1.Determination of Atractylodin, Magnolol and Honokiol in Xiangshapingwei Pills by GC

Objective:To establish a method for the determination of atractylodin, magnolol and honokiol in Xiangshapingwei pills by GC. Methods:An HP-5 column (30 m × 0. 32 mm × 0. 25 μm) with an FID detector was used, and the flow rate of the carrier gas (N2) was 2. 0 ml·min-1. The column temperature was raised by program: the initial temperature was 100℃ and maintained for 7 min, and then raised to 250℃ at the rate of 20℃·min-1 and kept for 10min. The inlet temperature was 300℃ and the detector tem-perature was 300℃. The injection volume was 1 μl and the split ratio was 20∶ 1. Results:The linear range of atractylodin, magnolol and honokiol was 4. 264-85. 280 μg·ml-1(r=0. 999 7), 9. 856-197. 120 μg·ml-1(r=0. 999 5) and 10. 040-200. 800 μg·ml-1 (r=0. 999 6), respectively. The average recovery wsa 98. 2%, 98. 6% and 99. 3% with RSD of 1. 5%, 1. 5% and 1. 9%, respec-tively (n=6). Conclusion:The method is simple, accurate and reliable, which can be used for the quality control of Xiangshaping-wei pills.
2.Clinical observation on effect of Meituokang combined with western medicine in treating patients with angina pectoris

AIM: To observe the therapeutic effect of Meituokang(Floium Ginkgo Extract and Tertram Ethypyrazine injection) combined with western medicine in treating patients with angina pectoris. METHODS: 74 patients with angina pectoris were randomly divided into two groups,the control group(38 cases) were only treated with the routine Isosorbide Mononitrate; Metoprolol;Amlodipine;Aspirin enteric coated tablets and the treatment group(36 cases) were treated with Meituokang Injection qd,iv gtt besides the conventional treatment method of the control group.The course lasted 14 days. RESULTS: In comparison,the total efficacy rate of angina pectoris and the electrocardiogram(ECG) were 94.44% and 80.56% in the treatment group and 76.32% and 52.63% in the control group,respectively.There was significant difference between two groups by statistical analysis(P
3.Statistical analysis of visits by emergency outpatients to our hospital for one day

Objective To grasp the basic situation concerning visits by emergency outpatients to big urban hospitals before the implementation of reform in the medical insurance system in our city. Methods An investigation was made by means of profile survey questionnaires into the form of medical security of emergency outpatients, the sources of patients, referrals of patients, and the evaluation of doctors. Results Current emergency outpatient services in big urban hospitals have the following features: fairly great medical demands and attraction, widespread sources of patients, and pretty strong advantages in comprehensive clinical treatment. At the same time, before the implementation of reform in the basic medical insurance system, existing medical security setups, which are in the stage of transition, are characterized by a variety of types, a pretty narrow coverage in the provision of medical security and a rather high proportion of self-paying patients. Conclusion It is imperative to actively promote reform of the medical insurance system, formulate regional health plans that conform to the situation in our country, and expand the functions of community medical healthcare.
4.Regulation of miR-1 and miR-133 a on L-type calcium channel Cavβ2 and α1C subunits in rat cardiomyocyte hypertrophy

Objective To investigate the regulation of miR-1 and miR-133 a on L-type calcium channel β2 subunit ( Cavβ2 ) and α1C subunit during rat cardiomyocyte hypertrophy .Methods Cardiomyocyte hypertrophy was in-duced by isoproterenol (ISO, 10μmol/L).The targets of miR-1 and miR-133a were predicted by online database microCosm and Targetscan , respectively .The 3′untranslated region sequences of Cavβ2 andα1C were respectively cloned into reporter vector and then transiently transfected into HEK 293 cells.The luciferase activities of samples were measured for demonstrating the expression of luciferase reporter vector .The protein expression of Cavβ2 andα1C were evaluated by Western blot .The expression levels of Cavβ2 andα1C were inhibited by RNAi to determine theeffectsofCavβ2andα1Concardiomyocytehypertrophy.Results 1)Cavβ2wasoneofpotentialtargetsof miR-1,α1C was the one of potential targets of miR-133a.2) The luciferase activities of HEK293 cells with the plasmid containing widetype Cavβ2 3′UTR sequence or α1 C significantly decreased ( P <0.05 , P <0.01 ) . 3 ) Upregulation of the miR-1 and miR-133 a by miR-1 mimic and miR-133 a mimic transfection suppressed pro-tein expression of Cavβ2 and α1C, respectively(P<0.01, P<0.05).4)Downregulation of Cavβ2 andα1C by RNAi could markedly inhibit the increase of cell surface area ( P<0.01 ) , mRNA expression of ANP andβ-MHC (P<0.05).Conclusions Cavβ2 is the target gene of miR-1 and α1C is the target gene of miR-133a.miR-1 and miR-133a can negatively regulate the expression of L-type calcium channel Cavβ2 andα1C subunit, inhibi-ting cardiomyocyte hypertrophy.
5.ATRA combined with IFN? to induce differentiation and growth inhibition of NB cells.

Objective To study the effect of ATRA and IFN? on growth and differentiation of SH-SY5Y cell line and the possible molecular mechanisms.Methods The expression of TrkA mRNA was detected by RT-PCR.Trypan blue exclusion was used to detect the antiproliferative effect of ATRA,IFN?and ATRA +IFN? on SY5Y cells.Morphologic changes were observed using phase-contrast microscopy.Results TrkA mRNA was upregulated by ATRA and IFN?.ATRA and IFN? could induce morphological differentiation and inhibit growth of SY5Y cells.The combination of ATRA+IFN? enhanced the induced differentiation and growth inhibition of SY5Y cells.Conclusion Synergistic effects of ATRA and IFN? on growth inhibition and inducing differentiation in SY5Y cells are observed and this may result from upregulating the expression of TrkA mRNA.

Objective To study the role of different subtypes of PKC in the neurite outgrowth of primary cultured neurons from embryonic rat spinal cord. Methods We observed the expression and distribution of protein kinase C(?、?Ⅰ、?Ⅱ、? subtypes) by using immunohistrochemical technique in the neurite outgrowth of the primary cultured neurons from the embryonic rat spinal cord by means of computer image analysis system to measure the lengths of neurites and the areas of bodies. Results The areas of the cultured neuronal bodies increased and the neurites prolonged while the expression of the four subtypes of PKC gradually increased before 7div(day in vitro )during primary cultured neurons growth. The expression both in bodies and neurites of neurons decreased after 7div and the processes gradually shrinked and at the same time bubbles appeared inside the bodies. 9.Influence of compound Chinese traditional medicine on immunity of chicken inoculated by infectious bronchitis virus

A model of infectious bronchit was developed in SPF chickens by repeated intranasal infectious routes,and then the influence of compound Chinese traditional medicine on cellular immunity and humoral immunity during preventing and curing infectious bronchitis was studied by MTT,flow cytometry and serum neutralization test in tracheal organ culture.The results showed that compared with the infected group,the compound Chinese traditional medicine group could significantly increase the weight gain of chickens(P<0.05),promote the growth of immunity apparatus,enhance the T lymphocytes proliferate response of chickens and increase serum neutralization antibody titers of chickens significantly(P<0.05),and the ratio of CD4+/CD8+T lymphocytes was improved significantly(P<0.01).The aforementioned results indicated that the compound Chinese traditional medicine could reinforce immune function via preventing both cellular and humoral immunity from depression in the chickens with IBV.
