1.The effect of 3-n-butylphthalide on learning and memory abilities in vascular dementia rats

Objective To investigate the effect of 3-n-butylphthalide (NBP) on learning and memory abilities in vascular demensia (VD) rats. Methods VD model rats was established by two-vessel method. 60 3-month-old Wistar male rats were randomly divided into VD group, sham-operate group and NBP group. Rats in NBP group were given NBP 120 mg · kg-1 · d-1 ,VD and sham-operate group were given equal quality vegetable oil.Morris water maze was used to assess the learning and memory abilities in each group and HE staining was used to observe the hippocampus morphology of the rats. Results The escape latency in hidden plat test was( (38.34 ±2.46 ) s, ( 14.83 ± 3.77s ), ( 75.74 ± 6.33 ) s ) and the original platform quadrant time was ( ( 26.45 ± 4.66 ) s,(35.21 ±3.78)s, ( 18.67 ±5.39)s) in NBP group,sham-operate group and VD group respectively. Compared to VD group, NBP group had obviously decreased escape latency in hidden plat test, increased original platform quadrant time and distinctly decreased the necrosis of the neurons in HE dyeing. Conclusion NBP can improve the learning and memory deficits in VD rats.
2.The Relationship of Blood Glucose and Blood Lipids with ?-cell Function and Insulin Resistance in Adult University Staff with Normal Glucose Tolerance and Impaired Blood Glucose

Objective To investigate the prevalence rates of diabetes, impaired fasting glucose (IFG), impaired glucose tolerance (IGT) and related factors in the university staff. Methods 617 subjects were selected from 1 519 cases with the fasting blood glucose level ≥5.6 mmol / L in the annually physical examination among 3 851 population in a university community. All 617 subjects were taken the blood glucose and insulin test for the fasting and 2 hour after 75 g glucose intake. The fasting lipids, BUN and Cr were determined. The demographic data including height, weight, waist circumference and blood pressure was also collected. Results The numbers of normal glucose tolerance (NGT), IFG, IGT, IFG+IGT and newly diagnosed DM were confirmed in 374 (60.62%), 60 (9.72%), 59 (9.56%), 41 (6.65%) and 83 (13.45%); totally, 243 (39.38%) cases with abnormal glucose metabolism. The highest DM prevalence was found in the group with BMI ≥28 kg / m2. In the group with hypertriglyceridemia, prevalence of DM, IFG and IFG +IGT was 12.50%, 8.33% and 8.33%. In the group with hypercholesteremia and hypertriglyceridemia together, prevalence of DM, IFG+IGT, IFG and IGT was 18.87%, 7.17%, 11.32% and 12.83%. DM prevalence increased with the higher triglycerides level. No differences of HOMA-IR was found among groups of DM (0.80?0.82), IFG (0.64?0.72), IFG+IGT (0.61?0.77), IGT (0.35?0.68), but all these obviously higher than that of NGT group (0.17?0.80). No differences of HOMA-B among the IGT (3.97?0.69), NGT (3.95?0.78), IFG (3.84?0.72), IFG+IGT (3.80?0.78) groups, but all these higher than that of NGT(3.69?0.88)group. Conclusion About 40% of these subjects in the university population had their blood glucose ≥5.6 mmol/L. In these subjects with blood glucose ≥5.6 mmol/L, about 40% of them had abnormal blood glucose level, including 13.45% diabetic patients. The highest prevalence of IFG, IGT, IFG+IGT was found in the group aged 60~70, the highest prevalence of DM in the group aged 70~80 and increased paralleled with the BMI and aging, was associated with lipids disorder, particularly with hypertriglyceridemia. IR existed in the pre-diabetes state and was worst in the DM, with most severe ? cell function failure.

4.Multi-mode analgesia of celecoxib after hip and knee replacement

BACKGROUND:Hip and knee replacement is a common surgery in the clinic; the procedure is relatively complex; and the risk of surgery is relatively high, so that postoperative analgesia is not satisfactory. Perioperative pain management has been a clinical concern. To find safe and effective analgesia has become one of the important tasks of joint surgeons. OBJECTIVE:To investigate the effect of celecoxib on multi-mode analgesia after hip and knee replacement. METHODS: A total of 80 cases undergoing hip and knee replacement in the Chongqing Dongnan Hospital from September 2012 to September 2013 were enroled in this study. These patients were randomly divided into two groups. In the control group, celecoxib was not used for analgesia after replacement. In the experimental group, celecoxib was used after replacement. The pain was observed at 1-5 days after surgery in the two groups. When the analgesic pump was removed, the drug dosage and opioid analgesics use were recorded. Side effects of drug use were also recorded. RESULTS AND CONCLUSION:In terms of analgesic efficacy, the analgesic effect was better in the experimental group than in the control group (95%, 85%, P < 0.05). 95% patients in the experimental group were satisfied with the analgesia, which was significantly higher than in the control group (65%;P< 0.05). No significant difference in pain visual analogue scale score was detected between the two groups immediately, 4 and 5 days after surgery (P > 0.05). Pain visual analogue scale score was significantly lower in the experimental group than in the control group at 1, 2 and 3 days post surgery (P< 0.05). The drug dosage was significantly more in the experimental group than in the control group after surgery (P < 0.05). The frequency of opioid use in the experimental group was significantly lower than in the control group (P < 0.05). The complication rate was significantly lower in the experimental group (8%) than in the control group (18%) (P < 0.05). These findings demonstrate that the analgesic effect of celecoxib was ideal after hip and knee replacement using multi-mode analgesia, which can reduce the dose of analgesic drugs and have smal adverse reaction.
5.Analysis of type 2 diabetic patients after a 3-year treatment

Objective To compare and analyze the metabolic control with the islet B cell function and the diabetic complications after a 3-year treatment in type 2 diabetic patients.Methods The T2DM patients were investigated who finished 3 years of treatment.The following measurements or determination was done:height,weight,waist,hips,blood pressure at standing and lying position,fasting glucose and insulin,glucose and insulin 2 h after taking food,HbA_1c,TC,TG,HDL-C,LDL-C,Cr,albuminuria and retinopathy.Results Totally 233 male and 178 female(56.53?10.33)yrs patients were analysized.The hypotensive,hypoglycemic and lipids agents were increased obviously and the number of the patients with insulin treatment increased too.HbA_1c was decreased,but no significant differences for the blood pressure and blood cholesterol,triglycerides after 3-year treatment.The fasting and post-meal insulin level,insulin resistance and the B-cell function index decreased significantly.Meanwhile,the patients with albuminuria and retinopathy increased from 15% and 26% to 23% and 33%.Conclusion Type 2 diabetic patients have not controlled the blood glucose,lipids and the blood pressure satisfactorily even if the agents for the blood sugar,lipids and blood pressure lowing are increased.Insulin resistance decreases but B cell function failure aggravates.The intensified therapy on the target should be paid more attention to.
6.Study of antibiotic resistance of clinical isolates of helicobacter pylori

Objective To analyze the susceptible range and antibiotic resistance of clinical isolates of Helicobacter pylori(H.pylori) in Shijiazhuang area.Methods (1)H.pylori strains:55.(2)Drugs:Metronidazole,Ornidazole,Azithrumyein,Clarithromycin,Amoxicillin,Tetracycline and Furazolidone.(3)Sensitive test:filter paper agar diffusion method.Results The antibiotic resistance rate of Metronidazole,Ornidazole,Azithrumyein,Clarithromycin,Amoxicillin,Tetracycline and Furazolidone was 92.73%,85.45%,45.45%,9.09%,0,0,0,respectively.Conclusion The antibiotic resistance rate of H.pylori from Shijiazhuang area to Metronidazole,Ornidazole and Azithrumyein are very high,and the resistant strains to Clarithromycin has been found.There are no resistant strains to Amoxicillin,Tetracycline and Furazolidone.
7.Study on methylation of the p73 gene in acute lymphoblastic leukemia and its clinical significances

Objective To investigate the effects of methylation of p73 gene on pathogenesis of acute lymphoblastic leukemia.Methods The restriction endonuclease enzyme digestion combined with PCR was used to detect p73 gene and methylation in 42 patients with acute lymphoblastic leukemia.Results Methylation of p73 gene was found in 28.6%(12/42).Patients without methylation of p73 gene had a higher complete remission rate of first chemotherapy and a longer survival time than those with methylation of p73 gene.Conclusion Methylation of p73 gene probably plays a role in the pathogenesis of acute lymphoblastic leukemia and may have clinical significance in predicting prognosis of ALL.
