.01 or P<0.05);but there was no significant difference in the HDL-C between 2 groups(P>0.05). Before treatment,OX40L showed positive correlation with CRP(r=0.746,P<0.01)and IL-6(r=0.763,P<0.01);after treatment,OX40L also showed positive correlation with CRP (r=0.755,P<0.01) and IL-6 (r=0.760,P<0.01). The incidence of MACE in high dose group were significantly lower than conventional dose group,the difference was statistically significant(P<0.05). CONCLUSIONS:Based on the conventional treatment,atorvastatin can significantly reduce lipid,inflamma-tion levels and incidence of MACE,but the effect is more significant in high dose.
5.Influence factors of modified mini-mental state examination evaluation on 6819 elderly people

Objective To explore the main influence factors of ederly people's cognitive function and provide the countermeasures. Methods A cross-sectional study was performed in the elderly volunteers who were sampled from halves of the counties of 22 provinces in China.Modified minimental state examination(MMSE)scale was adopted to investigate the 6819 Chinese elderly people who were investigated in the Chinese elderly health questionnaire in 2002.Simple Chi-square test and multiple logistic regressions were applied in the analysis. Results The results showed that the perfect rate of cognition function was 85.0%in male,and 80.43%in female.The rate was 97.7%in people at age of 60-69 years,94.8%,70-79 years,85.0%,80-89 years,71.11%,90-99 years,55.7%,100-111 years,94.8%.There existed significant cognitive function differences in gender,age,education,born place,marital status,1iving status,smoking history,drinking history,physical exercise,self-satisfaction with life,affective state and healthy status(all P<0.01).Non-conditional logistic regression analysis showed that influence factors of cognitive function of the elderly were age,education level,marital status,physical exercise and optimistic state. Conclusions The risk factors for cognitive function are aging and widowhood.Higher education,regular physical exercise,optimistic state are protective factors for cognitive function.
6.Influence of out-hospital nursing with no gap on the level of HbAlc for patients with diabetes

Objective To know the influence of out-hospital nursing with no gap on the level of HbAlc for patients with diabetes.Methods Divided 80 patients with diabetes into the intervention group and control group randomly,there were 40 cases in the each group.Out-hospital nursing cares with no gap were used in the intervention group,while the routine nursing cares were used in the control group.Fellowed up 1 year.Compared the FPG,2 hPG and HbAlc between the two groups at the time points of the 3rd,6th,9th and 12th month when out of hospital.Results All the indexes which had monitored were better in the intervention group than those of in control group.Conclusions Out-hospital nursing with no gap can remain the indexes in the normal level,and then promote their quality of life,which should be developed in clinical field.
7.Anti-bacterial Effects of Baicalin on One Escherichia coli Strain Carrying NDM-1 Gene

Objective To examine the bacteriostasis of baicalin against Escherichiacoli strain carrying NDM-1 gene.Methods Minimal inhibitory concentration(MIC)and minimal bactericidal concentration(MBC)were determined by broth dilution method.Synergy function was designed withcheckerboard method.Anti-infection effect in vivo of baicalin was observed in mice with bacteremia.Results In vitro antibacterial tests showed that the MIC and MBC baicalin were 8 mg/ml.Synergistic inhibitory effect was observed between baicalin and imipenem (FIC=0.125).In vivo inhibition experiments showed that ba-icalin had decreased the mortality of 25% of Escherichiacoli Strain carrying NDM-1 gene infection in mice.Conclusion Ba-icalin has antibacterial effect on NDM-1 E.coli.
8.Protective effect of non-mitogenic haFGF on cerebral ischemia-reperfusion injury in mice

Aim To investigate the protective effect of non-mitogenic human acidic fibroblast growth factor (nm-haFGF) on cerebral ischemia-reperfusion injury in mice. Methods Cerebral ischemia-reperfusion model was made by ligating bilateral carotid for 20 minutes in mice. These mice were randomly divided into model group( iv NS), two doses of nm-haFGF (iv 25、50 ?g?kg-1) groups, rhaFGF group(iv 50 ?g?kg-1) and sham- operated group. Step down test and Y-type electric maze were used to examine the effect of nm-haFGF on learning and memory of mice, then Even′s Blue(EB) level and NO level in brain of these mice were measured. Results The nm-haFGF significantly decreased numbers of errors of mice in 5 min in step down test and in Y-type electric maze test; EB and NO levels in brain of these mice were lower than those of model group respectively. Conclusion The nm-haFGF can protect cerebral ischemia-reperfusion injury in mice.
9.Comparison of wound healing in two mouse models of type 2 diabetes

Objective To compare the characteristics of ulcer wound healing in current commonly used C57BL/6J-db/db mouse models of spontaneous gene mutation-induced type 2 diabetes and in C57BL/6J mice with diabetes induced by streptozotocin (STZ), and to provide a basis for related experimental studies on diabetic ulcer in animal models.Methods To establish the mouse models of diabetic ulcer wound, observe the healing time and calculate the wound healing rate at 0, 3, 5, 7, 10, 14 days.Tissue samples were collected at days 7 and 14.HE and Masson staining, and immunohistochemistry (CD31 and PCNA) were used to observe the pathological changes of the wound tissues.Gene expressions of collagen-IIIα, fibronectin and α-SMA were detected by fluorescent quantitative analysis.Results The wound healing time of db/db mice was significantly delayed compared with the STZ mice, which was extended from 16.6±0.8 d to 20.2±1.3 d (P< 0.001).Compared with the STZ group, the growth of granulation tissue in the db/db group was slow, the length of newly formed epithelium was insufficient, the collagen deposition was disordered, and the wound healing was poor.At 7 days, the expression of CD31 and PCNA was significantly lower in the db/db group (P< 0.01), and at 7 and 14 days, the increase of collagen-IIIα and α-SMA genes up-regulation was significantly lower in the db/db group than in the STZ group.Conclusions Both the two types of diabetic mice show delayed wound healing.However, compared with the STZ-induced diabetic mice, the gene mutation db/db mice are more suitable for studies of diabetic ulcer wound healing as regarding the extent of the delay and the degree of difficulty of wound healing.
