Chinese Journal of Ultrasonography 2001;10(5):267-269
Objective To investigate the relation between ovarian blood-flow resistance index (RI) measured with color Doppler ultrasound and the dosage of pituitary desensitization medication in controlled ovarian hyperstimulation cycle. Methods Thirty infertile females were randomized into group at routine dose of gonadotropin releasing hormone agonist (GnRHa) (group A) and group at reduced dose of GnRHa (group B).Ovarian arterial blood-flow RI was checked on cycle day 8 and on the day of human chorionic gonadotropin ( HCG) administration with transvaginal color Doppler ultrasound. Results The ovarian arterial blood-flow RI on cycle day 8,the total dosage and the duration of human manopausal gonadotropin (HMG) administration were significantly reduced in group B. In both groups, the ovarian arterial blood-flow RI was significantly decreased in the ovary with more dominant follicles (follicle diameter >10 mm) compared with the opposite one. Conclusions In controlled ovarian hyperstimulation cycle, compared with routine protocol of GnRHa administration, reduced dose of GnRHa could decrease the ovarian arterial blood-flow RI,the total dosage and the duration of HMG administration without reducing pregnancy rate.

Purpose:To clone and construct an eukaryotic expressive vector of ribonuclease inhibitor (RI) gene ,as well as to observe the effects of the transfected pLNCX-ri on the growth of C6 glioma cells. Methods:A segment of RI gene of 1.4 kb was obtained by Nde I/Xho digestion and cloned into pLNCX. Transfective agent and selective antibiotic were lipofect AMINE and G418 respectively. The expression of pLNCX-ri in C6 glioma cells was detected by Western blotting. And SD rats were inoculated by the transfected C6 glioma cells. Results:An eukaryotic expressive vector of RI gene was constructed successfully. RI content was remarkably higher in the transfected cells than that of in the untransfected cells. After SD rats were inoculated by the transfected C6 glioma cells, the tumorigenic time was prolonged, the tumor weight was reduced and the density of tumor vessels was notably decreased. Conclusions:These results indicated that RI gene powerfully inhibited the growth of C6 glioma cells via decreasing tumor vessels formation.

Basic & Clinical Medicine 2006;0(09):-
Objective To observe the effect of tyrosine kinase Src on activating Notch-1 protein in pancreatic cancer cell line-BxPC3. Methods Notch-1 expression and c-Src expression were inhibited by siRNA interference, and the activation of Src was inhibited by its inhibitor PP2. The growth rate of BxPC3 cells was measured by MTT. The change of Notch-1 NICD level and Src protein level was measured by Western blot. Results The growth of BxPC3 cells decreased after inhibiting Notch-1 expression, as well as the cells were treated with PP2. The level of Notch-1 NICD decreased both after inhibiting c-Src expression and after Src activation was inhibited by PP2. Conclusion In BxPC3 cells, tyrosine kinase Src promotes the level of Notch-1 NICD, which directly influences the cell growth.
