Objective:To investigate the effect of fastigial nucleus stimulation(FNS) on preventing oxidative damage from myocardium after acute myocardial infarction in rats.Methods:Wistar rats were randomly divided into acute myocardial infarction(AMI),FNS before AMI model was made(FNS+AMI),bilateral fastigial nucleus lesion(FNL) before AMI(FNL+AMI) groups.At 24 hours after coronary ligation,blood samples and left ventricular myocardium were obtained for measurment of the activity of lactic dehydrogenase(LDH) and creatine phosphokinase(CK) in serum,as well as myocardial infarct size.At the same time,the malondialdehyde(MDA) level,total anti-oxidative capability (TAOC) and superoxide dismutase(SOD) activity in myocardium were measured too.Results:LDH,CK activities and MDA level in FNS+AMI group were significantly lower than that in AMI group( P 0.05).Conclusion:FNS can attenuate the damage of myocardium from infarct heart and reduce myocardial infarct size.The mechanism of the protective effect coming from FNS may be related to its antioxidative role.
3.Effects of oxidized HDL on the levels of MCP-1, ICAM-1 and free calcium in cultured human umbilical venous endothelial cells

AIM: To study the effects of oxidized high-density lipoprotein (oxHDL) on the expression of monocyte chemoattractant protein-1(MCP-1) and intercellular adhesion molecule-1(ICAM-1) and intracellular free calcium concentration ([Ca2+]i) level in cultured human umbilical venous endothelial cells(HUVECs). METHODS: The MCP-1 protein content in the medium of conditioned HUVEC was measured by ELISA, and the ICAM-1 on HUVECs was detected by indirect immunofluorescence, and [Ca2+]i was determined by Fluo-3/AM, the injury of cells was observed by scanning electron microscopy (SEM).RESULTS: oxHDL could induce the expression of MCP-1 and ICAM-1 in HUVECs. In oxHDL group (HUVECs were incubated with 100 mg protein/L oxHDL for 24 h), the levels of MCP-1, ICAM-1 and [Ca2+]i increased by 160%, 60% and 70% respectively compared with the control group (P<0.01). When HUVECs were incubated with 300 mg protein/L oxHDL for 24 h, cells were injured obviously. CONCLUSION: By inducing the expression of ICAM-1 and MCP-1 in endothelial cells, oxHDL may promote monocyte-endothelium adhesion and monocyte migration to intima, it may promote atherosclerosis as oxidized low-density lipoprotein (oxLDL).

5.EFFECTS OF DIFFERENT EDIBLE OIL ON LIPID AND LIPID PEROXIDATION IN RATS

Effects of different edible oils on lipid and lipld peroxidation and their correlation were studied in rats. The results showed: 1.The tea-seed oil and soybean oil exhibited a good potency of reducing serum TC and TG comparing with lard did. 2. The plasma LCAT activity in the lard feeding group (57.07 nmol/ml.h) was lower than that of groups consuming four different vegetable oils which produced similar LCAT activities (80.26-94.03 nmol/ml-h). 3. Lard increased liver lipid and promoted TG sediment in aorta, but lowered brain lipid level. 4. Of lipid peroxidation in serum, liver, heart and kidney, the rats fed lard exhibited a highest level, and tea-seed oil fed group was lower relatively soybean oil promoted tissue lipid peroxidation while rape-seed oil and Salad oil suppressed it. 5. After experiment, the SOD activity of erythrocyte increased in rats fed oils riched in monounsaturated fatty acid, and decreased in rats fed soybean oil and lard. 6. CONCLUSIONS: In late p hase of preconditioning ,the p38MAPK activity and expression of iNOS and COX 2 increase significantly in rat cardiomyocytes, activited p38MAPK mediates iNOS ,then promotes COX 2 expression.

8.Effect of activated nuclear receptors PPAR?,LXR?,RXR? on ABC1 and CD36 mRNA expression in THP-1 cell

AIM: To study the discrepancy and it's significance of ATP binding cassette transporter(ABC1) and CD36 mRNA expression while activate nuclear receptor RRAR?,LXR?,RXR? in THP-1 cells. METHODS: Cultured THP-1 cells were induced to become macrophage by PMA at concentration of 40 ?g/L for 48 hours. Troglitazone, 22(R)-hydrocholesterol and 9-cis-retinoid acid(9-CRA),activators for PPAR?,LXR? and RXR? respectively, were incubated with the macrophage for 24 hours. Total RNA of these cells were abstracted and reverse transcriptional polymerase chain reaction were performed to inspect the expression of ABC1 and CD36 mRNA in the cells. RESULTS: Expression of ABC1 mRNA increased in the presence of troglitazone, 22(R)-HC and 9-CRA. CD36 mRNA also increased while PPAR? activated. Activation of LXR? showed almost no effect on CD36 mRNA expression, and expression of CD36 mRNA was decreased by 9-CRA.CONCLUSION: The discrepancies existing in ABC1 and CD36 mRNA expression while activate nuclear receptors PPAR?,LXR?,RXR? in THP-1 cells implyed that LXR?,RXR? were better targets to deal with to improve cholesterol reverse transportation.
9.An experimental research of exponential curve electric stimulus promoting the restoration of peripheral nerve injury

Objective To find a new way to promote the regeneration of injured peripheral nerve after surgical repair, and to interpret its application and advantages. Methods Eighty adult male Wistar rats with sciatic transplantation were divided into group A(control group) and group B(exponential curve electric stimulus group). The effect of pace, hair and unfold claw reflex were observed, and the sciatic nerve function index(SFI), nerve conduction velocity, wet weight of triceps surae were measured at different time. The curative effects of two groups were compared statistically. Results The group of exponential curve electric stimulus had positive effect on the recovery of nerve conduction velocity, the slower of the denervated disuse of triceps surae, the recovery of SFI, the acceleration of distal nerve degeneration, at last promote the injured nerve regeneration and the good recovery of function. The group of exponential curve electric stimulus is of significant difference statistically with the control group. Conclusion Exponential curve electric stimulus can promote the injured nerve regeneration.
10.An experimental study of the influence of impact to lamina on spinal cord blood flow

Objective To study the influences of impact of different loads to lamina on spinal cord with a closed spinal cord injury model and to explore the mechanisms of blood vessels in spinal cord injury. Methods 120 normal male SD rats were randomized into 4 groups, with 30 in each. Group A served as the control group which received no impact. In Groups B, C and D,impacts of 10, 15 and 20 N were given to lamina respectively. The blood flow in the spinal cord central gray matter and the dorsal white matter, the blood pressure and the heart rate were observed at different experimental time points. Results No obvious change was observed in the blood pressure and heart rate in every group at different time points. The spinal cord blood flow (SCBF) in Groups C and D varied in a regular pattern. Conclusions The impact load to lamina can affect the spinal cord mainly by altering the spinal cord microcirculation. The influence is the most obvious on the blood flow of microcirculation of the gray matter.
