Objective:To establish an HPLC method to determine the entrapment efficiency (EE) and drug loading (DL) of curcumin (CUR)and quercetin (QUE)loaded self-microemulsifying drug delivery system. Methods:A centrifugation method was used to isolate the free drug. The content of drug was determined by HPLC.The analytical column was a Purospher STAR LP C18 column (250 mm×4.6 mm,5 μm) and the column temperature was 30 ℃.The mobile phase was acetonitrile-4% acetic acid (50∶50) and the flow rate was 1.0 ml·min-1.The UV detection wavelength was set at 370 nm and the injection volume was 10 μl.Results:CUR and QUE were linear within the range of 10.728-96.552 μg·ml-1 (r=0.999 8) and 1.08-9.72 μg·ml-1 (r=0.999 9),respectively.The average recovery was 99.98%(RSD=1.46%,n=9) and 100.34%(RSD=1.06%,n=9),respectively.In CUR-QUE-SMEDDS,the EE of curcumin and quercetin was (95.97±0.50)% and (95.91±2.52)%,and the DL was (25.82±0.15) mg·g-1 and (1.80±0.05)mg·g-1,respectively.Conclusion:The method is accurate, rapid and simple, and suitable for the determination of DL and EE in CUR-QUE-SMEDDS.

The aim of the study was to prepare and evaluate the quality of negatively charged selfmicroemulsifying drug delivery system of curcumin (NC-CUR-SMEDDS).Based on the CUR-SMEDDS,the optimum amount of excipients was confirmed by the single-factor design experiment taking mean particle size,Zeta potential, drug entrapment efficiency and the drug loadings of curcumin as the evaluation indices for the preparation of NC-CUR-SMEDDS.The quality of NC-CUR-SMEDDS was evaluated by observing its appearance status, transmission electron microscope micrographs and determining particle diameter,Zeta electric potential, drug entrapment efficiency and drug loading. As a result,it was found that the Zeta potential reached -43.43 ± 0.29 mV when 4% docusate sodium was added. The appearance of NC-CUR-SMEDDS remained clarified and transparent, and the microemulsion droplets appeared spherical without aggregation with uniform particle size distribution. The mean particle size was 14.08 ± 0.082 nm,the drug loading of curcumin was 26.48 mg·g-t and the drug entrapment efficiency was 94.12%.It was concluded that NC-CUR-SMEDDS with high entrapment efficiency and uniform particle size distribution met the requirement of inflammatory target binding in the colon.

Objective To investigate the clinical characteristics of childhood anaphylactoid purpura. Methods Seven hundred and sixty children with anaphylactoid purpura were included in this retrospective study. The clinicopathological features of childhood anaphylactoid purpura were analyzed, including age at onset, gender, season at onset, clinical and pathological manifestations, and complications such as purpura nephritis. Results Childhood anaphylactoid purpura commonly affected preschool and school-age children, and usually occurred in winter or spring.Of these patients,265 (34.87%) had gastrointestinal symptoms,298 (39.21%) had joint involvement, and 223 (29.34%) had renal impairment.Purpura nephritis mainly manifested as haematuria, proteinuria and nephritic syndrome, and was diagnosed in 91.91% (91) of the patients receiving renal biopsy. The pathological grade of purpura nephritis varied from Ⅱ to Ⅲ in these patients. The distribution pattern of purpura was associated with complications. Conclusions Anaphylactoid purpura has age and season predilection.Purpura in both lower and upper extremities is likely to be complicated by gastrointestinal haemorrhage and joint involvement, and lower extremity purpura is more frequently to be complicated by nephritis than purpura in both lower and upper extremities. In general, childhood anaphylactoid purpura is a mild condition with a good prognosis.

Objective To compare the reliability and validity of 2 incontinence- associated dermatitis (IAD) risk assessment tools (IAD risk assessment scale and PAT) in patients with incontinence, and to search for the best risk assessment tool. Methods A total of 101 IAD cases were chosen from the Affiliated Hospital of Nantong University.Reliability was evaluated by the inter- rater reliability, internal consistency, item sensitivity analysis and retest reliability; validity was evaluated by the structure validity and predictive validity. Results Cronbach α was 0.313 (IAD risk assessment scale) and 0.421(PAT);Cronbach α coefficient was the highest after the items that Change sheets and mats and relevant factors were removed respectively in two tools, which were 0.431, 0.428; The correlation between the total scores were 0.711 (P<0.01)(IAD risk assessment scale)and 0.498 (PAT)(P<0.01);Area under curve were 0.661 (IAD risk assessment scale),0.864 (PAT); the best cut-off value to forecast a poor prognosis was 53.5 (IAD risk assessment scale),6.5 (PAT). Conclusions The reliability and validity of IAD risk assessment scale and PAT are low. The change of sheets and mats and Related influencing factors are needed to be adjusted and revised.

ObjectiveTo discuss the epidemiological characteristic s of road traffic injuries in the Qinghai-Tibet Plateau and explore effective pre ventive measures against traffic accidents in the plateau as well as correspondi ng emergency treatment methods.MethodsA retrospective statis tical analysis was carried out in 1 894 cases of traffic injuries with detailed data admitted in our hospital from January 1980 to December 2000. Resu ltsMost of the traffic accidents occurred on the Qinghai-Tibet road, accounting for 68.0% (1 288/1 894). Of the patients with serious traffic inju ries, 54.0% (1 022/1 894) were in need of hospitalization. The patients who first entered into the plateau accounted for 61.1% (1 158/ 1 894). Death occurre d in 108 cases accounting for 5.7%. The duration from injury to treatment was ve ry long, average over six hours. Furthermore, the patients received no any manag ement before treatment in the hospital. ConclusionsHypoxia i n the plateau, unclear road sign and fatigue driving are the main factors leadin g to traffic accidents. Lack of health care units, serious hypoxia, long deliver y time and carriers' deficiency in medical knowledge are the main causes for gr eat amount of casualties.

Objective:To explore the influence of childhood abuse experience on relapse tendency of compulsory drug abusers, and the intermediary role of resilience and depression between them. Methods:A total of 261 drug abusers were investigated with childhood trauma questionnaire(CTQ), the Connor-Davidson resilience scale (CD-RISC), self-rating depression scale (SDS) and relapse tendency psychological questionnaire. All data processing and analysis were carried out by SPSS 25.0 software.Correlation analysis, Bootstrap analysis and other methods were used to test the mediating effect. Results:The scores of childhood abuse treatment, resilience, depression and relapse tendency were 44.690±18.550, 60.210±22.790, 48.370±12.450, and 15.300±9.833 respectively.Childhood abuse was negatively correlated with resilience( r=-0.396, P<0.01), and positively correlated with depression and relapse tendency( r=0.584, 0.298, P<0.01).Resilience was negatively correlated with depression and relapse tendency( r=-0.558, -0.147, P<0.05).Depression had a significant positive correlation with relapse tendency( r=0.286, P<0.01).Childhood abuse could directly predict the tendency of relapse( β=0.202, P<0.01), and it could also affect the tendency of relapse through the mediation of depression( β=0.082, 95% CI=0.007-0.165) and the chain mediation of resilience and depression( β=0.029, 95% CI=0.002-0.064). Conclusion:The experience of childhood abuse leads to the decrease of resilience of strong abstinence personnel and increases the level of depression, which finally increases the tendency of relapse.
