1.Reseach on host immune response in patients with chronic hepatitis B during antiviral treatment with Adefovir ester and Fuzheng Huayu

Objective To investigate the immune response of Adefovir ester combine the Fuzheng Huayu in the process of the treatment of chronic hepatitis B.Methods 100 cases with CHB from March 2010 to October 2014 collected in outpatients clinic or hospital were randomly divided into Adefovir ester treatment group and Adefovir ester joint the Fuzheng Huayu treatment group.Levels of AST, ALT, HBsAg and HBV DNA pre-and-post treatment were detected.CD3-CD56 +,CD244 +NK,CD3 +,CD3 +CD4 +,CD3 +CD8 +T cells and the contents of CD 4 +T cells in serum of two groups were measured by flow cytometry analysis.Results The levels of AST、ALT、HBV-DNA of two group post-treatment were all significantly lower than pre-treatment(P<0.05),and the level of AST and ALT in Adefovir ester and Fuzhengh Huayu treatment group at 3 months and 6 months were obviously lower than Adefovir ester treatment group.HBV-DNA contents between two groups had no significant difference.HbsAg levels of two groups were all significantly increased after 6,9,12 months’treatment (P<0.05),While there was no significant difference between two groups post-treatment.CD3-CD56 +cell at 6,9,12 months in two groups were all higher than pre-treatment(P<0.05), which in Adefovir ester and Fuzhengh Huayu treatment group were more higher than Adefovir ester treatment group(P<0.05).CD244 +NK,CD3 +T cell, CD3 +CD4 +Tcell at each point post-treatment in peripheral blood of two groups were all increased significantly(P<0.05),which in Adefovir ester and Fuzhengh Huayu treatment group increased more obviously than Adefovir ester treatment group (P<0.05).However, CD3 +CD8 +cells were reduced obviously after treatment in two groups(P<0.05), which besides 3 month point, its ratio in Adefovir ester and Fuzhengh Huayu treatment group decreased more significantly than Adefovir ester treatment group at other points( P<0.05 ).Cytokines in serum of two groups were all increased significantly post-treatment ( P<0.05 ) , which in Adefovir ester and Fuzhengh Huayu treatment group were more lower (P<0.05).Conclusion Adefovir ester and Fuzhengh Huayu treatment can improve the situation of AST, ALT, HBV DNA and HBsAg in patients with chronic hepatitis B,promote the expression of NK cells,T cells and Tcell secrited cytokines,and the efficacy is better than Adefovir estertreatment alone.

Objective:To realize Tangshan City’s medical institutions of health and human resources. Methods: Using gray predict model to prognosis Tangshan City’s main medical human resources, and find out the phenomena of lacking of medical administrative staff, disproportional of nursing staff, and shorting of assistant departments staff, such as pharmacy and medical laboratory.Results: In the next few years, we should focus on strengthening the ability of the health technical personnel, except practicing(assistant)doctors.Conclusion:Through increasing the orientation training and recruitment, improving treatment, changing ideas, etc, to achieve the rationalization of health personnel allocation. In order to coping with health care reform, we should complete the allocation and reserve of health human resources.
3.Study on Pureparation Techniques of Jiawei Sini Granules

Objective To optimize the conditions of Jiawei Sini granules by inspecting the macroporous resin adsorption refined methods.Methods Orthogonal design was used with saikosaponin A,paeoniflorin,hesperidin and glycyrrhizin as indicators determined by HPLC.Results The AB-8 macroporous resin,the biggest per gram of resin adsorption capacity were saikosaponin A 2.8 mg,paeoniflorin 62.8 mg,hesperidin 49.6 mg,glycyrrhizin 84.9 mg,with the concentration of durg 0.5 g/mL,the resin column diameter-height ratio of 1∶7 and adsorption velocity of 2(BV/h).Conclusion Macroporous resin adsorption method can be used for refining Jiawei Sini Granules.
4.Development of a device for imitation of ship shock motion

Objective:To develop an economical,simple and portable device for imitation of ship shock motion produced by non-contacting under water explosion.Methods:The device was developed based on aerodynamical principles and sensor detection technology,and was tested with animal experiments to verify its performance.The injuries caused by 4 grades of shock motions with different accelerations,duration and displacement were observed in 40 black rabbits(10 for each grade).Results:The device was safe and functionally stable,with a peak acceleration of 1 000 m/s~2,a shock duration of about 2 ms and a device displacement of 100 mm.The accelerations of 4 shock motion grades were significantly different and with good reproducibility.The damage to the rabbits were mainly haemorrhage of different extents in organs of pleuroperitoneal cavity,especially in the lungs,but no rupture of organs was found.The degree and involvement of damage had an increasing tendency with the increase of acceleration of shock motion.Conclusion:The device we developed can imitate the effects of ship shock motion.

Objective To study the effect of early administration of a mixture of Anisodamine and A-luminium hydroxide gel via nasal gastric tube in preventing in stress - induced digestive tract bleeding in patients with severe brain damage. Method One thousand one hundred and thirteen patients with severe brain damage were divided into three groups: control group 410 cases without any measures to prevent digestive tract bleeding within 72 hours after injury, treatment group A 540 cases when given cimetidine, ranitidine and Losec by intravenous injection or nasal feeding. Objective To explore the consistency and significance of optical coherence tomography (OCT) and clinical and histopathological findings in adoptively transferred uveitis in mice.Methods The adoptively transferred experimental autoimmune uveitis (EAU) model was established by intraperitoneal injection of antigen-specific T cells in C57BL/6 mice.Since 9 days after transferred,inflammation of eyes was observed by indirect ophthalmoscope with +90D lens and record clinical scores every 3 days.The disease was divided into 6 phases including onset phase,early phase,pre-peak phase,peak phase,resolution phase and late phase of EAU,which respectively corresponding to clinical score 0.5,1.0,1.5-2.0,2.5-3.0,1.0-2.0 and less than 1.0.Since 9 days after transferred,the retina and retinal thickness (RT) was measured by spectralis OCT about 1 disc from the disc edge in 10 time points including 9,11,16,21,25,30,35,40,50 and 60 days after transferred.The OCT score was recorded as from 0.0 to 4.0.After transferred 9,21 and 60 days,the mice were killed and eye balls were examined in histology.OCT score,clinical score and histology in the mouse were compared and analyzed.Results The disease was divided into onset phase,early phase,pre-peak phase and peak phase of EAU,which respectively corresponding to 9,16,21 and 26 days after transferred.In four phases,OCT score were 0.5,1.0,2.0 and 4.0 respectively.After transferred 30 days,which was in resolution phase of EAU,the inflammation cells in vitreous were decreased and OCT score was 3.0.After transferred 60 days,which was in late phase of EAU,inflammation cells in vitreous were disappeared and retina was atrophic topically.The histology showed the vitreous has slight inflammation cells and retinal structure was normal at onset of EAU.The vitreous has massive inflammation cells and retina structure was disorder at pre-peak of EAU.And in resolution phase of EAU,the inflammation cells in vitreous were slightly and retina was atrophic and thinned.The data in this study demonstrated that OCT score was well correlated with clinical score in EAU (r=0.957 9,P<0.000 1).Conclusion OCT and clinical and histopathological findings in adoptively transferred uveitis in mice were consistency and OCT is contribute to evaluate the disease dynamically and quantifiably.
7.The Expression and Significance of TSP-1 in Breast Cancer

Objective: To investigate the correlation of thrombospondin-1(TSP-1) expression and angiogenesis and clinical pathologic factors in breast cancer. Methods:Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of TSP-1 mRNA in the primary breast cancer tissues and the tissues set apart 5 cm from the tumor. Im-munohistochemistry was used to detect the expressions of vascular endothelial growth factor (VEGF), c-erbB2, estrogen receptor (ER), progesterone receptor (PR) and microvessel density (MVD). All data were statistically analyzed. Results: The positive rate of TSP-1 was significantly higher in breast cancer than that in tissues set apart 5 cm from tumor (P < 0.05). The expression of TSP-1 was positively correlated with tumor size only (P < 0.05). MVD was lower in TSP-1 positive group than that of negative group (P< 0.05). Conclusion: The MVD was decreased with up-regulation of TSP-1 expression. Thrombospondin-1 may repress the angiogenesis in breast cancer.
8.Experimental Study of Therapeutic Effect of Suhuang Zhike Capsules for Guinea Pig Model with Cough Variant Asthma

Objective To observe the effects of Suhuang Zhike capsules(SZC) on the guinea pig model of cough variant asthma(CVA), and to explore its therapeutic mechanism. Methods Fifty guinea pigs were randomly divided into normal control group, model group, prednisone acetate tablets group, and high-and low-dose SZC groups. With reference to the related literatures, we set up the guinea pig model of CVA. The observation indexes included cough frequency, lung histopathology, the percentage of eosinophils in bronchoalveolar lavage fluid (BALF), and the serum levels of interleukin 4(IL-4), interferon gamma(IFN-γ), tumor necrosis factor alpha (TNF-α), and immunoglobumin E(IgE). Results Compared with the normal control group, the cough frequency of the model group was significantly increased(P<0.05); compared with the model group, the cough frequency of high-dose SZC group was reduced obviously(P < 0.05). Compared with the normal control group , the percentage of eosinophils in BALF of the model group was significantly increased(P < 0.05); the percentage of eosinophils of the three medication groups was lower than that of the model group (P<0.05). The pathological changes of lung tissue in the medication groups were relieved compared to the model group. Compared with the normal control group, the serum IL-4, TN F-α and IgE levels of the model group were increased significantly (P < 0 . 05); compared with the model group , the serum IL-4 and TN F-α levels of prednisone acetate tablets group were significantly decreased, and the serum TNF-α and IgE levels of high-dose SZC group were decreased significantly (P < 0.05). Conclusion SZC could reduce the serum, TNF-α and IgE levels , and relieve the cough symptom, airway inflammation and the lung tissue pathology in CVA guinea pig, so as to achieve the therapeutic effect on CVA.
9.Clinical Characteristic of 34 Patients with Extranodal Nasal Type NK/T-Cell Lymphoma

Objective Analysis of clinical features and treatment of nasal type NK/T cell lymphoma, to improve the understanding of this special type of lymphoma.Method The clinical data of 34 patients with nasal type NK/T cell lymphoma confirmed by pathology were retrospectively analyzed.Result In 31 cases, 26 cases were EB virus (EBV) infection positive, recent Treatment Effectiveness in the positive group was significantly lower than the negative group (38.46%vs. 87.50%;P<0.05) . Follow up showed until October 2015, 19 of 34 patients died. Among them, 17 cases died of disease relapse or progression, 2 cases died of other diseases. 15 patients survived (11 of them were disease-free) and the median overall survival time was 37 months. The 2 year overall survival was related to clinical stage, B symptoms, EBV infection (the P values were 0.017/0.001, 0.005, and 0.039 respectively).Conclusion EBV infection is associated with patient's treatment response in NK/T-Cell Lymphoma; and the prognosis is associated with clinical features such as clinical stage, B symptom, and EBV infection.
10.Impact of injury severity on transplantation of embryonic neural stem cells following traumatic brain injury in rats

Objective To explore the influence of injury severity on transplantation of embryonic neural stem cells (NSCs) after traumatic brain injury (TBI).Methods The NSCs were isolated from the hippocampus of fetal rats aged at from 12-14 days.The cells were cultured and proliferated in the serum-free medium and identified in vitro.The animals received transplants in the bilateral hippocampal areas at day 3 following mild or moderate TBI separately.Conventional histology,TUNEL and immunohistology were examined to detect BrdU,NSE,GFAP,GalC,NGF and BDNF at day 14 post-implantation.Results BrdU-labeled positive cells in the bilateral hippocampus in the mild TBI group were more than those in the moderate TBI group at day 14 post-implantation.Significant differentiation of the astrocytes recognized as GFAP positive cells in the bilateral hippocampus was found at day 14 post-implantation.The expression of NGF and BDNF proteins was increased following TBI,the most evident in the mild TBI group.Conclusion The influence of injury severity on transplantation may be associated with the change of the microenvironment after TBI.
