Journal of Chongqing Medical University 1986;0(03):-
Objective:To investigate the effect of fastigial nucleus stimulation(FNS) on preventing oxidative damage from myocardium after acute myocardial infarction in rats. Methods:Wistar rats were randomly divided into acute myocardial infarction(AMI),FNS before AMI model was made(FNS+AMI),bilateral fastigial nucleus lesion(FNL) before AMI(FNL+AMI) groups. At 24 hours after coronary ligation, blood samples and left ventricular myocardium were obtained for measurment of the activity of lactic dehydrogenase(LDH) and creatine phosphokinase(CK) in serum,as well as myocardial infarct size.At the same time, the malondialdehyde(MDA) level, total anti-oxidative capability (TAOC) and superoxide dismutase(SOD) activity in myocardium were measured too. Results:LDH,CK activities and MDA level in FNS+AMI group were significantly lower than that in AMI group( P 0.05).Conclusion:FNS can attenuate the damage of myocardium from infarct heart and reduce myocardial infarct size. The mechanism of the protective effect coming from FNS may be related to its antioxidative role.

Objective To investigate the changes of high mobility group box 1 (HMGB1),glutathione peroxidase(GPX3) in thyroid carcinoma and thier relationship with pathological characteristics. Methods The levels of HMGB1 and GPX3 were detected by using ELISA in 153 cases of thyroid cancer,120 cases of benign thyroid nodules and 120 healthy controls. The changes of HMGB1 and GPX3 in the patients with thyroid cancer were analyzed.Their application value in thyroid cancer was analyzed by using the receiver operating characteristic(ROC) curve. Results The plasma HMGB1 level and the HMGB1 positive expression rate in the thyroid cancer group were higher than those in the benign thyroid nodules group and control group(P<0.05),while the plasma GPX3 level and GPX3 positive expression rate in the thyroid cancer group were lower than those in the benign thyroid nodules group and control group(P<0.05).There were no significant differences in the levels of plasma HMGB1,GPX3 and the positive rates of HMGB1 and GPX3 between the thyroid benign nodule group and the healthy control group (P > 0.05).The plasma H MGB1 expression in thyroid cancer was closely related with the infiltration degree, clinical stage and lymph node metastasis(P< 0.05),and the GPX3 expression was related with the tumor stage and lymph node metastasis(P<0.05).The ROC curve showed that the sensitivity and specificity of HMGB1 and GPX3 for diagnosing thyroid cancer were 76.3% and 85.5%,70.8% and 82.2% respectively. Conclusion Plasma HMGB1 and GPX3 expression levels are closely associated with the occurrence and progression of thyroid cancer.

Objective To study the influences of impact of different loads to lamina on spinal cord with a closed spinal cord injury model and to explore the mechanisms of blood vessels in spinal cord injury. Methods 120 normal male SD rats were randomized into 4 groups, with 30 in each. Group A served as the control group which received no impact. In Groups B, C and D,impacts of 10, 15 and 20 N were given to lamina respectively. The blood flow in the spinal cord central gray matter and the dorsal white matter, the blood pressure and the heart rate were observed at different experimental time points. Results No obvious change was observed in the blood pressure and heart rate in every group at different time points. The spinal cord blood flow (SCBF) in Groups C and D varied in a regular pattern. Conclusions The impact load to lamina can affect the spinal cord mainly by altering the spinal cord microcirculation. The influence is the most obvious on the blood flow of microcirculation of the gray matter.

Objective Churg-Strauss syndrome (CSS) is a rare multisystem vasculitis of unknown aetiology. To describe the clinical features of the disease, the treatment and long-term follow-up. Methods Eight CSS patients were selected from rheumatology department of guangdong province peoples hospital between 2000-2005. Data were obtained retrospectively. The patients′ clinical manifestation and laboratory result were studied. Results All patients had asthma and hypereosinophilia. The lung, skin and peripheral nervous system were the most commonly involved organs. The majority of patient received intravenous injection pulses of methylprednisolone 0.5 g/d?3 d, followed by per oral prednison 1 mg?kg-1?d-1 and intravenous injection pulses of cyclophosphamide 750 mg/m2. The outcome and long-term follow-up were good. Conclusion Churg-Strauss syndrome is a systemic vasculitis occurring in patients with a history of asthma, corticosteroids and immunosuppressive treatment may received good outcome.

[Objective]To investigate the changes and correlation of bone collagen and biomechanical property in osteoporosis rats.[Method]Normal 30 nullimating female 7 month SD rats were divided randomly to two groups, each included 15.The sham group was unovariectomized, and the OVX group was ovariectomized. To execute the rats after 12 weeks, the author get the 5th lumbar vertebra, and then detected the biomechanical property and HYP.The author observed the collagen morphology of the 4th lumbar vertebra using the microscope after the masson triad color dyeing.[Result]Twelve weeks later,HYP in the sham group was(0.197?0.035)mg/g.The bone collagen was regular, compact, and closed succession.HYP in the OVX group was(0.182?0.022)mg/g.The bone collagen was incompact, thin, collapse and poor succession. There was significant change of the HYP in two groups(P

AIM: To construct a recombinant plasmid carrying enhanced green fluorescent protein and human vascular endothelial growth factor 121 gene and to detect its expression in rats mesenchymal stem cells (MSCs). METHODS: Human VEGF121 cDNA was amplified with polymerase chain reaction (PCR) from pCD/hVEGF121 and was inserted into the eukaryotic expression vector pEGFP-C1. The recombinant plasmid pEGFP/hVEGF121 was identified with PCR, double enzyme digestion and DNA sequencing. Then this recombinant plasmid was transfected into rat's MSCs with lipofectamine. The expression of EGFP and VEGF121 protein were detected with fluorescence microscope and immunocytochemical staining, respectively. RESULTS: The recombinant plasmid was confirmed with PCR, double enzyme digestion and DNA sequencing. The fluorescence microscope and immunocytochemical staining results showed that the EGFP and VEGF121 protein were expressed in MSCs 48h after transfection. CONCLUSIONS: The recombinant plasmid carrying enhanced green fluorescent protein and human vascular endothelial growth factor was successfully constructed and expressed positively in rat MSCs. It provides a good basis for further research on differentiation of MSC and VEGF gene therapy for ischemial cardiovascular disease.
