1.Distribution of T lymphocyte subsets in tumor microenvironment in papillary thyroid carcinoma

Objective To analyze the distribution of T lymphocyte subsets in tumor microenvironment of papillary thyroid carcinoma (PTC). Methods The levels of CD3+ CD4+ CD8- ,CD3+ CD4- CD8+ ,CD3+ CD4+ CD8+ ,CD3+ CD4- CD8- T cell and CD4+/CD8+ ra- tio in tumor microenvironment of 35 cases PTC, 25 cases benign thyroid lesion and 15 healthy eases were detected by flow cytometry. Results Local tumor tissue of 35 cases PTC and 25 cases benign thyroid lesion, and blood of 15 healthy cases were collected. The ratio of CD4+/ CD8+ in blood, tumor microenvironment of benign thyroid lesions and PTCs decreased significantly ( P<0.01). The levels of CD3+ CD4+ CD8+ cell in tumor microenvironment of PTC and benign thyroid lesion were higher than that in peripheral blood( P<0.01). But the differ- ence of the level of CD3+ CD4+ CD8+ cell between PTC and benign thyroid lesion was not significant ( P>0.05). The levels of CD3+ CD4- CD8- cell in tumor microenvironment of benign thyroid lesion were higher than those in PTC and peripheral blood(P<0.01). The difference of the level of CD3+ CD4- CD8- cell between PTC and peripheral blood was not significant (P>0.05 ). The differences of CD4+/CD8+ ratio and the level of each of T lymphocyte subset such as CD3+ CD4+ CD8-, CD3+ CD4- CD8+, CD3+ CD4+ CD8+, CD3+ CD4- CD8- cell between 15 PTC cases with lymph node metastasis and 20 PTC cases without metastasis were not significant (P>0.05). Conclusions CD8+ T cell is the main immune cell in tumor microenvironment in PTC. The local cell-mediated immunity of PTC is in the status of obviously suppressed. But the obvious increase of CD3+ CD4+ CD8+ and CD3+ CD4- CD8- T cells in tumor microenvironment in thyroid benign lesion may be beneficial for creating the location for immune responding against tumor. The distribution of T lymphocyte sub- sets in tumor microenvironment in PTC is not associated with lymph node metastasis.
2.Expression of human platelet factor 4 mRNA and its relation with angiogenesis and lymph node metastasis in papillary thyroid carcinoma

Objective To study the expression of human platelet factor 4 (hPF4)mRNA and its relationship with angiogenesis and lymph node metastasis in papillary throid carcinoma(PTC). Methods RT-PCR was used to detect the expression of hPF4 mRNA in 40 cases of PTC and 10 cases of normal tissue surrounding carcinoma. Immunohistochemical stain was performed to detect microvessel density(MVD) whose endothelia were marked by antibody CD105. Results The positive-expression rate of hPF4 mRNA was 80 % respectively in normal tissue surrounding carcinoma. The positive-expression rate of hPF4 was 47.5 % in PTC cases. 5.Albumin-based SPECT/Photoacoustic Dual-modality Nanoprobe in Liver Imaging

Purpose Liver fibrosis,liver cirrhosis and liver cancerseriously threaten human health.To detect liver diseases by specific nanoprobes,and to provide reference for the formulation of treatment strategies.Materials and Methods In this study,bovine serum albumin was used as a skeleton and modified with galactose groups for targeting asialoglycoproteinreceptors on liver.The albumin was labeled with radioactive iodine by chloramines T method,and indocyanine green molecules was encapsulated to form liver-targeting nanoparticles.Physical and chemical characterization (particle size and spectral characterization),bio-distribution,SPECT imaging and photoacoustic imaging were carried out respectively.Results The size of the nanoparticles was about 86.4 nm and there were two obvious absorption peaks at 705 nm and 780 nm.Bio-distribution showed that the radiolabeled nanoparticles had a high distribution in liver at 60 min [(55.52 ± 5.39)%ID/g],while after the receptor was inhibited,the uptake in liver was reduced to (37.01 ± 7.38)%ID/g,indicating a significant inhibitory effect (P<0.05);the uptake of this material at 240nm still remained (34.22±4.44)%ID/g,while in other organs,the detected uptake was quite low,with a statistically significant difference (P<0.05).The results of SPECT and photoacoustic imaging were consistent with the data of bio-distribution,and images showed highest signal on liver.Thus,the probe was suitable for liver imaging.Conclusion 131I-labeled albumin nanoparticle is an excellent liver-targeting dual modal imaging probe,which can be used for the detection of liver diseases.
