3.Gene Disruption in Yeast

Gene disruption by homologous recombination is a powerfu l tool for investigating gene function in yeast. Since 1980’s, it has been deve loped a lot. PCR-mediated gene disruption technique makes the manipulation easi er and it can be used to precisely delete genes in yeast. Multi-gene disruption technique can delete several genes successively in the same yeast strain. After the completion of the yeast Saccharomyces cerevisiae genome sequencing, the gene disruption technique for systematic analysis meets the need of the functio nal genomics in yeast. It also enlighten the study on human functional genomics.
4.Carotid intima-media thickness and uric acid level and the research of the prognosis of patients with coronary heart disease

Objective To explore the carotid intima-media thickness and the value of uric acid in predicting the prognosis of coronary heart disease patients.Methods Using the random number table method,182 patients diagnosed with coronary heart disease were divided into two groups:group A was 92 cases of acute coronary syndrome (ACS),groups B was 90 cases of chronic stable angina,healthy check-up 90 cases as control group (group C),fasting blood test blood uric acid (UA),application of color Doppler ultrasound measurement carotid intima-media thick ness (IMT).Results Acute coronary syndrome (ACS) set of uric acid,IMT,plaque index were (428.5 ± 70.3) mmol/L,(1.58 ± 0.92) mm,(3.24 ± 1.81) and those in stable angina group were (356.4 ± 68.2) mmol/L,(1.32 ± 0.86) mm,(3.08 ± 1.62) and those of the control group were (251.2 ± 65.4) mmol/L,(0.92 ± 0.41) ram,(2.80 ± 1.18) to compare the difference was statistically significant(group A compared with group B(t =4.47,3.26,5.21,all P < 0.05),group A compared with group C (t =8.23,9.36,7.48,all P < 0.01);group SAP compared with control group the difference was statistically significant (t =6.34,5.26,7.12,all P < 0.05);Linear correlation analysis showed that uric acid and carotid intima-media thickness were positively correlated,r value was 0.67.The ACS group soft carotid atherosclerosis plaque detection rate was 60.9%,SAP group was 26.7% and the control group was 8.9%,the difference was statistically significant (x2 =21.60,P < 0.05;x2 =69.11,P < 0.01),group SAP compared with the control group,the difference was statistically significant (x2 =39.84,P < 0.01).Conclusion Uric acid levels and carotid artery atherosclerosis and coronary artery disease are closely related,which can provide theoretical basis for the risk assessment and prognosis of coronary heart disease.Early intervention can prevent the occurrence of serious cardiovascular events.

6.The application of mammotome system in diagnosis and treatment of breast tumor: a report of 115 cases

Objective To investigate the value of clinical application of mammotome under ultrasonic guidance for minimal excision and biopsy of breast tumor.Methods Excision and biopsy of 152 breast masses from 115 patients were performed using mammatome under ultrasonic guidance,and the patients were followed up.Results Total resection of 152 breast tumors were accurately performed by mammotome vacuum device and biopsy was performed.On pathological examination,86 cases were fibroadenoma,34 cases were adenosis,2 cases were fat necrosis,3 cases were gynecomastia,1 case was ductal inflammatory change and 26 cases were breast cancer.Postoperatively,hematoma was found in 3 cases,subcutaneous ecchymosis in 4 cases and skin injury in one case.All cases were followed up for a period of time and no residual or recurrent masses were found.Conclusions Mammotome vacuum system is an exact,effective and safe method.It is an ideal minimally invasive operation in treating benign breast masses with few complications.It can also be applied to biopsy for breast cancer before operation.
7.Modulating drug loading and release profile of beta-cyclodextrin polymers by means of cross-linked degree.

The purpose of the present study is to use beta-cyclodextrin polymers (beta-CDP) with different cross-linked degree (CLD) to form inclusion complexes with ibuprofen and examine the effects of structural and compositional factors of beta-CDP on its drug loading and release behaviors. A series of beta-CDP with different CLD were synthesized and characterized by Fourier Transform Infrared Spectroscopy (FT-IR) and 13C NMR spectrum. The beta-CDP was systemically characterized for the relation between the CLD of beta-CDP and the drug loading and release as well. The results of FT-IR and 13C NMR showed that similar peak-shaped vibration of beta-CDP and beta-CD implies that the polymer keeps the original characteristic structure of beta-CD. The CLD of the beta-CDP played a critical role in the drug loading and release, increasing the CLD resulted in reduction of drug loading, but increase in drug release.
8.Study on the competency of community health administrators

Objective To explore the competency of community health administrators in a survey of such administrators at large in Shanghai. Methods McClelland's Competency Dictionary was used to set two benchmark positions, while the behavioral event interviewing and questionnaires were made to survey 19 community health service centers in Shanghai. Results The competency model benchmarking on the head of a medical branch consists of 6 character clusters, 11 characteristic items, 27 evaluation dimensions and 27 typical behavior descriptions. The model benchmarking on the community team leader consists of 5 character clusters, 8 characteristic items, 21 evaluation dimensions and 21 typical behavior descriptions. Conclusion Findings of the survey provide objective criteria for competence appraisal of community health administrators, and serve as reference for formulating their development plans as well.
9.Effects of strict control of tobacco advertisement after implementation of the Advertising Law

Objective · To compare the prevalence of tobacco advertisement before and after implementation of the new edition of Advertising Law of the People's Republic of China and evaluate the effects of implementation of the new Advertising Law. Methods · Tobacco sales places, public places, and streets in Shanghai were randomly selected to observe tobacco advertisement and complete the survey. The survey was carried out in 10 districts of Shanghai in June 2015 with 10 tobacco sales places selected in each district, and in 4 districts of Shanghai in June 2016 with 25 tobacco sales places selected in each district. Sales places included convenient stores, tobacco stores, supermarkets, and kiosks. In addition, public transport waiting points, public transport vehicles, hospitals, shopping centers, restaurants, bars, and Internet cafes were selected into survey in 2016. Results · In 2016, the proportion of tobacco sales places having tobacco advertisement was lower than that in 2015, but the proportion of tobacco promotion behaviors was higher. In 2016, 128 public places and streets were investigated and only 1 had outdoor tobacco advertisement. Conclusion · The new edition of Advertising Law has significant effect on controlling tobacco advertisement, but tobacco sales places still have tobacco advertisement with various disguised promotion forms. Tobacco sales places have diversified business styles and minors often enter these places. It is recommended that a clear definition of disguised forms of tobacco advertisement should be made, tobacco advertisement at tobacco sales places needs to be supervised, the release of tobacco advertisement should be strictly reviewed, and existing tobacco advertisement should be severely punished, so as to prevent minors from exposing to tobacco advertisement and information luring smoking.
10.Effect and mechanism of Gujingdan on acute renal injury induced by gentamicin

AIM:To study the protective effect and mechanism of Gujingdan(Radix Astragali,Fructus Alpiniae oxyphyllae,Radix et Rhizoma Salviae miltiorrhizae,Herba Epimedii,Semen Euryales,etc.) on the acute renal injury caused by Gentamicin in rats.METHODS:90 rats were randomly divided into normal control group,pathologic model group,preventive group which included high,middle and low dosages of Gujingdan(4 g/kg,2 g/kg,1 g/kg) and positive control group which was treated with Shenkangning(2 g/kg) and treated group which included the same groups as the preventive group.The preventive groups were respectively given Gujingdan and Shenkangning for two weeks and ahead one week.On the seventh day,except the normal group the others were continuously injected gentamicin by abdominal cavity(100 mg/kg/day) seven days.At the same time,the treated group were given Gujingdan and Shenkangning by ig for seven days.When the experiment was over,the content of ?-N-acetyglocosamidase(NAG),serum creatinine(SCR) and blood urea nitrogen(BUN) were examined,and the renal histological changes were observed.RESULTS:NAG,BUN,SCR of the groups which were treated with Gujingdan were remarkly lower than that of the pathologic model group;SOD、ATP were higher and MDA was lower than that of the model group.Histological changes showed that renal tubular epithelial cells of the groups treated with Gujingdan were similar to the changes of normal group and much better than that of positive group and pathologic model group.CONCLUSION:Gujingdan has the protective effect on acute renal injury caused by Gentamicin.The mechanism may be that Gujingdan has the function of inhibiting oxidation,protecting and stabilizing cell membrane.
