Objective To study the chemical constituents of transformed products by Fusarium sacchari and to find the rare ginsenosides with anti-tumor effects from the leaves of Panax notoginseng. Methods The compound isolated was separated by repeated silica gel,Sephadex LH-20,and HPLC chromatography. The structure was identified by analysis of its spectral data.Results A dammarane-type triterpene, notoginsenoside-LZ,20(S)-3?-hydroxy-12?,23-epoxy-dammar-24-ene-20-O-?-D-xylopyranosyl (1→6)-?-D-glucopyranoside,was isolated from the transformed products.Conclusion Notoginsenoside-LZ is a new compound.

Objective To evaluate the application effect of mind map in admission guidance of patients receiving gamma knife treatment.Methods 153 cases of hospitalized patients admitted in our hospital were selected for this study,which had been divided according to the random number table method,the observation group(77 cases) was guided with the hospital mind map method,and the control group (76 cases) was given traditional oral hospital instruction,respectively.The database was established for the double entry,calibration and data cleaning,which were obtained from the result of gamma knife treatment of intracranial tumor disease related knowledge questionnaire,hospital satisfaction questionnaire and also patient adherence questionnaire assessment.Results The comparison of knowledge assessment questionnaire outcome,the admission guide satisfaction questionnaire assessment,the patients compliance questionnaire assessment outcome showed that the observation group was significantly different from the control group.Conclusions Application of mind map in admission guidance for hospitalized patients was a more efficient method than the traditional oral admission instruction,the method is simple,practical and effective,which is worthy of clinical practice.

Objective To study the triterpene saponins from Gynostemma pentaphyllum with antitumor activities.Methods The 75% EtOH extract of G.pentaphyllum was used for isolation by silica gel column chromatography and preparative HPLC.The structures of pure compounds isolated were identified by the spectral analysis and chemical evidence.Results Two compounds were isolated and identified as 23(S)-3β,20ξ,21ξ-trihydroxy-19-oxo-21,23-epoxydammar-24-ene 3-O-α-L-rhanmopyranosyl(1→2)-[β-D-xylopyranosyl(1→β)]-β-D-arabinopyranoside(1)and23(S)-21(R)-O-n-butyl-3β,20ξ-dihydroxy-21,23-epoxydammar-24-ene 3-O-α-L-rhamnopyranosyl(1→2)-[β-D-xylo-pyranosyl(1→3)]-β-D-arabinopyrannside(2).Conclusion Compound 2 is a new triterpene saponin with moderate antitumor activities against the HL-60,Colon205,and Du145 cell lines.

In recent years, many studies have demonstrated the potential of immune checkpoint inhibitors in the treatment of multiple types of cancers. Natural killer (NK) cells are an important part of the innate immune system and play an essential role in tumor immune surveillance. Their effects depend on their binding to inhibitory receptors, activating receptors, or both and the fact that they do not require major histocompatibility complex molecules to kill tumor cells. NK cells have shown great potential against both solid and hematologic tumors and have been increasingly identified as a promising therapeutic target for cancer immunotherapy. Some newly emerging checkpoint receptors and molecules have been revealed to mediate NK cell dysfunction in the tumor microenvironment, making them ideal targets for tumor immunotherapy. Thus, this paper will focus on the roles of these newly emerging immune checkpoint receptors in the regulation of NK cells and their potential application in tumor immunotherapy.

OBJECTIVE To study the analytic method for assaying of glutaral concentration by enzyme-labeled device at one time.METHODS Controlled solution of glutaral and sample solution were taken out and kept until the room temperature.Optical density value was detected at 450 nm.Content of sample was calculated by comparison with the known concentration of glutaral.RESULTS In the range of 0.75-2.50%,concentration of glutaral had linear relationship with the optical density value.Reproductive test:relative average error was within 2%,RSD≤2.51%;interference test:oxidation-resistant-sodium nitrite could make the result negative error.0.1% Sodium nitrite could make the test result decrease by 1.5% relatively.CONCLUSIONS The method is simple,fast and convenient,with accuracy which has met the requirement of hospital infection monitoring and acceptable interfering range made by additives,which is highly practical.

Objective To discuss the correlation between SIRT3 protein and clinicopathological parameters of gastric carcinoma.Methods Immunohistochemistry and Western blot were used to detect the expression of SIRT3 in the gastric carcinoma and normal gastric tissue.The correlation between the expression of SIRT3 and clinicopathological parameters was analyzed.Results The immunohistochemistry showed that the positive expression rate of SIRT3 protein in gastric carcinoma tissue (53.8 ％,43/80) was obviously lower than that in normal gastric tissue (86.0 ％,43/50),and the expression of SIRT3 protein showed close relationship with invasion depth,lymph node metastasis and TNM stage (P ＜ 0.05),rather than the age,gender,tumor size,or differentiation status (P ＞ 0.05).The Western blot showed that the expression rate of SIRT3 protein (SIRT3/β-actin) in gastric carcinoma tissue (0.655±0.317) was lower than that in normal gastric tissue (0.803±0.329) (P ＜ 0.05).Conclusion The expression of SIRT3 protein is lower in gastric cancer than that in normal gastric tissue,and relates to invasion depth,lymph node metastasis and TNM stage.SIRT3 may inhibit the occurrence and development of gastric cancer.

Objective To analyze the clinical features of,concurrence of allergic diseases in,as well as serum levels of total immunoglobulin E (IgE) and specific IgEs in,232 patients with alopecia areata (AA),and to estimate their relationship.Methods Serum samples were obtained from 232 patients with AA and 110 healthy controls from a health check-up center.Turbidimetric immunoassay and fluorescence-based enzyme-linked immunosorbent assay were performed to measure total IgE and specific IgEs to common antigens in these serum samples,respectively.Statistical analysis was carried out by t test,rank sum test,and chi-square test.Results The patients showed a male/female ratio of 127 ∶ 105 with an average age of (26.4 ± 13.8) years and clinical course of (25.3 ± 42.3) months.Among these patients,89 (38.4％) suffered from severe AA,and 67 (28.9％) reported at least one allergic disease.No significant difference was found in the positive rates of serum total IgE (30.2％ vs.21.8％,P ＞ 0.05),anti-Dermatophagoides pteronyssinus and-Cockroach specific IgEs or the serum levels of antiDermatophagoides pteronyssinus or-Cockroach IgE between the patients and healthy controls.The three most common specific IgEs were anti-Dermatophagoides pteronyssinus IgE (34.1％),anti-Dermatophagoides farinae IgE (31.9％) and anti-Cockroach IgE (22.5％) in patients with AA,whose serum levels were positively correlated with the level of serum total IgE separately.A significant increase was observed in the positive rates and serum levels of anti-Dermatophagoides pteronyssinus and-Cockroach specific IgEs in patients with severe and diffuse AA compared with the healthy controls and patients with mild AA,as well as in those of anti-Dermatophagoides pteronyssinus specific IgE in child patients with AA compared with the healthy controls and adult patients with AA (P ＜ 0.05 or 0.01).Conclusions Dust mite allergy may be not only a pathogenic factor for severe,diffuse and childhood AA,but also a risk factor for early onset and severe type of AA.

The target compounds were prepared from 5-aminobenzimidazolone by two steps reaction, and their AChE inhibitory activities were measured by Ellman method in vitro. The AChE inhibitory activity of compound 4d is the best of them, and its IC50 value is equal to 7.2 μmol·L(-1), which is better than that of rivastigmine; moreover the 4d had no inhibitory activities to BuChE. Therefore, the inhibitory activities of 5-aminobenzimidazolone derivatives to acetylcholinesterase are worth further researching.

Objective The study was to investigate the spectrum and incidence of the hot-spot deafness gene mutations from non-syndromic hearing loss(NSHL)in Guizhou province.Methods Genomic DNAs of 356 patients of NSHI in Guizhou were obtained from peripheral blood and a deafness gene test chip was used to screen nine hot spot mutation in the gene GJB2,SLC26A4,GJB3 and mitochondria 12SrRNA.Results Eighty-eight (24.72%)out of 356 patients were found carrying at least one pathogenic gene mutation.Among them,40 patients with the GJB2 mutations were found(11.24%),including 19 cases(5.34%)of homozygous matutions,5 cases(1.40%)of com-pound heterozygous mutations,and 15 cases(4.21%)of single heterozygous mutations.Twenty-nine patients with the SLC26A4 mutations were found (8.15%),including 9 cases(2.53%)of homozygous mutations and 19 cases (5.34%)of single heterozygous mutations.Homogenic mitochondrial 12SrRNA 1555A>G mutations were in 10 patients(2.81%)and 1494C>T mutations in 10 patients (0.56%),and heterogeneous mitochondrial 12SrRNA 1555A>G mutations in 7 patients(1.97%).GJB3 gene c.538C>T heterozygous mutations was found in 1 patients. Conclusion The mutations of GJB2 and SLC26A4 gene are two major pathogenic genes for patients with NSHL in Guizhou province.Our study provides a theoretical basis for the early diagnosis,genetic counseling and treatment of deafness.
