With the economic growth and better standards of living,the prevalence of non-alcoholic fatty liver disease (NAFLD) is expected to increase dramatically worldwide.NAFLD is a common chronic inflammation disease.NF-κB is a transcription factor that plays crucial roles in inflammation,immunity,cell proliferation and apoptosis.It can facilitate the occurrence and development of NAFLD,and the underlying mechanisms are related to insulin resistance,oxidative stress,alteration of intestinal flora,activation ofrenin angiotensin system,etc.
2.The expression, synthesis and release of Urotensin Ⅱ in hypoxic pulmonary hypertension and its significance

Objective To study the release of synthetic gene expression induced by vasopressin Ⅱ (U Ⅱ ) in hypoxic pulmonary hypertension. Methods Rat model of HPH was establish. RIA was used to observe the different time points of hypoxia in plasma and the dynamic changes of U Ⅱ , ADM content in bronchoalveolar lavage fluid (BALF). The impact of the release of U Ⅱ , as well as the relation-ship among U Ⅱ , ADM and HPH were explored to reveal the role of U Ⅱ in the pathophysiology of HPH. Results Rat HPH model was successfully established. Hypoxia promoted the expression, synthesis and release of U Ⅱ in lung tissue. U Ⅱ involved in the pathogenesis of HPH. HPH took place in the development of U Ⅱand was positive correlated with ADM. Conclusion The two peptides have opposite physiological effects on blood vessel, which suggest that these two peptides play an important role in maintaining the balance between the pul-monary circulation and lung ventilation as well as the stability of pulmonary artery pressure.
3.Preliminary Analysis on Syndrome Differentiation and Treatment of Chronic Heart Failure Based on Qiangxin Prescription of Professor LUO Quan

The national famous TCM doctor, Professor LUO Quan considers that the key pathogeny of chronic heart failure (CHF) is deficiency of yang qi, blood blocking and liquid stagnation, and should be treated with the method of tonifying qi and warming yang, activating blood and expelling stasis and inducing diuresis to alleviate edema. Qiangxin Prescription was created based on the above treatments, with obvious efficacy. This article conducted a preliminary analysis on the essence of medication of Qiangxin Prescription, took some example of Professor LUO Quan's experiences in using this prescription, with a purpose to be helpful to the treatment of syndrome differentiation and treatment of CHF.
4.Correlation between depression-related personality dimensions and personality traits and its effects on depression

Objective To explore the correlation between the higher-order personality dimension(neuroticism) and the lower-order personality traits (alexithymia,dependence,self-criticism,perfectionism) in the sample of university students,and explore the effect of the higher-order personality dimension and lower-order personality traits to depression in the same sample.Methods A convenient sample of 563 university students from the two Universities College in Hunan province.These students were assessed with Center for Epidemiological Studies-Depression Scale (CES-D),Depressive Experiences Questionnaire (DEQ),The twenty-item Toronto Alexithymia Scales (TAS-20),Frost multidimensional perfectionism scale (FMPS) and neuroticism subscale in EPQ.Results (1) There were significant relationships between the total score of CES-D,each depressive symptoms and 10 personality factors,such as neuroticism,doubts about action and so on (The coefficients ranged from 0.105 to 0.569,P <0.05 or P<0.01).(2)Factor analysis and multiple linear regression on the neurotic showed that doubts about action,concerned over mistakes,difficulties identifying feelings (DIF),difficulties describing feelings (DDF),dependency,self-criticism and neuroticism belonged to the factor 1 (the factor load coefficients ranged from 0.574 to 0.775).(3) Neuroticism can explained 32.3 % variance of depression (R 2adj =0.323),after 6 personality factors entered the regression equation,such as difficulties describing feelings (DDF),parental criticism and so on,the explained variance of depression increased to 43.2% (R2adj =0.432).Conclusion There are overlapping and interaction between personality traits which include self-criticism,dependency,difficulties describing feelings(DDF),difficulties identifying feelings (DIF),concerned over the mistakes and neuroticism personality dimensions.The depressed affect are effectively predicted by neuroticism which is a effectively predict factor of depression,personality traits included self-criticism,dependency,alexithymia and malajustment perfectionism have a gain function of depression base on the Neuroticism.
5.Observation of the Additional Efficiency of Aldosterone Blockers During Angiotensin Ⅱ Receptor Blockers Treatment on Hypertensive Left Ventricle Hypertrophy

Objective To evaluate the additional efficiency of aldosterone blockers (AB) during angiotensin Ⅱ receptor blockers (ARB) treatment on hypertensive left ventricle hypertrophy in the elderly. Methods Forty-five cases of hypertensive left ventricular hypertrophy detected by Doppler ultrasonic cardiogram examination were enrolled,and were randomized to receive treatment with ARB plus AB (Group A) or ARB plus diuretics (Group B) for 48 weeks. The indicators of left ventricle hypertrophy were checked before and after the treatment. Results The both regimens could significantly decrease the blood pressure,and there was significant difference in the effects on lowering blood pressure between the regimens. Both regimens could alleviate left ventricle hypertrophy,but the regimen of ARB plus AB was superior to the regimen in Group B. Conclusion The addition of AB treatment during the ARB treatment was a good antihypertensive regimen for reducing left ventricle hypertrophy.
6.Serum concentrations of laminin and fibronectin in patients with acute coronary syndromes

Objective To study the serum iaminin (LN) and fibronectin (FN) changes in acute coronary syndromes (ACS), and explore the role of them in assessing the severity of ACS. Methods This study included 46 ACS patients [25 with acute myocardial infarction (AMI) and 21 with unstable angina (UA)], 51 stable angina (SA) patients and 47 people without CHD as controls. Serum levels of LN, FN, fibrinogen and blood fat were assessed. Coronary angiography were performed on 49 of them. Results The serum concentration of LN was lower in ACS patients [(85.20±27. 57)ng/mL], higher in SA patients [(116. 80 ± 28. 80)ng/mL] as compared to that in the control group [(100.06±29.96)ng/mL], with significant difference among the groups (P<0.05). No difference was found in FN among the three groups. However, the subgroup analysis in the group with ACS showed that the serum concentration of FN was significantly higher in UA patients [(229.60±121.39)μg/mL ], and lower in AMI patients [(108.31±47.12) μg/mL ]. The serum LN and FN concentration could respectively enter the logistic regression equations of ACS patients and US patients. Neither LN nor FN concentration was correlated with narrowing of coronary artery of angiography. Conclusion Serum LN and FN level may be a useful indicator for stability of atherosclerosis plaque in coronary arterial disease patients, but could not predict the extent of narrowing in coronary angiography.
7.Enhancement of cardiac function of chronic heart failure rats by marrow stromal cell-based sarcoplasmic reticulum Ca2+ adenosine triphosphatase gene therapy

BACKGROUND: There are still few effective methods to repair injured myocardium after myocardial failure and pathologically rebuild reverral myocardium. As a new therapy, normal myocytes and therapeutic gene to interfere injured myocardium have advantageous effects in improving heart function.OBJECTIVE: To observe the efficiency and stability of adenovirus-medicated gene transferred into different passages of bone marrow mesenchymal stem cell (MSC) and investigate the effect of MSC-based sarcoplasmic reticulum Ca2+ ATPase gene (SERCA2a) gene therapy for rats with chronic heart failure. To compare the effects of gene therapy, cell transplantation and MSC-based SERCA2a gene therapy for chronic heart failure. DESIGN: Randomized controlled study.SETTING: Department of Senile Angiocardiopathy, General Hospital of Chinese PLA; Department of Biochemistry, Beijing Medical University. MATERIALS: Male Sprague-Dawley (SD) rats with 4 weeks old, clean grade and weighing 45-50 g provided by the Animal Experimental Center, Peking Medical University were used as donators of bone marrow. Other female SD rats of 12 weeks old, clean grade and weighing 200-250 g were used as receptors of cell transplantation and gene therapy. Sry gene of Y chromosome in male rats was used to evaluate whether transplanted cells of donators lived in myocardium of receptor rats. Ad-SERCa2a and Ad-EGFP were constructed by Doctor Lu Xiao-chun; MSC in the 3rd and 8th generations was isolating cultured on its own. METHODS: The experiment was carried out in the Zhou CY Laboratory (BSL-2), Department of Biochemistry, Beijing Medical University from July 2004 to December 2005. Thirty female SD rats received ligation at the left coronary artery to make models with chronic cardiac failure following acute myocardial infarction. And then, 29 rats were randomly divided into four groups, including gene therapy group (n=7), MSC group (n=7), gene-modified MSC group (n=8) and control group (n=7). Rats in the four groups were given the interventions of SERCA2a gene, MSC transplantation, MSC+Ad/SERCa2a and empty adenoviral vector, respectively. MSCs were separated and cultured, and then Ad-SERCA2a-GFP was used to transfer MSC in the 3rd and 8th generations.MAIN OUTCOME MEASURES: Ad-SERCA2a-GFP transfection rate of MSC was measured by using flow cytometer. Before and at 14 and 21 days after treatment, cardiac function was evaluated by ultrasonic echocardiogram. Expression of cytokine Ⅷ was tested by immunohistochemical staining. SERCA2a gene and protein expression were evaluated by RT-PCR and Western blot respectively, as well as SERCA2a enzyme activity. RESULTS: ① Transfection rate: The infection efficiency of adenovirus-medicated gene into different passages of MSC was over 80%, and there was no difference between passage three (P3) MSC and P8 MSC (P > 0.05). ② Heart function: Left ventricle wall was thickened obviously in group MSC and group MSC+Ad/SERCa2a on the 21st day after treatment, while volume was shortened and gradually rounded. Compared to control group, ejection fraction (EF) and shortening fraction (FS) of group Ad-SERCa2a, group MSC and group MSC+Ad/SERCa2a were elevated significantly on the 14th day after therapy (P < 0.01). While the elevation values of EF and FS began to reduce in group Ad-SERCa2a on 14th day after therapy, it continued to increase in both group MSC and group MSC+Ad/SERCa2a (P < 0.01). Improvement rate of EF at 21 days after therapy (EF D21) increased in group MSC and group MSC+Ad/SERCa2a respectively, but decreased in group Ad-SERCa2a. Compared to group Ad-SERCa2a, peak systolic flow velocity of anterior wall and interventricular septum in group MSC+Ad/SERCa2a increased significantly on the 21st day after therapy, and peak diastolic flow velocity of anterior wall and interventricular septum elevated in group MSC+Ad/SERCa2a, too (P < 0.01). ③ SERCA2a gene, protein expression and enzyme activity in group MSC+Ad/SERCa2a were significantly stronger in group MSC and control group. Parts of MSC transplanted into scar zone expressed Ⅷ.CONCLUSION: ① MSC is an effective platform for the targeted delivery of therapeutic gene. It suggests that different passages of MSC from P3 MSC to P8 MSC are regarded as high-effectively gene vehicles. MSC-based SERCA2a gene therapy showed much strong and lasting beneficial effect on exhausted myocardium. ② Effect of MSC transplantation on improving heart function may be related to promoting vascular neogenesis.
8.The clinical application of VATS lung volume reduction surgery

Objective To evaluate the efficacy of lung volum ereduction surgery(LVRS) on severe chronic obstrutive pulmonary emphysema (COPE).Methods Fifteen patients with COPE with LVRS were retrospectively analyzed.The lung volume was reduced with GIA by a degree of 20%-30%.The activity and the lung function indexes of patients before LVRS,half a year and 1 year after LVRS were analyzed.Results Fifteen patients recovered and were discharged in 8-54 (14.7 ± 8.8) days after LVRS.The forced expiratory volume in one second (FEV1),total lung capacity (TLC),residual volume (RV),and 6-minute walk distance (6MWD) of patients with LVRS were significantly improved after operation (P < 0.05).Conclusion The lung volume reduction surgery provides a quite new and effective treatment for some cases of severe chronic obstructive pulmonary disease.
9.Expression of Cx43 in human lung adenocarcinoma and its significance

Objective To investigate the expression of connexin43 (Cx43) protein and mRNA in human adenocarcinoma of lung and its clinical significance.Methods One hundred and forty-eight human adenocarcinoma of lung tissues were detected by immunohistochemistry for Cx43 expression and fluorescence in situ hybridization for mRNA.The relationship between Cx43 expression and clinicopathologcal factors was analyzed.Results The positive rates of Cx43 expression in well/moderately differentiated and poorlydifferemiated/indififerentiated lung cancer tissues were 48.7 % (38/78) and 30.0 % (21/70),respectively (P < 0.05).The positive rates of Cx43 expression in negative lymph node metastasis and positive lymphnode metastasis lung cancer tissues were 56.3 % (18/32) and 34.5 % (40/116),respectively (P < 0.05).The positive rates of Cx43 expression in stage Ⅰ-Ⅱ and Ⅲ-Ⅳ lung cancer tissues were 51.5 % (49/95) and 34.0 %(18/53),respectively (P < 0.05).The positive rates of Cx43 mRNA expression in adenocarcinoma of lung adenocarcinoma and normal lung tissues were 32.4 % (48/148) and 60.0 % (12/20) (P < 0.05).Conclusion The expression of Cx43 is closely related with well differentiation,invasion,status of lymphnode,clinical stage and prognosis of human lung adenocarcinoma.It may participate in the development and progression of human lung adenocarcinoma,and may be used to predict the prognosis of human lung adenocarcinoma.
10.Effect evaluation of two kinds of wet dressing on peripheral phlebitis above stage Ⅱ

Objective To observe the effects of Suprasorb G and Suprasorb H on peripheral phlebitis above stage Ⅱ. Methods 64 patients with peripheral phlebitis above Stage Ⅱ were randomly divided into the observation group (36 patients) and the control group (28 patients). The observation group used Suprasorb G, and the control group used Suprasorb H. All cases were tested with the change of stage of peripheral phlebitis, the pain score, the red swelling of the skin and the palpable vascular cord. Results After treatment with Suprasorb H or Suprasorb G, there were significant differences in the change of stage of peripheral phlebitis, red swelling of the skin, pain score and the change of palpable vascular cord by self-contrast before and after treatment. Significant differences existed in change of stage of peripheral phlebitis, and red swelling of the skin and the change of palpable vascular cord at 48 and 72 hours, but no change was seen in pain score between 2 groups. Conclusions Suprasorb G and Suprasorb H beth show good effect in treatment of peripheral phlebitis, but Suprasorb G is better than Suprasorb H in improving the stage of peripheral phlebitis, also in reducing the area of red swelling of the skin and the palpable vascular cord with passage of time. But there is no difference in releasing of pain caused by peripheral phlebitis between them.
