Neurotrophic factors are proteins that play an important role in the development and maintenance of neural networks. They released in the brain's response to injury and in neurodegenerative disorders and took part in the protective process. The fact that they can be used to prevent damage to neurons associated with brain lesions in animal models of neurodegenerative disorders supports the above mentioned hypothesis as well. However, they are proteins that do not easily cross the blood brain barrier. The induction of neurotrophic factors by systemically administered some medicines would be an elegant way to overcome problems of application.

Objective To investigate the risk factors of uroschesis in patients with cervical cancer after extensive hysterectomy.Methods The clinical data of 92 patients with cervical cancer who had undergone extensive hysterectomy plus pelvic lymph node dissection from August 2008 to June 2011 were analyzed retrospectively.The incidence rate and related risk factors of uroschesis after operation were analyzed.Results Thirty-one of 92 cases had postoperative uroschesis.The incidence rate of uroschesis was 33.70％.Univariate analysis showed that age,intraoperative blood loss,time of indwelling urinary catheter and urinary tract infection had significant correlation with postoperative uroschesis (P ＜0.05).While operative time,clinical stage,pathological type,preoperative adjuvant therapy,postoperative wound infection or fat liquefaction had no obvious correlation with postoperative uroschesis (P＞ 0.05).Multivariate Logistic regression analysis showed that age,time of indwelling urinary catheter and urinary tract infection were risk factors of postoperative uroschesis.Conclusions Age,time of indwelling urinary catheter and urinary tract infection are independent factors of postoperative uroschesis.The key method to reduce the incidence rate of postoperative uroschesis is to shorten the continuous catheterization time with comprehensive measure,to prevent and control urinary tract infection in early time.

Background and purpose:In order to look for the new biomarker with higher specificity and sensitivity, we investigated the relationship between expression of MGB-2 and chemotherapy response, and tried to find out whether MGB-2 may sever as a prognostic factor for the patients with ovarian carcinoma. Methods:The expression of MGB-2 was detected by flow-cytometry technology in ovarian carcinoma tissue. We analyzed the correlations among MGB-2 expression and clinical pathological factors, therapeutic effect and prognosis of the patients with ovarian carcinoma.Results:Positive detection of MGB-2 was 78%.(66/885). For the patients whose serum CA125 level did not drop to normal level after two courses of chemotherapy, the MGB-2 positive rate was significantly higher than in those whose serum CA125 level decreased markedly ( 85% vs. 62%, P=0.018). The positive rate was also higher in patients with recurrent disease than in those without recurrence (83% vs. 67%, P=0.097). In univariate survival analysis, the expression of MGB-2 was significantly associated with median survival time (P=0.021).Conclusions:MGB-2 expression is correlated with chemotherapy response and poor prognostic for the patients with ovarian carcinoma.

Objective To investigate the effect of chronic intermittent hypoxia on blood pressure,sympathetic nerve activity and myocardial mitochondrial oxidative stress status in aged rats.Methods Chronic intermittent hypoxia(CIH)rat models were established.A total of 84 male Wistar rats(3-4 and 23-24 months old,respectively)were randomly and equally divided into four groups according to age:young control group,young CIH group,aged control group and aged CIH group.CIH exposure lasted for 3 weeks.The arterial blood pressure,the frequency of carotid sinus nerve action potential,the lung function,the levels of catecholamine,aconitase/fumarate ratio (A/F),plasma superoxide dismutase(SOD)in the plasma were measured and compared between four groups.Results The arterial blood pressure was significantly higher in young CIH group than in the young control group[(150.4 ±25.6)mmHg vs.(102.2±7.4)mmHg,P＜0.01].There was no significant difference in the arterial blood pressure between the aged CIH group[(132.8 ±16.2) mmHg] and the aged control group[(127.1 ± 26.8)mmHg].The minute volume ventilation was significantly lower in the aged control group than in young controlgroup[(331.7±53.5)ml· min-1 · kg-1vs.(554.8±111.9)ml· min-1 · kg-1P＜0.05].The min-1 · kg-1 ventilation showed no significant difference between the agedCIH group[(354.1±51.9)ml· min-1 · kg-1]and the aged control group[(331.7 ±53.5)ml· min-1 · kg-1].The rate of CSN frequency change by hypoxia stimulation was lower in the aged control group than in young control group[(6.2± 5.5)times vs.(11.8 ±6.6)times,P＜0.01],and was lower in the aged CIH group than in the young CIH group[(22.2 ± 13.5) times vs.(44.2± 12.1) times,P＜0.01].The levels of plasma norepinephrine and epinephrine were higher in the young CIH group than in the control group[(39.0±8.9)nmol/L vs.(20.8±10.6)nmol/L,(48.1±13.6)nmol/L vs.(26.7 ± 14.3)nmol/L,both P＜0.05].The levels of plasma norepinephrine and epinephrine showed no significant difference between the aged control group and the young control group,also no significant difference was found between the aged CIH group and the aged control group.The A/F ratio was lower in the young CIH group than in the young control group[(0.26±0.13)vs.(0.58 ±0.04),P＜0.01].The A/F ratio was lower in the aged control group than in the young control group [(0.29±0.02)vs.(0.58± 0.04),P＜0.01],but there was no significant difference in the A/F ratio between the aged CIH group and the aged control group.The level of SOD was significantly lower in the young CIH group than in the young control group [(5.30 ±± 0.90) NU/mgprot vs.(6.10 ± 1.73)NU/mgprot,P＜0.05].There was no significant difference in SOD level between aged control group and aged CIH group.Conclusions The effect of CIH on blood pressure in aged rats is different from the youth rats.This study shows adecreasedsympathetic nervous over activity and a reducedmitochondrial oxidative stress in aged rats.Aging probably suppresses two important pathogenic mechanisms,by which CIH affects blood pressure,therefore releases CIH induced blood pressure abnormality.

In both clinical and animal experiments,it has been confirmed that the inflammation in brain is involved in the development of brain damage after the ischemia.However,the pro-inflammatory mechanism is extremely complicated.Recently,a growing number of reports indicates that many factors play their roles in the inflammation,such as leukocyte,interleukin-1?(IL-1?),tumor necrosis factor(TNF)and mast cells.They are released/presented in the region of cerebral ischemia and include both ischemic and secondary brain injury.

The purpose of the present study is to use beta-cyclodextrin polymers (beta-CDP) with different cross-linked degree (CLD) to form inclusion complexes with ibuprofen and examine the effects of structural and compositional factors of beta-CDP on its drug loading and release behaviors. A series of beta-CDP with different CLD were synthesized and characterized by Fourier Transform Infrared Spectroscopy (FT-IR) and 13C NMR spectrum. The beta-CDP was systemically characterized for the relation between the CLD of beta-CDP and the drug loading and release as well. The results of FT-IR and 13C NMR showed that similar peak-shaped vibration of beta-CDP and beta-CD implies that the polymer keeps the original characteristic structure of beta-CD. The CLD of the beta-CDP played a critical role in the drug loading and release, increasing the CLD resulted in reduction of drug loading, but increase in drug release.

OBJECTIVE: To investigate the expressions of transforming growth factor beta1 (TGF-beta1) and bone morphogenetic protein-2 (BMP-2) in human mandible fracture callus and their quantity changes in the process of healing. METHOD: Thirty callus samples from the fractured mandible bone stumps were collected during operation, and two callus samples were collected from the angle-square jaw patients as controls. The expressions of TGF-beta1 and BMP-2 were test by the immunohistochemistry technic-SABC-staining in different periods of human fractured mandibular callus and in osseous tissue of normal angle of mandible. RESULT: The TGF-beta1 and BMP-2 were expressed in callus of different periods but not in normal bone tissue. The expression of TGF-beta1 increased slowly during the first three weeks after fracture and reached its maximum in the third week, and then weakened gradually. The expression of BMP-2 increased gradually during the first two weeks after fracture and reached its maximum in the second week, then the expression weakened gradually. CONCLUSION (1) BMP-2 may be one of the factors promoting the repair of fracture. (2) TGF-beta1 could be another signal pathway in repairment of fracture. (3) There could exist some synergistic effects between TGF-beta1 and BMP-2 in the process of fracture healing.

Objective To investigated the effects of intermittent hypoxia on neuronal apoptosis and autophagy in hippocampus.Methods 30 Wistar rats were randomly divided into normal control group (NC),intermittent normoxia group (IN) and intermittent hypoxia group (IH).The spatial learning and memory function of the rat was assessed using Morris water maze test.The apoptotic cells and the ultrastructure of neurons in the hippocampus tissue were observed by TUNEL and transmission electron microscope,respectively.And the expression of autophagy marker protein LC3 and Beclin-1 were measured by Western blotting.Results The escape latency was significantly longer in IH than in NC and IN group.And the ratio of time spent in the target quadrant was lower in the IH group than in NC and IN group (P＜0.05).The apoptotic rate of rat hippocampal neurons (F =6.01,P=0.037),the amount of double-layer membrane structure-complicating autophagic vacuoles with karyopyknosis,and protein expression level of LC3 and Beclin-1 were significantly higher (all P ＜0.05) in intermittent hypoxia group than in IN and NC group.Conclusions Intermittent hypoxiainduced autophagy and apoptosis in rat hippocampus are significantly increased,which might be one of the possible mechanisms for cognitive dysfunction caused by intermittent hypoxia.

Objective To observe the effects of intensive-swimming training at different intensities on the neurological function and the expressions of protein kinase A after cerebral ischemia-reperfusion in rats,and to explore the possible neuroprotection mechanism of intensive-swimming training after cerebral ischemia-reperfusion in rats.Methods The animal model of cerebral ischemia-reperfusion was established in rats by using the intraluminal thread method to cause left middle cerebral artery occlusion (MCAO) for 2 h and then reperfusion.A total of 120 male model Sprague-Dawley rats were randomly divided into training group 1 (swimming for 5 min once a day),training group 2 (swimming for 10 min once a day),training group 3 (swimming for 20 min once a day) and control group (no training),with 30 rats in each group.Another 30 rats who were assigned to the sham-operation group were subject to no MCAO and no training.At the 3rd,7th and 14th day post operation,10 rats randomly selected from each group were examined for their neurological function by Menzies neurobehavioral test and forelimb placing test.Expressions of protein kinase A were detected by immunohistochemistry method.Expressions of PKA-mRNA were detected by using RT-PCR.Results The neurological function in sham-operation group was normal.The differences between sham-operation group and control group with regard to Menzies test and forelimb placing test scores were statistically significant at all the time points (P ＜ 0.05).The improvement of neurological function in all training groups,were significantly better than those in controls at the 3rd,7th and 14th d after swimming training,especially in training group 3 on the 14th day post-operation (P ＜0.05).At3rd,7th,and 14th days post-operation,the Menziesscores were(1.40±0.55),(1.00 ±0.71)and (0.80 ±0.45),respectively,and the forelimb placing scores were(5.20 ± 0.84),(3.80 ± 1.30) and (2.40 ± 1.14),respectively,in the training group 3.The number of positive cells and the expressions of protein kinase A mRNA were significantly higher in all training groups than those in the controls at the 3rd,7th and 14th days after swimming training,and the increase of positive cells and mRNA of PKA in the training group 3 was significantly more obvious at 14th day compared with other training groups(P ＜ 0.05).Conclusions Exercise training can increase the expression of positive cells and mRNA of PKA in rats after cerebral ischemiareperfusion,hence,it improved the injured neurological function and promote the recover of sensory and motor function,and the more intensive the training,the better the effects.
