1.The identiifcation analysis of rhinoceros horns products based on DNA barcoding 12s rRNA

Objective Through the DNA barcoding 12s rRNA sequences alignment and analysis of several rhinoceros horns products involved in cases to analysis the application feasibility of 12s rRNA in the rhinoceros horns products’ species identification. Methods Use rhinoceros horns products in 3 cases as materials, total DNA were extracted with improved method, PCR ampliifcation the DNA barcoding. Results The alignment and analysis of sequences show that 12s rRNA could identify rhinoceros horns products at the species level. Conclusion The DNA barcoding 12s rRNA could be used as a new way to identify the rhinoceros horns products which can’t be identiifed with morphological characteristics, provide a reliable basis for the qualitative and sentencing of cases.
2.The effectiveness of XueBiJing injection in therapy of sepsis: a multicenter clinical study

Objective To evaluate the effectiveness and safety of Xuebijing (XBJ) injection in therapy of sepsis.Methods It is a prospective,randomized controlled trial conducted in multicenter.Totally 731 patients with sepsis were enrolled from January to December 2011 at 37 centers and randomly (random number) separated into the XBJ group (n =392) and the control group (n =339).Basic therapy was adoptd in the both groups,but XBJ injection was additionally used in the XBJ group.The effectiveness of XBJ injection was evaluated by comparing the differences of MEDS score,APACHE Ⅱ score,DIC score,SOFA score,coagulation parameters,duration of mechanical ventilation and antibiotic therapy between the two groups.The influence on the outcome of XBJ injection was assessed by mortalities of 28-day and 90-day.The side-effects of XBJ injection were recorded.Results The score of each system and the abnormal rates of coagulation parameters were significantly higher in the XBJ group than in the control group at admission (P < 0.05).After combined treatment of XBJ injection and basic therapy,the score values of the XBJ group decreased distinctly,and the differences of pre-therapy and post-therapy in the two groups had statistical significance (P<0.01).The abnormal rates of coagulation parameters in the XBJ group decreased after therapy.The 90-day mortality of the XBJ group was lower than the control group (P < 0.05).No severe side-effects were found during study.Conclusions This study proved that the severity of sepsis can be alleviated by XBJ injection.Meanwhile,the organ function and the coagulation status can be protected,and then the prognosis can be improved.The safety of XBJ injection was good.
3.The independent predictors of 3-day mortality in patients with systemic inflammatory response syndrome in emergency department

Objective To study the independent predictors of 3-day mortality of patients with systemic in-flammatory response syndrome(SIRS) in emergency department. Method From Dec 2006 to Sep 2007, 640 pa-tients with SIRS admitted to emergency department were enrolled. Those died within one hour were excluded. Ev-ery patient was examined on plasma B-type natriuretie peptide (BNP) level and serum cardiac Troponin Ⅰ (cTNI) level. Early warning score (EWS) was calculated. Rank test was used to compare the difference of BNP,cTNI and EWS between survivors and non-survivors. Logistic regression analysis was used to determine the independent pre-dictors for death and receiver operator characteristic curve was used to determine the cutoff values. Results The mean levels of age, BNP, cTNI and EWS of patients died in 3 days were 73.5(65~80),468.5 pg/mL(161.5~1862.5 pg/mL),0.37 ng/mL(0.07~2.61 ng/mL) and 6(5~9), respectively. In survivors, the mean levels of age, BNP, cTNI and EWS were 71(60~77), 239 pg/mL(56.3~783.8 pg/mL), 0.07 ng/mL(0.03~0.26 ng/mL)and 4(3~6), respectively. There were significant difference in age, BNP, cTNI and EWS between two groups (P < 0.05). Age ≥ 76, BNP ≥ 115 pg/mL, cTNI ≥ 0.135 ng/mL and EWS ≥ 6 were independent predic-tors of death. Conclusions Levels of BNP,cTNI and EWS were predictors of death in SIRS patients during emer-gency department stay.
4.Expression and significance of EMMPRIN and VEGF-C of breast carcinoma

Objective To study the correlation of EMMPRIN and VEGF-C expression with invasion and metastasis of breast carcinoma. Method The expression of EMMPRIN and VEGF-C was detected by immunohistochemical technique in 21 cases hyperplasia of mammary glands, 10 cases of in-tubule carcinoma and 68 cases invasive carcinoma. Results The positive rate of EMMPRIN expression in hyperplasia of mammary glands, in-tubule carcinoma and invasive carcinoma were 4. 76% (1/21), 40. 00% (4/10)73.53% (50/68), respectively;Significant difference was found among three groups ( P < 0.01 ). The positive rate of VEGF-C expression in hyperplasia, in-tubule carcinoma and invasive carcinoma were14. 29% (3/21) ,60. 00 (6/10) ,77.94% (53/68), respectively. The positive rates of VEGF-C expression in situ and invasive ductal carcinoma were remarkably higher than that of hyperplasia of mammary glands ( P <0. 01 ). The expression of EMMPRIN significantly correlated with the expression of VEGF-C in breast tubule carcinoma ( rs =0. 390, P <0. 01 ), Lymph nod metastasis and the number axillary Lymph nod metastasis were related to EMMPRIN and VEGF-C ( P < 0. 01 ). Conclusion The over-expression of EMMPRIN and VEGF-C were important elements to promote the invasiveness and metastasis of breast carcinoma. Synergistic effect between EMMPRIN and VEGF-C may take place in lymph node metastasis of breast carcinoma.
5.Experience of TAN Qing-gang in Differentiation and Treatment for Postoperative Breast Cancer

TCM plays a more and more important role in the treatment of breast cancer today. Many veteran TCM doctors hold unique opinions about it, especially for postoperative complications. Doctor TAN Qing-gang is one of the most famous and experienced TCM doctors in the field of TCM in Enshi, Hubei Province. He is expert in the diagnosis and treatment on miscellaneous diseases of TCM. This article summarized the experience of Doctor TAN in treatment for postoperative breast cancer.
7.Establishment and biological characteristics of six human renal cell carcinoma cell lines

Objective To establish human renal cell carcinoma (RCC) cell lines,and to investigate the cell phenotypes and expression of tumor associated antigens. Methods Fresh tumor tissues from pathologically proven human RCCs were primary cultured and passed generation to generation until a stably grow in vitro.The cell cloning form,chromosome and graft into nude mice in vivo were examined for cell lines.Immunofluorescent stain and flow cytometric analysis were carried out for cell's phenotype,RT PCR examination for MN/CA9 gene expression. Results Six human RCC cell lines have been established,including four of the cell lines derived from clear cells,one from mixed clear granular cells and one from papillary cells.All the cell lines showed the characteristics of malignant cells.All the lines highly expressed HLA ClassⅠ and HER2/neu.Three lines weakly expressed HLA ClassⅡ and one cell line highly expressed CD86 but all the lines did not express CD80.RT PCR showed that three cell lines have the expression of MN/CA9 gene. Conclusions These newly established RCC cell lines would provide a useful in vitro model for studies related to biological characteristics,tumor associated antigen,immunogenity and immunotherapy of human RCC.
8.c-erbB-2 protein of human renal cell carcinoma can be recognized by cytotoxic T lymphocytes

ObjectiveTo provide experimental proof for the specific immunotherapy in patients with renal cell carcinoma (RCC).MethodsThe expression of c-erbB-2 and HLA-A2 molecules in RCC were examined by flow cytometery and HLA-A2 cDNA RT-PCR.Tumor lysatic antigens (Tuly) was used to load DCs (DC-Tuly) to induce the generation of c-erbB-2 specific CTLs derived from peripheral blood mononuclear cells of RCC patients in vitro and CTLs was used to kill RCC cells which express or not express c-erbB-2 and HLA-A2.The cytotoxic activity of induced CTLs was further studied by antibodies (anti-c-erbB-2 and anti-CD 8) blocking assay.Resultsc-erbB-2 protein is tumor associated antigens for RCC.DC-Tuly induced specific CTLs highly killed c-erbB-2+ HLA-A2+ autologous and allogenic RCC,but non-specific ones lowly killed c-erbB-2- HLA-A2+ or c-erbB-2+ HLA-A2-RCC cells. The cytotoxic activity against tumor cells was blocked by anti-c-erbB-2 and anti-CD 8 monoclonal antibodies.ConclusionsThese results suggest that this specific CTLs adoptive immunotherapy for RCC patients with c-erbB-2+ HLA-A2+ could be a novel approach for clinical use.
9.Inhibitory effect of different target-side antisense oligonucleotides on bFGF expression in SWO-38 cells

AIM: Three different antisense oligonucleotides complementary to basic fibroblast growth factor (bFGF) mRNA were compared in inhibitory effect on gene targeted expression. METHODS: After transfecting bFGF antisense oligonucleotides (asODN) into SWO-38 cells by lipofectin,the proliferation of cells was identified by MTT method, apoptosis was examined by flow cytometric cell cycle analysis and the expression levers of bFGF were detected by Western-blotting. RESULTS: There were 49%,33%,51% inhibition of cell growth and 35%, 27%, 18% cell apoptosis after asODN1, asODN2 and asODN3 treatment.In addition,the decrease in bFGF protein was 63%, 42%, 11%, respectively. CONCLUSION: The data suggeste that asODN1 is a potent target to bFGF mRNA, which inhibits cell growth and induces apoptosis in SWO-38 cells.
10.Teaching Exploration and Feeling about Starting the New Course of Sociology of Sexuality in Our University

According to the changeover of modern medical teaching mode and the undergraduates' need of knowledge structure,we started the course Sociology of Sexuality in our university.In more than 3 years teaching experience and exploration,we arranged the content for each part of each chapter based on the teaching outline,teaching scheme and the students' knowledge and ability structure.As to teaching method,we adopted multimedia computer instruction,heuristic instruction,interactive instruction,participating teaching and audio-visual digital teaching method and so on.We are to introduce such a teaching means of exchange communication,and such an examination method combined with the traditional test as well as testing the capability of handling the actual problem,which can help to implementing the teaching work for Sociology of Sexuality.
