2.Different biological characteristics of high-and low-expression of ATP-binding cassette transporter in drug resistant human nasopharyngeal carcinoma cell

Objective To investigate the different biological characteristics of high-and low-expression of ATP-binding cassette transporter(ABCG2)in human multi-drug resistant nasopharyngeal carcinoma CNE2/DDP cell line(abbreviated as ABCG2High cell and ABCG2Low cell),and to analyze drug-resistant characteristics of the two cell lines.Methods ABCG2High and ABCG2Low cells were isolated by flow cytometry,while their cloning efficiency,cell cycle distribution,and the expression of ABCG2 before and after the 5th passage were examined as well.mRNA expressions of drug-resistant genes,such as ABCG2,Bcl-2,MDR1,MRP and MGMT,were detected by reversed transcriptive polymerase chain reaction(RT-PCR).MTT assay was utilized to detect the light absorbance(A)from day 1 to 14 to determine growth kinetics.Moreover,the drug sensitivity of the two cell lines to fluorouracil,cisplatin,vincristine,carboplatin,epirubicin,daunorubicin,paclitaxel and mitomycin were detected by MTT assay.Results The cloning efficiency of ABCG2High and ABCG2Low cells was 25.24%?1.18% and 16.28%?1.11%,respectively,on day 7.ABCG2High cells in S phase accounted for 41.5%,while the ABCG2Low cells in the G1/M0 phase went up to as high as 63.9%.The expression of ABCG2 in original ABCG2HighCNE2/DDP cells was 98%,while in ABCG2Low CNE2/DDP cells was 2%.However,at the 5th passage,the expression of ABCG2 decreased to 75% in the former cells and increased to 20% in the latter ones.The expression of ABCG2 mRNA in ABCG2High cells was obviously higher than that in ABCG2Low cells,while there was no difference in expression of other drug resistance genes between two cell lines.The growth velocity of ABCG2High cells exceeded that of ABCG2Low cells in early stage,but it slowed down 7 days later.MTT assay showed that IC50 of ABCG2High cells to 8 kinds of anti-tumor drugs were twofold higher than that of ABCG2Low cells,with statistically significant difference between them(P
3.Diagnosis and treatment of aggressive fibromatosis after thyroidectomy

Objective To investigate the diagnosis and treatment of aggressive fibromatosis after thyroidectomy.Methods One patient who were admitted into our hospital on January 16,2014 were discovered to have left cervical mass at the regular follow-up 8 months after opertaion.The ultrasonic examination showed that there were low echo area of 58 mm ×30 mm in the 3 ~5 muscular layer at the left neck. And it was diagnosed as aggressive fibromatosis through the puncture disease inspection.Whereafter,the patient were readmitted into hospital and recieved radical resection of the tumor on left side of the neck.The clinical,pathological and radiological data of the patient were collect-ed and analyzed so ao to discuss the diagnosis and treatment experience combined with related literatures.Results Postoperative pathologic examination confirmed that it was aggressive fibromatosis.The patient was actively cooperated with perioperative treatment,and the postoperative recovery was good with no local recurrence during the follow-up up to now.Conclusion Early diagnosis and surgical treatment are needed for patients with aggressive fibromatosis after thyroidectomy.
4.Clinical study of oxaliplatin combination with calcium folinate and flrorouracil in treatment of advanced esophageal carcinoma

Objective To evaluate the efficacy and toxicity of oxaliplatin in combination with calcium folinate and flrDrouracil in treatment of advanced esophageal carcinoma. Methods 61 patients with advanced esophageal cancer were divided into treatment group ( 30 cases) and control group (31 cases). Treatment group was given oxaliplatin combined with calcium folinate and flrorouracil; control group was given cisplatin and calcium folinate and flrorouracil. Results The overall response rate was 43.3% in the treatment group and 41.9% in control group(P>0.05).The median time to progression( TTP) was 8.1 months vs.7.9 months(P>0.05).Compared with control group,the treatment group, the side effects of myelosuppression, stomasitis and alopecia were not significant difference (P > 0. 05 ) , grade Ⅰ -Ⅳ nausea and vomiting( P = 0. 028 ) , diarrhea (P = 0. 039 ) and renal toxicity ( P = 0.044 ) were lower,while the peripheral nerve toxicity ( P = 0. 010) was higher. Conclusion The effect of oxaliplatin combined with calcium folinate and flrorouracil had satisfactory effect in the treatment of advanced esophageal carcinoma, and the poisonous side effect was low. It could be used as first-line chemotherapy regimen.
5.The inflammatory and immune mechanisms following cerebral ischemia

Hypoxia-ischemia induces an inflammatory and immune response in the nervous system that may be important for development of brain injury. Recent data implicate that cytokines, chemokines, adhesion molecules,are involved in the recruitment of inflammatory-immune cells. A cytokines which promote emigration of leukocytes from the vascular lumen into the injured brain tissue are produced at the site of incipient cerebral infarction. The expression of mRNA for alpha- and beta-chemokines preceded the appearance of immune cells suggesting that these molecules may have a role in the inflammatory response to insults. The humoral and cell-medated immunity under the conditions of focal brain ischemia was discussed. These results together indicate that cell-cell interaction by adhesion molecules and cytokines is an important component in the pathogenesis of ischemic cerebral diseases, especially at the acute phase.Recently accumulated data show that depleting the amount of circulating leukocytes or administering anti-inflammatory chemicals such as cytokine blocking agents, anti-adhesion molecule antibodies, and immunosuppressants effectively minimize the size of ischemia induced cerebral infarction.
6.Stimulating action of capitate phrynium stem and leaf extract on the rodent animals uterus

AIM: To study the exciting effects of ethanol extract of capitate phrynium (Phrynium Capitatum Willd) stem and leaf on the uterine smooth muscle of rat,mice,guinea pig and rabbit pretreated by diethylstilbestrol. METHODS: Conventional method was adopted to prepare animal uterine specimens both in vivo and in vitro. Different doses of the extract were given to the specimens and recorded their tensility, frequency and activity of contraction with the help of BL-420 biological experimental system. IC_(50) of the extract was calculated, with oxytocin as standard comparison. RESULTS: 95% ethanol extract and 50% ethanol extract of capitate phrynium and leaf had obviously stimulating action on the uterine smooth muscle of rat, mice, guinea pig and rabbit in a dose-dependent manner in vitro, whichinclude accelerating contraction frequency, raising tension (P<0.05, P<0.01). In addition, 50% ethanol extract also had a significant stimulating action on uterine smooth muscle of rabbit in vivo(P< 0.01). CONCLUSION: Ethanol extracts of capitate phrynium have an effect on rodent uterine. Smooth muscle indicates that clinical application of Chinese practitioner used to treat the dysfunctional uterine bleeding proves to be effective.

8.Calcium folinate, 5-fluorouracil bolus and continuous infusion combined with cisplatin in advanced esophageal carcinona

Purpose: The aim of this study is to assess the anti tumor efficacy and safety of high dose folinic acid plus 5 fluorouracil bolus and continuous infusion 48 hours combined with cisplatin in treating advanced esophageal carcinoma. Methods:Thirty patients with advanced esophageal carcinoma were treated with high dose folinic acid plus 5 fluorouracil bolus and continuous infusion 48 hours combined with cisplatin.Results: There were two complete responses, fourteen partial responses, thirteen no changes and one progressive disease in this series with total response rate of 53.33%. The main side effects include nausea and vomiting, mucositis, bone marrow suppression and alopecia. Other side effects were uncommon. All side effects were tolerable and mild except for nausea vomiting. Conclusions: High dose folinic acid plus 5 fluorouracil bolus and continuous infusion 48 hours combined with cisplatin may be a safe and effective therapy for the patients with advanced esophageal carcinoma.
9.Reflection on Opening of Section for Outpatients for Risk Assessment and Communication of Medication Use in Pregnancy by Pharmacists

Objective:To discuss the necessity of the opening of a pharmacist section for outpatients for risk assessment and com-munication of drugs in pregnancy by surveying the use situation and risk reasons of the drugs was in pregnancy. Methods:Referring to the risk classification of medicines in pregnancy formulated by FDA and integrating various factors in pregnant women, such as drug dosage, administration time, genetic factors, prenatal care and potential diseases et al, pharmacists established the section for outpa-tients for risk assessment and communication of drugs in pregnancy. Results:The visiting number of pregnant women was gradually in-creased after the establishment by reviewing the clinical data from the hospital information system. The new pharmacy service mode pro-vided by pharmacists for obstetrical patients was positively recognized by physicians and patients. Conclusion:The section for outpa-tients for risk assessment and communication of drugs in pregnancy should be established in order to promote the pharmaceutical knowl-edge in pregnancy and improve the medication safety.
10.Sunitinib malate-induced high expression of NKG2D ligands in nasopharyngeal carcinoma cell ABCG2~(high) CNE2/DDP

Objective:To investigate the inducing effects of sunitinib malate on expression of NKG2D ligands in nasopharyngeal carcinoma cell ABCG2high CNE2/DDP.Methods:ABCG2highCNE2/DDP cells and Allo-NK cells were isolated by magnetic activated cell sorting(MACS).Flow cytometry was used to evaluate the purity of isolated cells and the expression of NKG2D-ligands on target cells before and after incubation with sunitinib malate.Then the cytotoxic sensitivity of treated and un-treated ABCG2high CNE2/DDP cells to Allo-NK cells were measured by LDH releasing assay.Results:The positive rate of ABCG2 in ABCG2highCNE2/DDP cells was(91.40?2.32)%.More than 90% of isolated Allo-NK cells were proven to be CD3-CD16+CD56+ cells.The expression of MICA,MICB,ULBP1,ULBP2 and ULBP3 on ABCG2high CNE2/DDP cells incubated with sunitinib malate increased from(2.92?0.33)%,(4.27?0.33)%,(5.80?0.62)%,(11.10?3.15)%,and(7.75?1.14)% to(89.12?4.56)%,(66.10?2.22)%,(67.56?4.19)%,(69.37?8.83)%,and(63.28?3.31)%,respectively.At the E ∶T ratios of 10 ∶1 and 20 ∶1,the cytotoxic sensitivities of ABCG2high CNE2/DDP cells to Allo-NK cells increased from(15.32?13.86)% and(27.26?6.81)% to(41.12?4.12)% and(57.25?2.37)%,respectively,after treatment with sunitinib malate,with significantly difference found in the cytotoxic sensitivities of target cells in each group before and after sunitinib malate treatment(F=15.58,P=0.000).Conclusion:Sunitinib malate can up-regulate expression of NKG2D-ligands(MICA/B,ULBP1-3)in ABCG2high nasopharyngeal carcinoma cells,which results in higher cytotoxic sensitivity to Allo-NK cells.
