1.Supplementary effects of Xuanfei Zhisou Mixture on cough variant asthma in patients with syndrome of wind pathogen invading lung

AIM To observe the supplementary effects of Xuanfei Zhisou Mixture (Schizonepetae Herba,Peucedani Radix,Platycodi Radix,etc.) on cough variant asthma in patients with syndrome of wind pathogen invading lung.METHODS Forty-eight patients,receiving budesonide/formoterol turbuhaler,were randomly and equally divided into trial group and control group using Stata 12.0 software.The patients in the trial group took Xuanfei Zhisou Mixture additionally.Cough symptom score,cough visual analogue scale,quality of life,blood eosinophil count and adverse reaction in two groups were compared.RESULTS Compared with the control group,the trial group provided more benefits in terms of cough symptom score,cough visual analogue scale and quality of life with statistical differences.There was no statistical difference in blood eosinophil count between the two groups.Adverse reactions such as hoarseness and oropharyngeal fungal infection in the trial group,and palpitation and tremor in the control group occurred in one patient in the process of the treatment,respectively.CONCLUSION Xuanfei Zhisou Mixture combined with budesonide/formoterol turbuhaler is an effective treatment method for cough variant asthma with syndrome of wind pathogen invading lung.
2.Bacterial Contamination of Auxiliary Equipments of Ventilator and Corresponding Countermeasures

OBJECTIVE To investigate the bacterial contamination status of ventilator systems and corresponding measurements.METHODS Bacterial culture samples of auxiliary equipments of 36 ventilators,including inlet and outlet tube,mask,humidifier,water collector,nebulizer,and air filter,were respectively collected.The culture results were reported to the infectious disease control office of the hospital.After improved the sterilization management,then repetition of bacterial culture was performed and the results were compared with the former.RESULTS The first cultural results demonstrated severe bacterial contamination and the positive rate of inlet tube,humidifer,water collector,nebulizer,and air filter was even higher than 50%.After countermeasures were conducted,the positive rate of second bacterial culture of all ventilator systems was significantly dropped to less than 5.5%.CONCLUSIONS Bacterial contamination of ventilator systems commonly exists which can be significantly diminished by improved conception of asepsis and standardized sterilization.

6.The unreasonable use of high beam at nighttime among motor vehicle drivers

Objective:
To observe the incidence of unreasonable use of high beam at nighttime among motor vehicle drivers, so as to provide the evidence for the prevention and control of road traffic injury.
Methods:
Four roads into city and five urban roads were selected in Yongkang of Zhejiang Province. An automatic recording system was used to collect the unreasonable use of high beam among motor vehicle drivers on the selected roads from 19:00 to 5:00 on Monday, Wednesday, Friday and Sunday during a week in July 2020. The regression tree model was used to analyze the relationship of the unreasonable use of high beam with road, time and traffic flow.
Results:
A total of 89 989 motor vehicles were observed, and 2 419 motor vehicle drivers had unreasonable use of high beam, with an incidence rate of 2.69%. The incidence rate of the unreasonable use of high beam was 3.14% in the roads into city, which was higher than 2.30% in the urban roads ( P<0.05 ). The incidence rates of the unreasonable use of high beam in the roads into city and in the urban roads were 5.15% and 2.90% on Wednesday, which were higher than those on Monday ( 2.89% and 2.34% ), Friday ( 2.90% and 1.92% ) and Sunday (2.06% and 2.12%). The highest incidence rate of the unreasonable use of high beam in the roads into city was 6.07% between 4:00 and 5:00, and in the urban roads was 4.50% between 2:00 and 3:00. The results of regression tree classification analysis showed that the highest incidence rate was 8.13% on the roads into city in the east, west and south directions, and on the urban roads in the east and north directions with less than 317 vehicles per hour on Wednesday.
Conclusion
It is more likely for motor vehicle drivers to use high beams unreasonably at nighttime on the roads into city with less traffic flow.
7.Acquiring homozygous tetraploid germplasm by PEG-mediated protoplast fusion of Rhodiola sachalinensis.

OBJECTIVETo acquire homozygous tetraploid germplasm of Rhodiola sachalinensis.
METHODPEG-mediated protoplast fusions were conducted using callus of Rh. sachalinensis as materials. Protoplast fusion products were embedded and cultured in low-density, low-melting-point agar and marked according to the protoplast size, and single-celled sister lines were established to acquire genetically homozygous tetraploid germplasm.
RESULTR(D) and R(M) of newborn daughter cells or protoplasm, metaphase cells or protoplasm were approximately in line with the formula R(D) = 0.793 7R(M). The change range in diameter of the diploid cells without fusion, two protoplasts fusion product were: 16.7 microm < or = R < 21.3 microm, 21.0 microm < or = R' < 26.8 microm respectively. There is an overlap between the two diameter ranges. The protoplast inoculation density of 1 x 10(4) cells x mL(-1) was appropriate when protoplasts were anchored by low-intensity, low-melting-point agar. Under the conditions of this density, plating efficiency was high and single cell origin of the sister lines microclones grew rapidly, and it was easy to mark the single cell microclones, and separate from each other to subculture. The chromosome counts results showed that chromosome numbers of diploid and tetraploid of single cell lines were 26 and 52, respectively. The result from flow cytometry assay showed that there is no presence of chimerism in single-cell regeneration plantlets.
CONCLUSIONThe results of this study provide a scientific basis for polyploid breeding of Rh. sachalinensis.

8.Research progress and prospect of metabonomics in glaucoma

Metabonomics is an important component of systems biology.It reveals the essential metabolic characteristics of the activities of organisms using qualitative and quantitative detection and analysis of the dynamics of all endogenous low-molecular-weight metabolites within an organism before and after stimulation such as pathophysiological stimuli or genetic modification.Therefore, it provides insight into the pathogenesis, early diagnosis, treatment and prognosis of diseases.Metabonomics relies on chromatography, mass spectrometry, nuclear magnetic resonance spectroscopy and other analytical chemistry techniques to obtain data.The examination specimens are usually body fluids including blood, tear, aqueous humor and tissues such as trabecular meshwork, vitreous body, retina, etc.Glaucoma is an optic nerve disease characterized by optic disc damage and visual field defect.Previous animal and human studies have provided some preliminary results on metabolites associated with glaucoma.Metabonomics, genomics, transcriptomics, and proteomics constitute a bioinformatics system.Joint multi-omics research will be the direction of future development.On the basis of an overview of metabonomics, this paper reviewed the research progress of metabonomics in glaucoma based on different tissues and body fluid specimens.
9.Metagenomic next-generation sequencing of a case of intracranial hemorrhage with positive herpes simplex virus 1 and literature review

Objective:To investigate the clinical features of Herpes simplex virus encephalitis(HSE) with cerebral hematoma as the prominent manifestation and the significanc of metagenomic next-generation sequencing (mNGS) in the diagnosis of HSE.Methods:The clinical manifestations, diagnostic process, clinical treatment and prognosis of a case of HSE with cerebral hematoma as the prominent manifestation at Shanghai Children′s Medical Center Affiliated to Shanghai Jiaotong University School of Medicine in June 2019 were retrospectively analyzed.The relevant literatures were also searched and reviewed.Results:A 4-year-old boy presented with slight fever, headache, convulsion and vomiting was considered to have intracranial space-occupying lesions and possible intratumoral hemorrhage after undergoing imaging examination at a local hospital.The patient was checked by head CT in Shanghai Children′s Medical Center Affiliated to Shanghai Jiaotong University School of Medicine, which showed that there were many bleeding foci in the brain, indicating the possibility of complications of blood system diseases.Therefor the child was given the examination of blood routine and coagulation routine, but the results were normal, the bone marrow cytology was negative, the cerebrospinal fluid(CSF) of lumbar puncture was biochemically normal, and mNGS were 8×10 6/L.Besides, CSF smear, culture and next-generation sequencing were negative, the autoimmune encephalitis CSF testing was negative, and brain biopsy suggested inflammation.The mNGS brain tissue showed herpes simplex virus 1 was positive in two specimens, confirming the diagnosis of HSE.After 3 weeks of antiviral treatment with Aciclovir, the child′s condition improved.After a 5-month follow-up, the patient had quadriplegia and only had activities such as blinking and swallowing. Conclusions:When the intracerebral hemorrhage such as hematoma caused by encephalitis clinically can not be ruled out, the possibility of HSE should be considered, and mNGS is helpful for identifying the central ner-vous system pathogen.
10.Neuroimaging advances in the regression of white matter hyperintensity of presumed vascular origin

As one of the imaging features of cerebral small vessel disease, white matter hyperintensity (WMH) of presumed vascular origin is quite common in the elderly. The burden of WMH is thought to progress slowly over time and is significantly associated with cognitive decline. However, the pathogenesis of WMH remains unclear and there is no effective treatment available. Recent studies have reported that some WMH lesions can regress during follow-up, along with progression and regression occurring alternately, suggesting that longitudinal changes in WMH are not unidirectional. This article presents a systematic review of current neuroimaging studies on WMH regression to enhance the understanding of dynamic changes in WMH and to provide new theoretical evidence for WMH intervention.
