The pharmacokinetics/pharmacodynamics ( PK/PD) of antimicrobials aims to establish and evaluate dose-concentration-response relationship, and subsequently to describe and predict the effect-time courses resulting from a drug dose.Nowadays, PK/PD is playing an important role in dosage regimen optimizing, reducing toxicity and drug tolerance of antibiotics.And it is also of great significance in determining susceptibility breakpoints and development in new drug.This paper reviews the progress on PK/PD studies of antibacterial and antifungal agents to provide theoretical basis for safe and effective individual dosage regimens.
2.The status and demands of prenatal education among pregnant women in Chengd

Objective To investigate the status and demands of prenatal education among pregnant women,and to provide reference for better prenatal education. Methods A total of 750 pregnant women were recruited from 20 hospitals in Chengdu and were investigated with a serf-designed questionnaire. Results 74.42% of the pregnant women had participated the prenatal education,25.58% of them didn't take even one prenatal class. The most preferred knowledge was newborn care. They most preferred to take prenatal classes at weekends in small group. The women's educational level,family income, delivery experience and times of prenatal check-up were the key factors to affect the participation in the prenatal educa-tion. Conclusions It is suggested to innovate the contents and means of prenatal education in order to attract the pregnant women to participate prenatal education actively.
3.MRI assessment of acute myocardial infarction with transplantation of autologous mesenchymal stem cells in swine:an experimental study

ObjectiveTo investigate the effects of autologous bone marrow-derived mesenchymal stem cells (MSCs)transplantation on acute myocardial infarction in swine models using MRI. MethodsFourteen Chinese mini-pigs(27±3 kg)were divided into control group(n=7)and transplantation group(n=7).Acute myocardial infarction(AMI)model was made by occlusion of the left anterior descending coronary artery for 90 minutes,and then 10 ml autologous MSCs(3 × 106 cell/ml)were injected into LAD by over-wire-balloon catheter after one week. MRl was performed to assess the cardiac function and myocardial perfusion 1 week after AMI and 6 weeks after transplantation.The implanted cells in vitro were analyzed by immunofluorescence.ResuitsThe left ventricular ejection fraction(LVEF)in transplantation group was increased from(42.7 ±7.5)%to(50.1±10.1)%,which was significantly different from that in control group(P<0.01).In addition,the dyskinetic segments in infarcted region and the infareted area were decreased by 4 and 3.2 cm2 respectively(P<0.01),and the left ventricular weight index was increased by 4.1 g/m2 in transplantation group(P<0.05)compared with control group.The DAPI-labeled cells in infarcted and peri-infarcted region indicated the survived MSCs.Immunofluoreseence also confirmed that those cells expressed cardiomyocyte-specific troponin T,connexin 43 and vessel-specific smooth muscle actin.Capillary density in both infarcted and peri-infarcted region were higher in transplantation group than the control group(P<0.01).Conclusion MRI is a reliable imaging method for assessing the effects of stem cell transplantation in acute myocardial infartion of swine models.
4.Inhibitory effects of small interference RNA (siRNA) on herpes simplex virus type 2

ObjectiveTo investigate the antiviral effect of siRNA on herpes simplex virus type 2 (HSV-2) in vitro.MethodsDifferent siRNA duplexes targeting HSV-2 VP16 and DNA polymerase genes were designed and chemically synthesized.The antiviral effect of siRNA duplexes was evaluated by virus yield reduction assay.In detail,Vero cells cultured in 24-well plates were transfected with different siRNAs 4 hours before(preventive effect experiment) or 1 hour after(therapeutic effect experiment) the inoculation with HSV-2(0.02 to 0.05 plaque forming unit/cell).After additional culture for 40 to 48 hours,the expression of target genes in Vero cells was measured by real-time quantitative reverse transcription(RT)-PCR.Results Both the siRNA duplex VP2-1 targeting HSV-2 VP16 and DP2-1 targeting DNA polymerase gene resulted in a remarkable decrease in viral titer by about 1 log10 in both the preventive and therapeutic effect experiment.As the preventive effect experiment showed,the expression of HSV-2 VP16 mRNA was decreased markedly by 58.6% and 79.5% by VP2-1,respectively,and that of HSV-2 DNA polymerase mRNA by 59.8%and 77.4% by DP2-1,respectively,at 6 and 12 hours after the infection.ConclusionsThe siRNA duplexes targeting HSV-2 VP16 and DNA polymerase genes could inhibit the proliferation of HSV-2.Hence,HSV-2 VP16 and DNA polymerase genes may serve as the target genes of RNA interference to inhibit HSV-2 replication.
5.Laboratory indicators of lung infection for patients with chronic obstructive pulmonary disease

Objective To evaluate the clinical value of white blood cell(WBC) ,neutrophilic granulocyte percentage(NEUT% ) and C‐reactive protein(CRP) in lung infection of early chronic obstructive pulmonary disease(COPD) ,and analyse the pathogen dis‐tribution .Methods 186 cases of hospitalized patients with COPD were enrolled as research group and 60 healthy ones as control group .Compared WBC ,NEUT% ,CRP levels between the two groups .According to sputum culture results ,patients were divided into normal flora group and pathogen growth group;according to the type of pathogen ,patients were divided into G+ and G- group . Analyse WBC and CRP levels in each groups and compare sensitivity ,specificity and positive predictive value of the two indicators . Distribution of positive sputum culture results were statistically analyzed .Results The levels of three indicators in research group were higher than those in control group(P<0 .05) .WBC and CRP levels in normal flora and pathogen growth groups were not sig‐nificantly different(P>0 .05) .Based on sputum culture positive results ,the number of fungi is 86 ,and Candida albicans were the most accounting for 35 .85% .The number of bacteria were 73 strains .Differences in WBC ,NEUT% and CRP between G+ group and G- group were not significant(P> 0 .05) .Conclusion WBC ,NEUT% and CRP levels in COPD patients were higher than those in healthy group .But due to many factors which could affect the levels of WBC ,NEUT% and serum CRP ,a preliminary diag‐nosis of COPD lung infection can′t be made just through the three indicators .

The First Affiliated Hospital of Zhejiang University founded an online hospital campus leveraging the Internet-plus environment. Supported by the offline hospital, this campus made full use of Internet features and advantages in its attempts to optimize the distribution of medical resources,to improve the accessibility of medical service, to reduce medical costs, and to expand the service coverage among others. Meanwhile,this online system, via interconnection with the hospital′s HIS system, achieved online medical insurance payment,prescriptions checking and drug delivery service.

8.Adipose-derived stem cells combined with gelatin sponge for repair of rabbit radial defects

BACKGROUND:Adipose-derived stem cells discovered recently are a new kind of adult stem cells, and have a strong multi-differentiation capacity. However, there are rare studies concerning in vivo osteogenic capacity of adipose-derived stem cells.
OBJECTIVE:To investigate the effect of adipose-derived stem cells combined with gelatin sponge on repairing bone defects.
METHODS:Adipose-derived stem cells from rabbit inguinal fat pads were isolated and cultured, and then
induced using an osteogenic medium containing bone morphogenetic protein 2 fol owed by injection of gelatin sponge. Radial defect models of rabbits were prepared. Compound of adipose-derived stem cel s and gelatin sponge was implanted into the lesion side, while gelatin sponge alone was implanted into the contralateral side. Rabbits were kil ed at weeks 6 and 12 after bone defect repair for X-ray examination, CT scan, and hematoxylin-eosin staining.
RESULTS AND CONCLUSION:Lane-Sandhu X-ray and Lane-Sandhu histological scores after compound implantation were significantly higher than those after repair with gelatin sponge alone. It indicates that
adipose-derived stem cel s combined with gelatin sponge can promote bone defect healing of rabbits, showing an obvious osteogenic capacity in vivo.
9.Characteristics of hypertrophic cardiomyopathy on delayed contrast-enhanced MRI

Objective To analyze the characteristics of hypertrophic cardiomyopathy (HCM) on delayed contrast-enhanced cardiac magnetic resonance imaging (CMRI). Methods All patients underwent delayed contrast-enhanced CMRI. The left ventricle was divided into 9 segments to assess the location,extent and function of the hypertrophic segments. The t test was applied for the statistics. Results Of 154 patients, delayed enhancement of hypertrophic segment was found in 95 cases and non-delayed enhancement in 59 cases. The thickness and number of hypertrophic segment in patients with delayed enhancement were larger than those with non-delayed enhancement [ (24. 8 ± 5. 5 ) mm vs (20. 4 ±3.8) mm, t = 3.82, P < 0.05; (3.3 ± 1.9) vs (2.4 ± 1.7), t = 2. 26, P < 0.05 ], and the age was younger [ (46. 0 ± 15.2) years vs (55.0 ± 11.9) years, t = - 3. 67, P <0. 05 ]. The diffuse enhancement was found in 62 patients, and confluent enhancement in 33 patients. Confluent enhancement was found in all 14 patients after the alcohol ablation procedure. Conclusion The age, thickness and number of hypertrophic segments in patients with delayed enhancement are different from those with non-delayed enhancement.
10.Studies on antibacterial activity and endotoxin neutralization of murine BPI N-terminal functional fragment(muBPI_(25) protein)in vitro

Objective:To establish an experimental model for intracellular antibacteria and endotoxin neutralization in vitro to detect the antibacterial and endotoxin neutralization activity of the muBPI_(25) protein.Methods: RAW264.7 cells were transfected with pcDNA3.1(+)muBPI_(36-259), and then were infected with intracellular bacterial of either G ~+/G~-to establish the experimental model of intracellalar antibacteria.The RAW264.7 cells were co-transfected with the pSecTag2B-muBPI_(36-259) and dual-luciferase reporter gene plasmids for establishment of the experimental model of endotoxin neutralization.Results:The experimental model of intracellular antibacteria confirmed that the muBPI_(25) protein could inhibit/kill Salmonella typhi.The experimental model of endotoxin neutralization indicated that the muBPI_(25) protein could neutralize endotoxin.Conelusion: We firstly demonstrate that murine BPI N-terminal functional fragment(muBPI_(25) protein)can inhibit/kill Salmonella typhi,and can neutralize, its lysating product, endotoxin.
