1.Study on the Quality Control Standard of Gehuang Huoxue Capsules

Objective To establish the quality control standards of Gehuang Huoxue Capsules. Methods TLC methods for identification of Radix Astragali and Radix Gentianae Macrophyllae were established. A simple HPLC method was established for the determination of Puerarin. Results The spots on TLC plates were clear without interference in the blank reference. The liner range of Puerarin was 0.020 79~0.665 2 mg/mL, r= 1.000 0. The average recovery of Puerarin was 100.1% and RSD=0.92%. Conclusion The method is simple, accurate, reproducible and can be used for the quality control standards of Gehuang Huoxue Capsules.
2.Observation effect of levocarnitine combined with shuxuetong in treatment of the patients with chronic renal failure

Objective To study the effects and safety of levocarnitine and shuxuetong in treatment of the patients with chronic renal failure.Methods 94 patients with chronic renal failure were selected as research object,and they were randomly divided into group A(levocarnitine and shuxuetong group) 47 cases and group B(shuxuetong group) 47 cases.Group A were treated with levocarnitine oral solution and shuxuetong,and group B were treated with shuxuetong,then the percentage of effectiveness,incidence of adverse reactions of group A and group B and serum BUN,Scr,Hcy were analyzed.Results The percentage of effectiveness of group A was higher than that of group B,but the incidence of adverse reactions of the two groups had no difference,and the serum BUN,Scr,Hcy of group A and group B before the treatment had no differences(P>0.05),those of group A and group B after treatment were compared there were significant differences(all P<0.05).Conclusion The effects of levocarnitine oral solution and shuxuetong in treatment of patients with chronic renal failure were better.
3.Influence of different dialysis frequence on infection of maintenance hemodialysis patients

Objective To study the influence of different dialysis frequence on infection and prognosis of maintenance hemedialysis patients. Methods Fifty-three maintenance hemodialysis patients with chronic kidney disease were divided into 3 groups. Groups A, B, C dialysed thrice per week (19 cases),twice per week (15 cases) and thrice per two weeks (19 cases), respectively. All the patients were dialysed for 1 year. The rate of infection, death number and death reason were recorded. Results All the cases were infected 62 times (17 times in group A, 18 times in group B and 27 times in group C). Pulmonary infection,blood vessels infection, urinary tract infection and digestive tract infection happened 35 times(56.45% ), 14 times (22.58%), 8 times (12.90%) and 5 times (8.06%) respectively. Eight cases died in three groups,and the total death rate was 15.09%, with 1 case in group A,1 case in group B and 6 cases in group C.There was significant difference in prognosis among three groups (χ2 =6.293,P <0.05). Conclusions Patients who dialysis less than twice per week have higher rate of infection and death. Dialysis program should be adjusted and dialysis frequence should be increased.

Many studies suggested that MUC1 mucin is involved in the mechanism of Helicobacter pylori( Hp)infection. It is important to understand the relationship between MUC1 mucin and Hp for preventing Hp infection and gastric cancer. This article reviewed the relationship between MUC1 mucin and Hp.
5.Therapeutic Observation of Acupuncture plus Speech Training and Psychological Intervention for Dysarthria in Hepatolenticular Degeneration

ObjectiveTo verify the therapeutic efficacy of acupuncture plus speech training and psychological intervention in treating dysarthria in hepatolenticular degeneration (Wilson’s disease).MethodSixty patients were randomized into a control group, a treatment group, and a combined group, 20 in each group. The three groups all received conventional treatments including removal of copper, liver protection, and brain protection, based on which, the control group also received speech training, the treatment group received acupuncture based on the treatments given to the control group, and the combined group received psychological intervention based on the treatments given to the treatment group.ResultAfter 2-month treatment, in comparing the Frenchay Dysarthria Assessment, the combined group showed a more significant improvement than the treatment group and control group (P<0.05); the total effective rate was 75% in the combined group, versus 25% in the treatment group and 10% in the control group, and the total effective rate of the combined group was significantly higher than that of both treatment group and control group (P<0.05).ConclusionAcupuncture plus speech training and psychological intervention can markedly improve the dysarthria symptoms of patients with Wilson’s disease, and recover their speech function and help them to go back to society.

Shiwei Dangguiyin(SWDGY)is mainly composed of Radix Angelicae Sinensis,Radix Adenophorae,Radix Notogenseng,Radix Bupleuri,etc. Oral administration of SWDGY could significantly inhibit the metatarsal swell- ing eaused by dimethylbenzene in rats,raise the pain threshold in hot-plate test and depress the torsive reaction caused by acetic acid in mice.In vitro SWDGY exerted bacteriostatic and bacteriocidal effects on Staphylococcus aureus,Bacil- lus pyocyaneus,Escherichia coli,Streptococcus A,B and C.It was shown that SWDGY possessed anti-inflammatory,analgesic and antiseptic effects in vitro.In mice LD_(50) of SWDGY by oral administration was more than 840g/kg.Affer cral adminstration in a daily dose of 189.Sg/kg continuously for one month in rats, no toxic reactions appeared,This dosage was 118.6 times as much as the clinical one.
7.Synthesis and anti-myocardial ischemia activities of novel Danshensu derivatives

Objective To design and synthesize novel Danshensu derivatives in order to improve their stability and cardio-protective effect against myocardial ischemia .Methods Novel Danshensu derivatives were synthesized chiefly viaesterification , methoxy phenol hydroxyl protection ,nitration and amine solution reaction .All the target compounds were evaluated their bio-logical effects on anti-myocardial ischemia with H9c2 cells in vitro .Results 15 compounds were synthesized and further con-firmed by 1 H NMR and MS .Pharmacological studies showed that few synthetic derivatives at 5μmol/L performed a higher bi-ological effect than Danshensu .Conclusion Most derivatives didn't show the best activity at the chosen concentration gradient that a further pharmacological trial need to be done .
8.Traditional Chinese medicine combined with radiotherapy for treatment of middle and advanced esophageal neoplasms: an analysis of prognostic factors

Objective To explore the prognostic factors of middle and advanced esophageal neoplasms treated by traditional Chinese medicine combined with radiotherapy.Methods A total of 462 esophageal neoplasms patients treated by traditional Chinese medicine combined with radiotherapy from March 2009 to March 2012 in Civil Administration Hospital of Hebei Province were retrospectively analyzed.The overall median survival time and the survival rate of 1,3 and 5 years were calculated by using life table.Log-rank test was used for single-factor analysis and Cox model was used for multiple-factor analysis.Results The median survival time of 462 esophageal neoplasm patients was 24.2 months (12.9-36.6 months).The survival rate of 1,3 and 5 years was 80.3 %,30.6 % and 10.5 % respectively.Single-factor analysis showed that the age (x2 =89.773,P =0.000),tumor diameter (x2 =102.373,P =0.000),clinical staging (x2 =128.903,P =0.000),concomitant with other diseases (x2 =65.366,P =0.000) and the short term effect (x2 =64.948,P =0.000) were associated with prognosis of middle and advanced esophageal neoplasms treated by traditional Chinese medicine combined with radiotherapy.Multiple-factor analysis showed that the age (RR =1.625,95 % CI 1.251-2.111,P=0.000),clinical staging (RR =2.437,95 % CI 1.874-3.168,P=0.000),concomitant with other diseases (RR =1.1.628,95 % CI 1.278-2.076,P =0.000) and the short term effect(RR =1.865,95 % CI 1.594-2.182,P =0.000) were the independent prognosis factors for middle and advanced esophageal neoplasms treated by traditional Chinese medicine combined with radiotherapy.Conclusion Elderly patients,esophageal neoplasms in stage Ⅲ,concomitant with other diseases and poor short-term effect could influence the prognosis of the patients with middle and advanced esophageal neoplasms treated by traditional Chinese medicine combined with radiotherapy.
9.Clinical and ASS1 gene variant analysis of three Chinese pedigrees affected with Citrullinemia type I.

OBJECTIVE:
To analyze the clinical and genetic characteristics of three Chinese pedigrees affected with Citrullinemia type I (CTLN1).
METHODS:
Three children diagnosed at the Children's Hospital Affiliated to Shandong University from 2017 to 2020 were selected as the study subjects. Genomic DNA was extracted from peripheral blood samples of the probands and their parents. Next generation sequencing (NGS) was carried out to detect pathological variants of the probands. Sanger sequencing was used for validating the candidate variant among the pedigrees.
RESULTS:
The probands have respectively carried compound heterozygous variants of c.207_209delGGA and c.1168G>A, c.349G>A and c.364-1G>A, c.470G>A and c.970G>A of the ASS1 gene, which were respectively inherited from their parents.
CONCLUSION
The newly discovered c.207_209delGGA and c.364-1G>A variants have enriched the mutational spectrum of the ASS1 gene. And the mutation spectrum of Chinese CTLN1 patients is heterogeneous.

10.Analysis of clinical features and ATRX gene variants in a Chinese pedigree affected with X-linked alpha thalassemia mental retardation (ATR-X) syndrome.

OBJECTIVE:
To explore the clinical characteristics and genetic basis of two brothers featuring X-linked alpha thalassemia mental retardation (ATR-X) syndrome.
METHODS:
An infant who had presented at the Qilu Children's Hospital in 2020 for unstable upright head and inability to roll over and his family were selected as the study subjects. The clinical features of the child and one of his brothers were summarized, and their genomic DNA was subjected to targeted capture and next generation sequencing (NGS).
RESULTS:
The brothers had presented with mental retardation and facial dysmorphisms. NGS revealed that they had both harbored a hemizygous c.5275C>A variant of the ATRX gene located on the X chromosome, which was inherited from their mother.
CONCLUSION
The siblings were diagnosed with ATR-X syndrome. The discovery of the c.5275C>A variant has enriched the mutational spectrum of the ATRX gene.
