1.Effects of MPP~+ on expression of inhibin,activin and its receptorⅡA and cell ability in PC12 cells of rats

Objective To observe the effects of MPP+ on the expression of inhibin, activin and its receptor ⅡA and cell ability in PC12 cells of rats.Methods The changes of expression of activin?A, activin?B, inhibin? and receptorⅡA mRNAs were assayed by RT-PCR, the change of cell viability was detect by tyrpan blue exclusion method in cultured PC12 cells at 3 h,6 h,12 h,24 h after MPP+ was added and compared with control groups.Results The expression of activin?A, activin?B and activin receptorⅡA mRNAs in PC12 cells were down-regulated significantly after MPP+ administrated at every time point while the expression of inhibin? mRNA remained unchanged. The cell viability decreased at the points of 12 h and 24 h after MPP+ administrated.Conclusion MPP+ may cause the injury of PC12 cells by downregulating the expression of activin and its receptor.
2.Study on Evaluation of the Scientific Research Performance of Provincial Key Disciplines in a Medical University

Objective This article was aimed to take exploratory evaluation on the scientific research performance of provincial key disciplines in a medical university,and to provide a scientific basis for the discipline evaluation related topics.Methods This paper took 29 provincial key disciplines in a medical university as the evaluation system.We calculated the scientific research performance of key disciplines.Data envelopment analysis (DEA) method was used to evaluate the scientific level of performance,while the use of TOPSIS and gray correlation analysis method to rank the efficiency of its research,and the results obtained by the three methods was used for statistical correlation analysis.Results According to the results calculated by DEA Method,there were 18 DEA efficient units and 11 non-DEA efficient units.While in this 11 non-DEA efficient units,8 of them needed to reduce re source inputs and the other 3 units needed to increase research outputs.Meanwhile there were significant positive correlations between evaluation results of TOPSIS Method andGrey Correlation Analysis Method with Data Envelopment Analysis Method,respectively [the correlation coefficient:rs 1 =0.797(P =0.000); rs 2=0.583(P =0.007); rs 3-0.536(P =0.003)].Conclusions Compared to TOPSIS Method and Grey Correlation Analysis Method,DEA Method not only could evaluate the ef fectiveness of discipline,but also provide evidence to improve the non-DEA efficient units.

Objective To apply the big data on medical equipment quality inspection to scientific management of medical equipment.Methods The big data on medical equipment metrological verification and quality control were analyzed,and medical equipment performance curve was drawn based on the function on data standard deviation and time.Results The big data provided support for medical equipment bidding and purchase,preventive maintenance and precision benefit analysis.Conclusion Medical equipment big data is of great significance to enhance medical support ability,and contributes to the management of medical equipment and strategic equipment.
4.The prospective trends and bias of clinical scientific research regarding recent grants in a hospital

Based on the investigation of a hospital's grants during 2004-2005 and the authorized during 2002-2005 (385 in total), we tried to find the trends and bias of clinical sciences at present. We analysed on the types of grants (clinical or basic), research objects, experimental methods, discipline involved, modes of cooperation, and background of applicants. A close connection between research and clinical trial, interdisciplinary study, patient-centered practice and international competitions are becoming more and more important. To meet the needs of clinical research, policies and devotions such as technical platforms are needed.
5.Expressions of HGF and its receptor c-met in non-small-cell lung cancer and its association with lymphangiogenesis

Objective To test the expression of HGF and its receptor c-met and to evaluate their relationship to lymphangiogenesis in NSCLC. Methods Immunohistochemical study was used to determine the lymphatic microvessel density (LMVD) and the expression of HGF and its receptor c-met in 113 patients with NSCLC and 20 normal lung tissue samples.Results The levels of expression of HGF (67.3 %) and c-met (74.3 %) in NSCLC tissues were significantly higher than that in normal lung tissues (20.0 %, 25.0 %respectively), (u=4.008, 4.342; both P <0.05).The expressions of HGF and c-met in NSCLC tissues had no correlation to age, gender, tumor size and differentiation grade.However, the expression of HGF and c-met was positively correlated to TNM stage and lymph node metastasis.In NSCLC samples, HGF and c-met were significantly associated with LMVD contrast to normal sample (HGF: 16.3051±5.3753 vs 10.9934±4.9668,t =4.58; c-met: 15.6692±5.5386 vs 11.3700±5.3875, t =3.97, both P < 0.05).Conclusion The high expression of HGF and its receptor c-met is closely correlated with LMVD and metastasis of lymph nodes, which indicates that HGF and c-met may play an important role in lymphangiogenesis and lymphatic metastasis in NSCLC.
6.Content Analysis and Comparative Study of Amino Acids in Ejiao from 18 Manufacturers

OBJECTIVE:To provide reference for quality evaluation of Ejiao. METHODS:The contents of 17 amino acids in 18 batches of Ejiao from 18 manufacturers were analyzed by automatic amino acid analyzer;the content data was processed by prin-cipal component analysis (PCA) and hierarchical clustering analysis (HCA),in order to understand the relationship among these amino acids and categorize the Ejiao products. RESULTS:Except for a batch of Ejiao,the other 17 batches contained 17 various amino acids,including 7 necessary amino acids(Thr,Val,Met,Ile,Leu,Phe and Lys)for human being,especially the contents of Gly and Pro were higher. The total content of amino acids was arranged from 66.1% to 82.0%,showing great difference. Four main components were extracted by PCA,the cumulative contribution of which was 89.578%,and accordingly it may be consid-ered that Pro,Ala,Glu,Asp,Leu,Ile and Thr were characteristic amino acids of Ejiao products. In hierarchical clustering analy-sis,3 tree figures of commercial Ejiao product were achieved and Euclidean distance was varied among 0-25. CONCLUSIONS:There is great difference in total control of amino acid in commercial Ejiao product;Ejiao products from various manufacturers dif-fer in their amino acid contents. Ejiao is rich in collagen,and therefore,using amino acid determination would be helpful to moni-tor its quality.

A possible association between the polymorphism of glucose transporter 9 (GLUT9) rs1 3137343 and hyperuricemia was investigated in Chinese male population and the calculated genotype frequencies and allelic frequencies by PCR method and direct sequencing were reported.Data showed that there was statistical difference in GLUT9 rs13137343 genotype frequencies between hyperuricemia cases and controls(x2 =7.024,P =0.030).
9.Correlation of Epstein-Barr virus infection with the expression of miR-101, EZH2, and COX-2 in gastric cancer

Objective:This study was conducted to investigate Epstein-Barr virus (EBV) infection in gastric cancer in Wuwei ar-ea of Gansu province (China) and the roles of miR-101, EZH2, and COX-2 in EBV-associated gastric carcinomas (EBVaGC). Meth-ods:Tissue microarray technique, immunohistochemistry, and in situ hybridization were performed to detect the expression of EBV-en-coded small RNAs (EBERs), miR-101, EZH2, and COX-2, in gastric cancer tissues (n=120) and the corresponding adjacent tissues (n=120). Results:The positive rate of EBV was 10.0% in 120 cases of gastric cancer tissues. EBVaGC was not significantly associated with lymph node metastasis and developed most often in the cardia and body (P<0.05). The differences in the positive rates of miR-101, EZH2, and COX-2 in 120 cases of gastric cancer tissues and corresponding adjacent tissues were significant (P<0.05). The differences in the positive rates of miR-101, EZH2, and COX-2 in 12 cases of EBVaGC and in 108 cases of EBV-negative gastric can-cer (EBVnGC) tissues were significant (P<0.05). The infection of EBV in gastric cancer tissues was positively related to miR-101 ex-pression. By contrast, miR-101 expression was negatively related to lymph node metastasis and expression of COX-2 and EZH2 in EB-VaGC tissues (P<0.05). Conclusion:EBV infection was related to gastric cancer in Wuwei area of Gansu province. EBVaGC and EB-VnGC have significant differences in lymph node metastasis and in the location of cancer. MiR-101, EZH2, and COX-2 were related to the development of EBVaGC.
10.Experimental study on bacteriostatic effect of recombinant human lactoferrin on Helicobacter pylori

Objective To evaluate the bacteriostatic effect of recombinant human lactoferrin(rhLF) on Helicobacter(H .) py‐lori and its influence on CagA ,Ure and gastric mucosal IL‐8 .Methods The minimum inhibitory concentration(MIC)and the influ‐ence of different drug concentrations on the proliferation of H .pylori were detected .The effects of rhLF on the mRNA and protein expressions of CagA and Ure in H .pylori were detected by RT‐PCR and Western blot ,respectively .The animal study :Balb/c mice were adopted and assigned randomly into four groups ,including the standard triple+rhLF(group A) ,rhLF(group B) ,standard tri‐ple(group C) and normal saline(group D) .The histopathological HE staining was used to observe the gastric inflammation and ELISA was used to detect the IL‐8 level of gastric tissue in each group .Results MIC was 0 .5 mg/mL ,moreover rhLF inhibited the bacterial growth and proliferation with a concentration‐dependent manner .rhLF could reduce the expression of H .pylori major viru‐lence factor CagA ,mRNA and protein of Ure .Comparing the group A with the group B ,C and D ,the gastric mucosal inflammation score and the IL‐8 levels of gastric tissue homogenates had statistically significant differences(P<0 .05) .Conclusion rhLF inhibits the growth and proliferation of H .pylori ,moreover inhibit the expression of major virulence factor CagA in H .pylori ,mRNA and protein of Ure in different degrees ,weakens its pathogenicity ,meanwhile reduces the IL‐8 level in mice gastric mucosa ,and allevi‐ates H .pylori related gastric mucosal inflammatory response .
