1.Comparative study on the different molecule weight of poly-L-lactic acid in the biological function of composite materials

BACKGROUND: With the development of technology, poly-L-lactic acid/β-tricalcium phosphate composite materials show good characters in the tissue engineering, which can promote osteoblast proliferation, reduce rejection reactions, and improve bone healing in a dose-dependent manner. OBJECTIVE: To test the influence of poly-L-lactic acid with different molecule weights on the structure and function of poly-L-lactic acid/β-tricalcium phosphate composite scaffolds. METHODS: Poly-L-lactic acid with molecule weights of 200 000 and 380 000 were combined with β-tricalcium phosphate to produce composite scaffolds by using freeze-drying method. Porosity and pore size of the samples were measured. The fetal rabbit bone mesenchymal stem cells (BMSCs) were cultured and expanded in vitro. They were harvested and seeded into the prepared poly-L-lactic acid/β-tricalcium phosphate scaffolds. The 3-(4, 5-Dimethylthiazol-2yl)-2, 5 diphenyltetrazolium bromide (MTT) and alkaline phosphatase were examined for comparison between normal cultured BMSCs and those cultured on the different poly-L-lactic acid/β-tricalcium phosphate scaffolds. RESULTS AND CONCLUSION: Images of scanning electron microscope showed that the cells adhered to the scaffolds greatly. The value of MTT and alkaline phosphatase showed no significant difference (P > 0.05). The molecule weight of poly-L-lactic acid has no influence on the biological function of composite materials.
2.Investigation of Demodex Infection Among Medical Students

Squeezing and smear method was used to detect Demodex infection for college students,the overall infection rate was 30.81% with Demodex folliculorum as the major one,D.brevis and mixed infection as the second ones.Certain relations were found between facial signs,lipid content and the infection.Prevalence was higher in those students who used to share washing materials(towels,etc.)with family members.
3.Clinical observations on the efficacy of total glucosides of paeony combined with ebastine in the treatment of chronic idiopathic urticaria

Objective To evaluate the efficacy and safety of total glucosides of paeony combined with ebastine for the treatment of chronic idiopathic urticaria.Methods A randomized,open-labeled,positive drugcontrolled,parallel-group study was carried out.Sixty patients with chronic idiopathic urticaria were randomly divided into two groups using a random digit table:combined group treated with total glucosides of paeony 600 mg thrice daily combined with ebastine 10 mg per day,control group treated with ebastine 10 mg per day only.The treatment lasted 12 weeks followed by a four-week follow-up.Adverse reactions were recorded and treatment efficacy was evaluated by using urticaria activity score over seven days (UAS7).Results The UAS7 was 9.28 ±4.59,5.83 ± 4.44 and 7.52 ± 5.57 in the combined group on week 8,12 after the initiation of treatment and week 4 after the withdrawal of treatment,respectively,significantly lower than that in the control group at the three time points (13.29 ± 4.72,P < 0.05; 9.86 ± 5.46,P < 0.01; 16.21 ± 5.34,P < 0.01).Significant differences were observed in the response rate between the combined group and control group at the end of the 12-week treatment (75.9% vs.42.9%,x2 =4.56,P < 0.05).There was a decreased recurrence rate in the combined group combined with the control group at the end of the follow-up (13.6% vs.50.0%,x2 =3.90,P <0.05).No obvious adverse reactions were noted in either of the two groups.Conclusion Total glucosides of paeony could markedly enhance the efficacy of ebastine for the treatment of chronic idiopathic urticaria with a reduction in recurrence rate.
4.Growth, reproduction and blood physiological and biochemical indexes in an atopic dermatitis mouse model

BACKGROUND:The skin lesions, pathological and immunological characteristics of Nc/Nga mice are consistent with human atopic dermatitis, and as an atopic dermatitis animal model, it has great research value. But there are no reports on the physiological and biochemical parameters of Nc/Nga mice in China. OBJECTIVE:To observe the growth and reproduction and blood physiological and biochemical parameters of atopic dermatitis model of NC/Nga mice METHODS:The data of reproductive performance of NC/Nga mice from the first to the third generation was analyzed, including mean litter size, weaning rate, pregnancy rate and generation interval. The body mass of 60 Nc/Nga mice with 1-56 days old was measured, 30 mice of female and male, then the growth curve was draw. The blood samples from the infraorbital vessels were col ected to detect the blood physiological and biochemical parameters in the mice. RESULTS AND CONCLUSION:The mean delivery interval of Nc/Nga mice was (25.8±3.1) days, mean litter size was (7.5±2.5) mice, mean weaning rate was (97.2±1.2)%, the mean pregnancy rate was (97.0±1.4)%, and there was no significant difference among the three generations of these mice (P>0.05). The body mass of Nc/Nga mice was increased with the time increasing of days, the body mass of the mice was maximal within 1-2 weeks post-weaning, and there was no significant difference in body mass between males and females at 6 weeks post-weaning (P>0.05). Comparison of the blood physiological and biochemical parameters of mice at the same age between males and females showed that the levels of red blood cells and hemoglobin in female mice were significantly higher than those in the male mice;the platelet count in the male mice was significantly higher than that in the female mice (P<0.05);the triglycerides and albumin levels in the female mice were higher than those in the male mice (P<0.05). The results indicate that the gender and age may influence the blood physiological and biochemical parameters of Nc/Nga mice.
5.Comparison of pressure changes in the common bile duct after laparoscopic common bile duct exploration with primary closure of bile duct or Ttube biliary drainage

6.Clinical analysis transabdominal incision total gastrctomy for gastric carcinoma in 170 cases

Objective To investigate the indications and suitable surgical operation routes of total gastrectomy for gastric carcinoma and the reconstruction procedure of digestive tract following total gastrectomy.Methods The clinical data of total gastrectomy by abdominal incision in 170 patients with GC from 1991 to 2001 were reviewed.Results Radical total gastrectomy was prfomed in 132 cases,palliative total gastrectomy in 38 cases,total gastrectomy with combined re section of other organs in 18 cases,Roux en Y esophagojejunostomy in 110 cases following total gastrectomy.Interposition with jejunum in 60 cases following total gastrectomy.There are 20 cases with dumping syndrome and 6 cases with reflux esophagitis occurring in Roux en Y esophagojejunostomy,but none of cases occurs in those by interposition with jejunum.Conclusions (1)Transabdominal incision is the better choice for patients of GC,especially gastrocardiac carcinoma.(2)The total gastrectomy can raise the survival rate and quality of life of patients with GC,if the indications are stricted.(3)Interposition with jejunum following total gastrectomy is superior to Roux en Y esophagojejunostomy.
7.Influence factors of the timing of endoscopic examination in nonvariceal upper gastrointestinal bleeding

Objective To explore the risk factors affected the timing of endoscopic examination in nonvariceal upper gastrointestinal bleeding (NVUGIB).Methods From January 2001 to December 2011 , 441 patients diagnosed as NVUGIB under endoscope were enrolled,and they were divided into the group receiving endoscopy within 24 hours (233 cases)and the group receiving endoscopy beyond 24 hours (208 cases).The demographic and clinical characteristics of the two groups were analyzed.Student′s t test was used for measurement data comparison and chi-square test for numerical data.Logistic regression analysis was used to analyze the multivariate factors which influenced the timing of endoscopy examination.Results The rates of complication in the group receiving endoscopy within 24 hours and the group receiving endoscopy beyond 24 hours were 9.0% (21/233)and 19.7% (41/208),and the difference was statistically significant (χ2 =10.411 ,P =0.001 ).The mean systolic blood pressure were (118 ±19 )and (122±19)mmHg (1 mmHg=0.133 kPa),respectively,and the difference was statistically significant (t=2.099,P = 0.036 ).On admission the mean hemoglobin were (98 ± 27 )and (93 ± 28 )g/L, respectively,and the difference was not statistically significant (t=1 .640,P =0.102).The mortality rate were 1 .3% (3/233 )and 5 .3% (11/208 ),respectively,and the difference was statistically significant (χ2 =5 .723,P =0.017).With multivariate Logistic regression,multivariate factors analysis indicated that complication was the main factor influenced the timing of endoscopy examination (OR =2.710,95 %CI :1 .502 to 4.899,P < 0.01).However,age,gender,systolic blood pressure,hemoglobin levels on admission,location of lesion and clinical symptoms were not related with timing of endoscopy examination (all P >0.05).Conclusion Complication is a risk factor in the timing delayed endoscopy.
8.Construction of the DNA vaccine plasmid against CDR3 region of immunoglobulin heavy chain of the human B-cell lymphoma cell line Namalwa

Objective:To analyze whether the DNA vaccine against the CDR3 regio n of immunoglobulin heavy chain of the human B-cell lymphoma could elicit the s p ecific idiotypic antibody,the authors established a model with the human B-cell lymphoma c ell line Namalwa.The CDR3 gene fragment of Namalwa membranous immunoglobulin hea vy chain was amplified.The sequenced CDR3 fragment was used as the antigen gene to construct the DNA vaccine plasmid. Methods:The authors acquired the CDR3 gene fragment using Ig superfamily primers by means of RT-PCR and the murine monocyte chemot ic protein(MCP-3) as the adjuvant molecular.The fused gene fragment of CDR3 and MCP-3 was obtained by recombinant PCR and then cloned into the eukaryonic plas mid vector pcDNA3.1 to construct the DNA vaccine plamid.Then the vaccine plasmid was transiently expressed in the eukaryonic cell COS-7. Results:The authors acquired th e DNA vaccine plasmid in which the mIgCDR3 of the Namalwa cell was used as the a ntigen gene by the molecular biology. Conclusion:The transient transfection assa y proved that the recombinant plasmid could express in eukaryonic cells in right way.They have constructed an expression plasmid containing fused MCP3-CDR3 seq uen ce which could be used in further study of DNA vaccine against B-cell lymphoma in vivo. 9.Effect of Nicorandil on Myocardial Ischemia-reperfusion Injury in Isolated Rat's Heart

Objective: To investigate the effect of potassium channel opener nicorandil on myocardial ischemia-reperfusion injury (MIRI) in isolated rat's heart. Methods: Langendorff system was established for isolated heart reperfusion. A total of 44 rats were randomly divided into 4 groups: Normal control group, Ischemia-reperfusion (I-R) group, Adenosine/I-R (A/I-R) group, the I-R heart was treated by adenosine 100 μmo/L and Nicorandil/I-R (N/I-R) group, the I-R heart was treated by nicorandil 200 μmo/L.n=11 in each group. Left ventricular developed pressure (LVDP), the maximal rise/fall rate of left ventricular pressure (±dp/dtmax), reperfusion arrhythmia (RA) and myocardial infarction (MI) size were recorded; the expressions of left ventricular tissue acetaldehyde dehydrogenase 2 (ALDH2), Bcl-2 and Bax were examined and compared among different groups. Results:①Compared with I-R and A/I-R groups, N/I-R group had increased LVDP at 30 min and 45 min reperfusion, allP<0.05.②Compared with N/I-R and A/I-R groups, I-R group showed reduced ±dp/dtmax at 30 min and 45 min reperfusion, allP<0.05; while ±dp/dtmax in N/I-R group was higher than A/I-R group at 30 min and 45 min reperfusion, all P<0.05.③Compared with I-R group, both N/I-R and A/I-R groups presented reduced RA score, allP<0.01, RA score was similar between N/I-R group and A/I-R group,P=0.771.④compared with I-R group, both N/I-R and A/I-R groups had the smaller MI size,P<0.01 and N/I-R group showed the smaller MI size,P<0.05.⑤Compared with Normal control group, I-R group had decreased mitochondrial ALDH2 expression and increased Bcl-2, Bax expression; ALDH2 level was elevated in N/I-R group had reduced in A/I-R group; Bcl-2 and Bax levels were increased in both N/I-R and A/I-R groups. Compared with I-R group, both N/I-R and A/I-R groups had increased ratio of Bcl-2/Bax,P<0.05; the anti-apoptosis effect was similar between N/I-R group and A/I-R group,P>0.05. Conclusion: Nicorandil may reduce MIRI and protect myocardium in isolated rat's heart, the overall effect is better than adenosine. Nicorandil can up-regulate the expressions of mitochondrial ALDH2, Bcl-2 and down-regulate the expression of Bax.
10.hBMP-2 gene modified tissue engineered bone repair the segmental bone defect of rabbit's radius

Objective To compare and evaluate the defect-repaired capabilities of human bone morphogenetic protein-2(hBMP-2) gene modified tissue engineered bone in the segmental bone defect model of rabbit's radius.Methods Rabbit's bone mesenchymal stem cells (BMSCs)were transferred with hBMP-2 gene through Adeno-XTM adenoviral expression systems,then seeded onto the compound scaffold of calcium phosphate cemept(CPC)and fibrin glue(FG)to construct a new kind of gene modified tissue engineered bone after proliferation in vitro for three weeks(Group A).Meanwhile,the compound scaffold of calcium phosphate cement(CPC)and fibrin glue(FG),which were seeded by rabbit's bone knesenchyrmal stem cells(BMSCs) after proliferation in vitro for three weeks(group B)and the compound scaffold without cells(Group C)acted as control groups.Then,three kinds of reconstructive modalities were implanted into segmental bone defect of donator rabbit's radius.Besides these three groups,bone defect model of rabbit's radius without treatment(Group D)represented blank group.The defect-repaired capabilities were assessed by gross observation,radiograph,Single Photo Emission Computed Topography (SPECT)and histological analysis in the 4th week,8th week and 12th week after operation.The rates of bone healing in the different groups were compared each other.Results All defects that had been treated with implants(Group A,B,C)exhibited new bone formation and could attain osseous tissue healing 12 weeks after operation,but defects in blank group(Group D)were repaired only by fibrous tissue.The defects in the Group A regenerated more new bone,bridged earlier and stronger than those in the Group B and Group C.The quantity and rate of new bone formation in the Group B and Group C had no significant difference and the rates of bone healing in different groups showed the same results.Conclusion hBMP-2 gene modified tissue engineerod bone have better potential to form new bone and the rate of bone healing in repairing bone defects is higher,so this way is an optimal kind of material for artificial bone graft.
