Objective:To study the protective effect of electro-acupuncture on glutamate(GLU),glycine(GLY),aspartate(ASP)and NF-?B expression in rat with cerebral ischemia reperfusion.Methods:The middle artery occlusion(MCAO)model was established and high pressure liquid chromatography(HPLC)fluorescence assay was used to detect contents of glutamate(GLU),glycine(GLY)and aspartate(ASP)in rat brain.The effect of electro-acupuncture on the nervous dysfunction score,the water content of cerebral constitution and the expression of NF-?B were observed.The group with non-acupoint group was as control.Results:The nervous dysfunction score,the water content of cerebral constitution,NF-?B,GLUand ASP in model group were higher than those of the sham-operated group.GLY levels had no obvious changes,there was no significant diference between the model group and the non-acupoint group.The nervous dysfunction score,the water content of cerebral constitution,the expression of NF-?B,GLU and ASP in electro-acupuncture group were lighter than that of the model group,GLY levels had no obvious changes.Conclusion:Electro-acupuncture could protect brain cell from damage after focal cerebral ischemia reperfusion injury by inhibiting the cerebral expression of NF-?B and excitatory amino acid.

Objective:Choosing efficient expression vector express anti HBsAg Fd and L chain in E.coli to produce anti HBsAg human Fab.Purificating inclusion bodies to denaturating and refolding the protein.Methods:Expression vector of PQE32 Fd、PQE32 L was constructed and transformed into E.coli strain M15,efficient expression clone was screened by SDS PAGE.Results:After induced by IPTG,M15 PQE32 Fd、M15 PQE32 L expressed insoluble recombinant protein in inclusion bodies.The Fd、L chain gene sequence conform with that reported in NCBI BLAST.Conclusion:The M15 PQE32 expression system is stable and efficient for expressing anti HBs human Fd、L chain.The denaturation and protein refolding of the anti HBsAg Fab expressed in inclusion bodies need to be studied further.

Objective To investigate effects of 1,25-Dihydroxyvitamin D3 on the cell cycle and apoptosis of K652 cells. Methods Vitamin D receptor (VDR) expression was identified by western blot analysis. Cell growth, apoptosis and cell cycle were evaluated by MTT assay, acridine orange/ethidium bromide (AO/EB) and flow cytometry. Results VDR was present in K562 cell nucleus. 10-8mol/L 1, 25-Dihydroxyvitamin D3 could markedly inhibit K562 cells growth and induced cell apoptosis. Most of cells were arrested in G2/M phase, the ratio of apoptosis increased from 4.1% in control group to 26.5 % in treatment group (P<0.01). Conclusion 1,25-Dihydrexyvitamin D3 exhibited obvious inhibitory effects on cell proliferation and cell cycle, and induced apoptosis in K562 cells.

OBJECTIVE: To explore the molecular basis for a pedigree affected with spondyloepiphyseal dysplasia congenita (SEDC). METHODS: The proband was subjected to whole exome sequencing. Suspected variant was verified by Sanger sequencing. RESULTS: All patients from the pedigree were found to carry a novel missense variant c.1394G>C (p.Gly465Ala) of the COL2A1 gene. The variant was not reported previously. Provean, Polyphen-2 and Mutation Taster software predicted that the variant is highly likely to be pathogenic. CONCLUSION The c.1394G>C (p.Gly465Ala) variant of the COL2A1 gene probably underlies the SEDC in this pedigree.

Objective:To investigate the effect of integrating nursing home service with rehabilitation therapy for persons with senile dementia.Methods:One hundred elderly persons with senile dementia were randomly divided into an observation group and a control group, each of 50. Both groups were given routine nursing care, while the observation group was additionally provided with rehabilitation training. Before and after the intervention, both groups were evaluated using the mini-mental state examination (MMSE), the Barthel index and QOL-AD.Results:After the intervention, the average MMSE, Barthel index and QOL-AD scores of both groups were significantly improved, with the average improvement in the observation group significantly greater.Conclusions:Rehabilitation training can effectively improve the cognitive functioning, performance in the activities of daily living and life quality of persons with senile dementia.
