1.Study on the relationship between sex hormone and lipid metabolism in overweight and obese boys aged 9 to 12 years

Objective:To explore the specific changes and correlations of sex hormones and lipid metabolism in overweight and obese boys aged 9-12 years.Methods:Seventy-two male subjects (9 to 12 years old) were divided into normal weight group ( n=42), overweight group ( n=15), and obese group ( n=15). Plasma sex hormone levels and lipid levels were detected by chemiluminescence immunoassay and ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC/QTOF/MS). All data were analyzed by multivariate statistics analysis. Results:(1) The serum concentration of sex hormone-binding globulin (SHBG) in obese group and overweight group was significantly lower than that in normal weight group, and the serum concentration of SHBG in obese group was significantly lower than that in overweight group ( P<0.05). The serum concentrations of dehydroepiandrosterone (DHEA) in overweight and obese group were significantly higher than those in normal weight group ( P<0.05). (2) There were 171 kinds of differential lipid compounds between normal weight group and overweight group, 218 species of differential lipid compounds between normal weight group and obese group, and 34 species of differential lipid compounds between overweight and obese group. Among them, 150 kinds of lipid compounds in normal weight group were significantly different from those in overweight and obese group. (3) SHGB was positively correlated with phosphatidyl cholines (PC) (20∶0/0∶0) and PC[18∶1(9E)/0∶0]( r=0.6, P<0.05), DHEA was positively correlated with diacylglycerol (DG)[17∶2(9Z, 12Z)/22∶0/0∶0]( r=0.5, P<0.05). Conclusions:9-12 years old overweight and obese boys had early growth and development, but sex hormone levels were not synchronized with growth and development. The serum DHEA level in overweight and obese boys was significantly increased, while the level of DG [17∶2 (9Z, 12Z)/22∶0/0∶0] was significantly increased, and the serum SHGB concentration was significantly decreased, while the vast majority of PC was significantly down-regulated. The mechanism of these changes needs to be further studied.
2.Removal of injected polyacrylamide hydrogel and repair of deformity after breast augmentation

Objective To explore surgical methods of removing polyacrylamide hydrogel (PAHG) and the right time of repairing the deformity of breast after removing PAHG.Methods We operated with endoscopy to remove PAHG through the lower mammary areolar incision.According to injection influence,we made the dicision whether to put the silicon gel prosthesis (hereafter referred to as prosthesis) simultaneously,and to fix the porsthesis,and to rebuild the imframammary fold of breast using biological repair membrane (hereafter referred to as membrane).Results In all 46 patients,39 patients' injection and their envelope were removed entirely.7 patients left part of the envelope because of its thin and wide characters.14 of them accepted prosthesis augmentation mammaplasty simultaneously and 5 of these accepted membrane repair.4 patients received prosthesis augmentation in stage Ⅱ.All patients' incision were primary healing.The incision scars were not obvious.1 patient with breast cancer suffered breast excision; 1 patient who received prosthesis and membrane simultaneously appeared prosthesis displacement after 3 months and fixed again.1 patient who received membrane appeared hydrops in residual cavity,and the membrane was removed finally.Conclusions This method with endoscopy through mammary areolae is necessary for cleaning PAHG entirely.We can use prosthesis to repair the deformity of breast after removing PAHG,and if necessary use membrane to fix the implant and rebuild the inframammary fold of the breast.
3.Prediction and prevention of stroke-associated pneumonia

Pneumonia is one of the most common medical complications after stroke.Stroke-associated pneumonia (SAP) can not only increase the length of hospital stay and medical cost of patients,but also an important risk factor for mortality and morbidity in patients with stroke.All these indicate that the importance of prediction and prevention of SAP.This article reviews the advances in research on the prediction and prevention of SAP.
4.Effects of artesunate on cell proliferation and apoptosis of human Tenon's capsule fibroblasts

Objective To investigate the effects of artesunate (Art) on cell proliferation and apoptosis of human Tenon's capsule fibroblasts (HTFs),and discuss the countermeasures of bleb scarfing in glaucoma.Methods In vitro,HTFs were cultivated and applicated by different concentrations (50 μg · mL-1,100 μg · mL-1,150 μg ·mL-1,200 μg · mL-1) of Art for 48 hours.The effect of Art on cell proliferation was assessed by MTT method.The rate of apoptosis induced by Art was determined by flow cytometry.Western Blot was performed to detect the relative expression levels of Bax and Bcl-2 after Art was treated.Results After treated with Art for 48 hours,compared with blank control group,Art (50 μg · mL-1,100 μg · mL-1,150 μg · mL-1,200 μg · mL-1) group exhibited notable anti-proliferative effect on HTFs with concentration-dependence (all P < 0.05).The results of flow cytometry showed that the apoptosis rates (8.80% ±0.88%,11.60% ±0.56%,16.30% ±1.03%,23.40% ±1.62%) of HTFs were significantly enhanced with the increase of Art concentration (all P < 0.05).The relative expression levels of Bax were obviously high with the increase of Art concentration,while Bcl-2 levels were significantly low with the increase of Art concentration (all P < 0.05).Conclusion Art can inhibit the proliferation and induce cell apoptosis of HTFs possibly by enhancing the expression of Bax and reducing the expression of Bcl2.Art may be a potential drug in preventing fibrous scar formation after glaucoma filtration surgery.
5.Regulatory role of BTLA signal in the initiation and early phase of T cell activation

Objective:To observe the expression of BTLA on T cells during activation and further analyze its inhibitory effects on T cell activation in different phases.Methods: T cells from PBMC were enriched by negative selection using magnetic beads.Expression of BTLA,CTLA-4 and PD-1 on freshly isolated human T cells and kinetics expression of BTLA,CILA-4 and PD-1 on CD3 mAb stimulated T cells were examined by flow cytometry.T cells were stimulated by anti-CD3 mAb combined with anti-CD28 mAb in the presence of anti-BTLA mAb 8H9,then T cell proliferation was tested by MTT assay in the different culture time.Immature DCs were generated from monocytes cultured in the medium containing GM-CSF and IL-4, and further driven to maturation by anti-CD40 mAb.Expression of HVEM on DCs was measured by flow cytometry.T cells were co-cultured with DCs in the presence of soluble 8H9 or anti-HVEM antibody to block HVEM-BTLA interaction,T cell proliferation was measured by MTT assay.Results:Freshly isolated T cells exhibited high levels of BTLA expression, but not CTLA-4 and PD-1.After T cell activation, BTLA expression decreased on first 2 days, with rapidly increasing to high levels.Unlike BTLA, expression of CTLA-4 and PD-1 was gradually increased during T cell activation.8H9 significantly inhibited the proliferation of T cell stimulated by CD3 mAb and CD28 mAb.8H9 could still exhibit inhibitory effect on T cell proliferation after 24 h or 48 h of preactivation by CD3 mAb plus CD28 mAb stimulation.HVEM was highly expressed on immature DCs, and down-regulated on mature DCs.Blockade of BTLA by soluble 8H9 or anti-HVEM antibody enhanced DC-mediated T cell proliferation within 48 h.Conclusion: BTLA signal enhances the threshold of T cell activation and plays importantly negative regulatory role in the initiation and early phase of T cell activation.

Objective To observe the protective effect of?-asarone,the active components from Rhizoma Acori Tarinowii,on human umbilical venous endothelial cell strain ECV304 injury induced by oxidized low-density lipoprotein(ox-LDL)and to investigate its influence on proliferation of vascular smooth muscle cells(VSMC). Methods The well-grown ECV304 cells were randomized into 5 groups:normal group,control group,high-, middle-and low-dose?-asarone groups.Except the normal group,ECV in the other groups were co-cultured with ox-LDL 25?g/mL,and the?-asarone groups were additionally with?-asarone 30,15 and 7.5?g/mL added respectively.Flow cytometry was used to detect the cell apoptotic rate and the mitochondrial membrane potential (MMP)of ECV304 as well as the proliferation index and the DNA content in different cell phases of VSMC. 7.Individualized education for 30 children with learning disability

ObjectiveTo study the method and effect of individualized education on children with learning disability (LD).Methods30 children during 7-10 year old in a local boarding elementary school were chosen and guided individually as the following: sensory integration dysfunction drill, subtle motion drill, life skills drill, cognitive capability drill, behavior therapy, game therapy, music therapy and parents education. One and a half year later, the index of Sensory Integration Rating Scales and study marks were determined and compared with that of the normal children.ResultsThere is not significant difference between the LD children and the normal ones in the sensory integration of behavior and the learning achievement.ConclusionIndividualized education is a valid approach improving the potentiality of the LD children\'s physiology and psychology.
8.Jiangtang decoction in combination with insulin in the treatmnet of type 1 diabetes mellitus in children

Objective To investigate the efficacy of the combination ofJiangtang decoction and insulin in the treatment of type 1 diabetes mellitus in children.Methods A total of 43 children with type 1 diabetes mellitus were enrolled and randomly allocated to either a insulin group (control group) and a insulin plusJiangtang decoction group (treatment group) using the table of random digits, with 23 patients in the control group and 20 in the treatment group. The patients in the control group were treated with insulin, and those in the treatment group were administrated withJiangtangdecoction on the basis of the treatment in the control group for 2 months. Body mass index (BMI), fasting plasma glucose (FPG), postprandial blood glucose (PBG) and hemoglobin A1c (HbA1c) were measured.Results The levels of FPG, PBG and HbA1c were significantly decreased in the treatment group (twere 3.376, 54.493, and 2.798, respectively; allP<0.01), and the PBG level was significantly decreased in the control group (t=29.714,P=0.000). The levels of FPG, PBG and HbA1c in the treatment group were significantly lower than those in the control group (twere 2.715, 2.191 and 5.627, respectively;P<0.05 orP<0.01). There was no difference in BMI between the treatment group and the control group (t=0.159,P=0.874). The proportion of patients with HbA1c <7.5% in the treatment group (60.0%, 12/20) was higher than that in the control group (26.1%, 6/23;χ2=5.055,P=0.025).Conclusion Combination ofJiangtang decoction and insulin can decrease the level of FPG, PBG and HbAlc, and its therapeutic effect is superior to insulin alone for type 1 diabetes in children.
9.Different beginning time of acid stimulation to reduce the acute damage of salivary glands after high-dose iodine-131 therapy for the post-surgery DTC patients

Objective To compare different beginning time of acid simulation to reduce the acute damage of salivary glands after high-dose of iodine-131 treatment for the post-surgery patient with differentiated thyroid cancer (DTC) and screen out the optimal beginning time of acid simulation .Methods Total 309 cases of post-surgery DTC patients accept high-does ioding-131 treatment (average dose of ioding-131 :4 .28 ± 0 .43 GBq) ,and all patients was divided into three group randomly ,the beginning time of acid stimulation(vitamin C :10 mg per time ,three time a day ,lemonade:50 mL per 2 h) for each group is 2 ,12 and 24 h .Then observe the incidence and time of acute damage of salivary glands for each group .At same time ,we analyses the relation between acute dam-age of salivary glands with sex and age .Results The incidence of acute damage of salivary glands is 13 .21% (2 h) ,24 .51% (12 h) , 26 .73% (24 h)respectively ,the incidence of 2 h is lower than those of 12 h and 24 h obviously(P<0 .05) ,and there is no difference between 12 h and 24 h(P>0 .05) .The occurrence time of acute damage of salivary glands for 84 .85% patients is between 10-24 h .And there is no difference of incidence of acute damage of salivary glands between different sex and age group .Conclusion 2 h maybe the optimal beginning time of acid simulation to reduce the acute damage salivary glands for the post-surgery DTC patients after high-dose iodine-131 treatment in this study .Sex and age are no influence to the occurrence of acute damage of salivary glands .
10.Establishment of a HL-60 cell line with stable RYBP silencing

Objective To establish stable HL-60 cell line with stable RYBP gene silencing using lentivirus-mediated RNA interference. Methods Five special shRNAs for RYBP gene were cloned to lentivirus vector.Recombinate lentivirus vectors were packed into lentivirus,which were used to infect HL-60 cells, and took empty vector and non-specific shRNA as control groups. Stable infected cells were selected with puromycin in 8 μg/ml concentration.The expression levels of RYBP were analyzed by Western blot,and confirmed the most effective RYBP shRNA.Then the level of mRNA was analyzed by real-time PCR.Results Stable infected cells were selected by puromycin successfully.Comparing to control groups,the expression of RYBP were reduced at different degrees (P < 0.01). And RYBP shRNA2 took the most silencing effect, the RYBP mRNA was decreased by more than 95 % (P < 0.05).Conclusion The shRNA2 targeting RYBP gene can effectively inhibit the expression of RYBP. HL-60 cell line with stable RYBP gene silencing were constructed successfully,which had provided experiment fundament for further studying the function of RYBP.
