1.Reliability and validity of public mood scale in college students

Objective To develop the public mood scale and test its reliability and validity,and explore the components construct of the public mood.Methods 423 college students responded to the inventory.Correlation analysis,exploratory factor analysis and confirmatory factor analysis were applied to the data analysis.Results Public mood scale was consisted of two dimensions,positive emotions and negative emotions.Factor analysis indicated that the two factors explain 61.3% of the variance with factor loading ranging from 0.68 to 0.86;The Cronbach' s coefficients of positive and negative public mood were 0.81 and 0.76 respectively; Conffirmatory factor analysis indicated that the public mood scale had ideal structure validity.Joint factor analysis showed that public mood and personal mood were separated.Positive public mood had positive correlations with Big Five personalities,which were extraversion,agreeableness,responsibility and openness (r =0.22-0.33,P< 0.01),negative correlation was found between neuroticism and positive public mood(r=-0.25,P<0.01).Positive public mood had significant positive correlation with government satisfaction,economic development confidence and social stability(r=0.34-0.45,P<0.01),while they had significant negative correlations with negative public mood (r =-0.32--0.24,P< 0.01).Conclusion The public mood scale obtained in the present study has ideal validities and reliabilities and it is suitable to assess the public mood.

The fine ultrastructure and localization of acid phosphatase in cell ultrastructuralevel of a HGPRT-human promyelocytic leukemia cell mutant(HL-60-AR)arestudied by scanning electron microscopy,transmission electron microscopy,freezeetching,and electron microscopic cytochemistry techniques.The results of theobservation show that the ultrastructural characteristics of HL-60-AR cells aresimilar to that of HL-60 cells.There are microvilli and ridges over cell surface.Thecells have large nucleus with prominent nucleoli,and numerous nuclear pores.Thereare less developed Golgi complex,expanded rough endoplasmic reticulum andabundant polyribosomes.After treatment with retinoic acid(RA)at 10~(-6) mol/L for 5days,HL-60-AR cells differentiate along myeloid pathway and have a decreasednucleocytoplasmic ratio accompanied with nuclear condensation and segmention.A (?)ignificant increase of specific granules is demonstrated.Microvilli of the cellsdisappear,surface features of the treated cells become more irregular and largeprotrusion and blunt pseudopodia appear.Increase of acid phosphatase content localizedon azurophilic granules(lysosomes)and Golgi complex is showed.The application offour kinds of electron microscopic techniques might provide the best way foridentifying cell ultrastructure.
4.MRI assessment of acute myocardial infarction with transplantation of autologous mesenchymal stem cells in swine:an experimental study

ObjectiveTo investigate the effects of autologous bone marrow-derived mesenchymal stem cells (MSCs)transplantation on acute myocardial infarction in swine models using MRI. MethodsFourteen Chinese mini-pigs(27±3 kg)were divided into control group(n=7)and transplantation group(n=7).Acute myocardial infarction(AMI)model was made by occlusion of the left anterior descending coronary artery for 90 minutes,and then 10 ml autologous MSCs(3 × 106 cell/ml)were injected into LAD by over-wire-balloon catheter after one week. MRl was performed to assess the cardiac function and myocardial perfusion 1 week after AMI and 6 weeks after transplantation.The implanted cells in vitro were analyzed by immunofluorescence.ResuitsThe left ventricular ejection fraction(LVEF)in transplantation group was increased from(42.7 ±7.5)%to(50.1±10.1)%,which was significantly different from that in control group(P<0.01).In addition,the dyskinetic segments in infarcted region and the infareted area were decreased by 4 and 3.2 cm2 respectively(P<0.01),and the left ventricular weight index was increased by 4.1 g/m2 in transplantation group(P<0.05)compared with control group.The DAPI-labeled cells in infarcted and peri-infarcted region indicated the survived MSCs.Immunofluoreseence also confirmed that those cells expressed cardiomyocyte-specific troponin T,connexin 43 and vessel-specific smooth muscle actin.Capillary density in both infarcted and peri-infarcted region were higher in transplantation group than the control group(P<0.01).Conclusion MRI is a reliable imaging method for assessing the effects of stem cell transplantation in acute myocardial infartion of swine models.
5.Influence of α-Zearalanol on lipometabolism and hemorheology in experimental hyperlipidemia rabbits

ObjectiveTo investigate the effect of α-Zearalanol(α-ZAL) on lipometabolism and hemorheology in ovariectomized(OVX) hyperlipidemia rabbits.Methods44 adult virgin female rabbits were divided into 5 groups,group A: normal control;group B: sham+CHO;group C: OVX+CHO;group D: OVX+CHO+17βE_2;group E: OVX+CHO+α-ZAL.Cholesterol(CHO) was fed to rabbits for 12 weeks.Before and after feeding CHO,the serum lipid(TC,TG,LDL-C,HDL-C) were measured;Blood viscosity,plasma viscosity,aggregation index of RBC(AIRC) and fibrinogen were also assayed respectively.ResultsThe serum levels of TC,TG and LDL-C in group B and E were significantly decreased compared with those in group C(P<0.05);the level of blood viscosity,plasma viscosity and AIRC platelet aggregation rate in group D and E were also significantly decreased compared with those in group C(P<0.05).Conclusionα-ZAL can improve vascular function through the adjustment of lipometabolism and hemorheology.
6.Characteristics of hypertrophic cardiomyopathy on delayed contrast-enhanced MRI

Objective To analyze the characteristics of hypertrophic cardiomyopathy (HCM) on delayed contrast-enhanced cardiac magnetic resonance imaging (CMRI). Methods All patients underwent delayed contrast-enhanced CMRI. The left ventricle was divided into 9 segments to assess the location,extent and function of the hypertrophic segments. The t test was applied for the statistics. Results Of 154 patients, delayed enhancement of hypertrophic segment was found in 95 cases and non-delayed enhancement in 59 cases. The thickness and number of hypertrophic segment in patients with delayed enhancement were larger than those with non-delayed enhancement [ (24. 8 ± 5. 5 ) mm vs (20. 4 ±3.8) mm, t = 3.82, P < 0.05; (3.3 ± 1.9) vs (2.4 ± 1.7), t = 2. 26, P < 0.05 ], and the age was younger [ (46. 0 ± 15.2) years vs (55.0 ± 11.9) years, t = - 3. 67, P <0. 05 ]. The diffuse enhancement was found in 62 patients, and confluent enhancement in 33 patients. Confluent enhancement was found in all 14 patients after the alcohol ablation procedure. Conclusion The age, thickness and number of hypertrophic segments in patients with delayed enhancement are different from those with non-delayed enhancement.

Objective: To observe the therapeutic effect of acupuncture therapy for treatment of chronic atrophic gastritis. Methods: According to the theory of TCM, 36 cases of chronic atrophic gastritis were differentiated and treated by acupuncture. Gastroscopy and electrogastrograph (EGG) of the body surface, gastric acid and pepsin, blood gastrin, blood prostaglandin E (PGE), gastric mucosal cells and G cells are recorded. Results: Acupuncture therapy had a better effect in improving clinical symptoms, the markedly effective rate was 97.2% and the disappearing rate of clinical manifestations 97.02%;after acupuncture treatment,blood PGE concentration increased; gastric acid secretion increased; gastric dynamics was enhanced and the amplitude of EGG rose; acupuncture could significantly facilitate secretion of gastrin. Conclusion: Acupuncture is a convenient treatment method for atrophic gastritis with quick effect and without side effects.

AIM: To investigate the effect of 17? estradiol (17?E 2)on atherosclerosis and hemorrheology in ovariectomized (OVX) rabbits. METHODS: Thirty four female rabbits were divided into 4 groups, group A: normal control; group B: sham+CHO; group C: OVX+CHO; group D: OVX+CHO+17? E 2. Cholesterol(CHO) was fed to rabbits for 12 weeks, before and after feeding CHO, the serum lipid (TC, TG, HDL-C, LDL-C, ApoA1, ApoB) and area of aortic atherosclerotic plaques were measured. Blood viscosity, plasma viscosity (?p), aggregation index of RBC (AIRC) and fibrinogen were also determined, respectively. RESULTS: ① The ratio of area of aortic atherosclerotic plaque to total area of aortic intima was 0.02?0.003 (in group A), 0.42?0.15 (group B), 0.67?0.23 (group C), and 0.12?0 11 (group D), respectively. 9.Research progress and prospect of metabonomics in glaucoma

Metabonomics is an important component of systems biology.It reveals the essential metabolic characteristics of the activities of organisms using qualitative and quantitative detection and analysis of the dynamics of all endogenous low-molecular-weight metabolites within an organism before and after stimulation such as pathophysiological stimuli or genetic modification.Therefore, it provides insight into the pathogenesis, early diagnosis, treatment and prognosis of diseases.Metabonomics relies on chromatography, mass spectrometry, nuclear magnetic resonance spectroscopy and other analytical chemistry techniques to obtain data.The examination specimens are usually body fluids including blood, tear, aqueous humor and tissues such as trabecular meshwork, vitreous body, retina, etc.Glaucoma is an optic nerve disease characterized by optic disc damage and visual field defect.Previous animal and human studies have provided some preliminary results on metabolites associated with glaucoma.Metabonomics, genomics, transcriptomics, and proteomics constitute a bioinformatics system.Joint multi-omics research will be the direction of future development.On the basis of an overview of metabonomics, this paper reviewed the research progress of metabonomics in glaucoma based on different tissues and body fluid specimens.
10.Detection of viable myocardium by low dose of dobutamine cine MR imaging in miniswine.

OBJECTIVETo evaluate the diagnostic value of dobutamine stress magnetic resonance imaging (MRI) for myocardial viability.
METHODSTen male miniswines underwent left ventriculography and coronary angiography, followed by stenosis of the left circumflex coronary artery (LCX) using ameroid constrictor. More than one month later, left ventriculography and coronary angiography were performed again, followed by cine-MRI at rest and during stress with incremental dose of dobutamine 5 - 20 micro g.kg(-1).min(-1). Traditional and/or breath-hold cine-MRI were used to evaluate regional left ventricular wall motion, corresponding to basal, midventricular and apical short-axis tomograms. Regional wall motion score index (WMSI) was calculated. The miniswines were finally sacrificed for pathological examination. Triphenyl tetrazolium chloride (TTC) delineated myocardial infarction. Microscopy was used to identify myocardial cellular changes.
RESULTSOne pig died, one pig suffered from aneurysm and another showed no negative findings. The other seven pigs were found with hypokinetic (n = 4) or akinetic (n = 3) myocardial regions related to stenosed LCX. Their mean WMSI at rest for the lateral and posteroinferior walls (ischemic regions) of the left ventricle was 2.27 +/- 0.32, as compared with 1.00 +/- 0.00 (P < 0.01) for the corresponding nonischemic anteroseptal regions. Further, the mean WMSI for the ischemic regions was 2.27 +/- 0.32 at rest compared with 1.40 +/- 0.39 (P < 0.01) at the dose of dobutamine 5 micro g.kg(-1).min(-1). However, the mean WMSI at the doses of dobutamine 10 and 20 micro g.kg(-1). min(-1) were 1.70 +/- 0.76 and 1.75 +/- 0.83, respectively, with no significant difference as compared with the mean WSCI at rest (P > 0.05). The pathologic examination showed viable myocardium at the ischemic regions.
CONCLUSIONLow-dose dobutamine (5 micro g.kg(-1).min(-1)) recovers hypokinetic or akinetic myocardial regions, and dobutamine stress MRI can be used to detect myocardial viability.
