1.Research on experiential avoidance, cognitive fusion and type D personality of pregnant

Objective To explore the relationship among experiential avoidance,cognitive fusion and type D personality in pregnant women.Methods 309 pregnant women were assessed with self-made general information questionnaire,acceptance and action questionnaire-2nd edition (AAQ-Ⅱ),cognitive fusion questionnaire(CFQ) and type D personality scale(DS14).Results Average score of AAQ-Ⅱ and CFQ of 309 pregnant women were (16.41±7.55) and (24.24± 10.90),respectively.According to the type D personality questionnaire scores,the subjects were divided into four groups.Average score of each group on AAQ-Ⅱ and CFQ was respectively (21.25 ± 8.18) and (31.12± 10.99) for type D personality group (Group 1),(19.80±6.14) and (27.67±9.76) for negative emotion group(Group 2),(15.12±6.40) and (22.16±9.56)for social inhibition group (group 3) and (14.02±6.61) and (21.18±10.03) for group without negative emotions and social inhibition (group 4).The differences between groups were statistically significant (F=19.44,17.372,all P<0.01).Correlation analysis revealed that AAQ-Ⅱ and CFQ were positively related to NA and SI (r=0.484,0.251,0.477,0.302,P<0.05).NA and SI all entered the AAQ-Ⅱ regression equation and CFQ regression equation respectively:Y1 =4.655+ 1.032X1 + 0.836X2,Y2 =8.013+ 1.233X1 + 0.745X2 (Y1 as AAQ-Ⅱ,Y2 as CFQ,X1 as NA,X2 as SI).Conclusion Pregnant women with type D personality have the worst psychological flexibility.The two demensions of type D personality are predictive to experiential avoidance and cognitive fusion.
2.Anti-cancer mechanism of bufalin

Bufalin is one of the major active components of Chan Su,a traditional Chinese medicine.Studies at home and abroad have confirmed that bufalin can inhibit cell proliferation significantly in many human leukemia and solid cancer cell lines by inhibiting endothelial hyperplasia and angiogenesis,and inducing cell differentiation and apoptosis. With the deepening of study,it is showed that bufalin has extensive anticancer activities and it has potential clinical value with very low drug concentration.
3.Effect of pretreatment with candesartan on the expression of vascular endothelial growth factor after cerebral ischemia-reperfusion in rats

Objective To investigate the vascular protective effect of pretreatment with candesartan on cerebral ischemia in rats.Methods Forty-eight male Sprague Dawley rats were randomly divided into sham operation,ischemia-reperfusion,low-dose candesartan group was randomly redivided into reperfusion 24 h and 72 h subgroups (n = 6).A model of middle cerebral artery occlusion was induced by intraluminal suture method after 4 weeks of drug gavage.Blood pressure was measured preoperatively.The neurological scores were performed after 24 and 72 hours of reperfusion.Then the rats were decapitated and the brains were removed.The infarct volume was measured using 2,3,5-triphenyltetrazolium chloride staining.The expression of vascular endothelial growth factor (VEGF) protein in ischemic regions was detected by immunohistochemical staining and Western blot analysis.Results The neurological scores of the low-dose and high-dose candesartan groups at 24 and 72 hours after reperfusion were significantly better than those of the ischemia-reperfusion group (P = 0.008and 0.001,respectively),and the infarct volume was reduced significantly (P=0.010 and 0.000,respectively).At the second week after medication,the blood pressure was decreased significantly in the high-dose candesartan group,and there was no significant antihypertensive effect in the low-dose candesartan group.The positive expression of VEGF mainly distributed in the vascular endothelial cells around the infarcts.Its expression was further upregulated with the time.It reached the peak after 72-hour reperfusion.The result of Western blot analysis was consistent with that of immunohistochemical staining.Conclusions Candesartan may reduce the infarct volume by upregulating the expression of VEGF in the ischemic region and improve the neurological score.
4.Reduction of membrane potential in GH3 cells with ghrelin

ObjectiveTo investigate the in vitro effect of ghrelin on membrane membrane potentail of GH3 cells.MethodsNystatin-perforated whole-cell recording configuration was used.ResultsThe membrane potentail of GH3 was decreased from-(72±4.78) mV to(54.8±4.58) mV(P<0.01) after application of ghrelin(10-7~10-8 M)ConclusionGhrelin can transiently and significantly decreased the amplitude of membrane potentail,which may contribute to the ghrelin-stimulated GH secretion from GH3 cells.
5.Research on postpartum depression and experiential avoidance of parturient

Objective To explore the relationship between postpartum depression and experiential avoidance of parturient and to provide empirical evidences for acceptance and commitment therapy in mental treatment of postpartum depression.Methods 309 pregnant women were assessed with self-made general information questionnaire,self-rating depression scale (SDS) and acceptance and action questionnaire-2nd edition (AAQ-Ⅱ) in predelivery periods and 3-7 days after childbirth respectively.Results AAQ-Ⅱ scores in the predelivery or postpartum depression group ((18.54±8.25),(19.07±7.82)) were higher than that in the normal group((15.47±7.03),(14.57±6.57)),and the difference was statistically significant(t=-3.15,-5.07,all P<0.01).It was found that the predelivery or postpartum AAQ-Ⅱ scores were positively associated with the SDS scores of predelivery periods and postpartum periods respectively(r=0.34,0.34,0.24,0.42,all P<0.01).Hierarchical multiple regression analyses were then conducted.In the first block,neonatal exceptional conditions and the predelivery SDS significantly explained variance in postpartum depression(β=0.09,0.62,all P<0.01).In the second block,the predelivery AAQ-Ⅱ still had a significant effect on postpartum depression (β=0.13,P=0.006,△R2=0.01,P=0.006),despite control the age of parturient women,postpartum complication,neonatal exceptional conditions and the predelivery SDS.Conclusion A function to predict the occurrence of maternal postpartum depression is obtained from the experiential avoidance which is attributed to the risk of maternal postpartum depression.
6.Analysis of ER-α36 expression in 653 cases of breast cancer tissues

Objective To study the expression status of estrogen receptor-alpha36(ER-α36)in breast cancer tissue and its rela-tionships with the occurrence,development and clinical prognosis of breast cancer.Methods 653 cases of breast cancer tissues were selected in this study.The real-time reverse transcription polymerase chain reaction(RT-PCR)and immunohistochemistry were used to detect the expression of ER-α36,estrogen receptor-alpha66 (ER-α66),progesterone receptor(PR)and human epidermal growth factor receptor-2(Her-2).The relationships between the expression of ER-α36,ER-α66,PR and Her-2 and the pathological charac-ter were analyzed.Results The expression rate of ER-α36 in all cases was 40%.The expression rate of ER-α36 in Her-2 positive tissues(63%)was significantly higher than that in the Her-2 negative group(44%,P <0.05).The expression rate of ER-α36 in ER-α66/PR/Her-2 negative tissues(66%)was significantly higher than that in the non-three-negative group(35%,P <0.05).The differences of ER-α36 expression rate between ER-α66 positive samples and negative samples or between PR positive and negative samples showed no statistical significance(P >0.05).The expression rate of ER-α36 in stage Ⅲ+Ⅳ breast cancer tissues(54%) was significantly higher than that in stage Ⅰ+Ⅱ breast cancer tissues(28%,P <0.05).The expression rate of ER-α36 in breast cancer tissues with lymph node metastasis (55%)was significantly higher than that in breast cancer tissues without lymph node me-tastasis (23%,P <0.05).Conclusion The results indicate that ER-α36 may play a very important role in the occurrence,develop-ment and lymph node metastasis of breast cancer,and be associated with the expression of Her-2,breast cancer staging and lymph node metastasis.ER-α36 is expected to become a new tumor marker and clinical diagnosis and treatment target.
7.Effects of decreasing lipidemia by mappianthus iodoies flavone in SD rats

Objective To explore effect of decreasing blood lipid by mappianthus iodoies flavone in rats model.Methods We selected sixty healthy male SD rats,which were randomly divided into six groups:normal control group (NC),model control group (MC),simvastatin group positive control group (PC),low dose group of mappianthus iodoies flavonoids (MIF1 group),middle dose group of mappianthus iodoies flavonoids (MIF2 group),high dose group of mappianthus iodoies flavonoids (MIF3 group).After 6 weeks,absolute diet 12 h,the rats of blood samples were drawn from orbit and it was collected to detect serum total cholesterol (TC),serum triglyceride(TG),low density lipoprotein cholesterol (LDL-C),high density lipoprotein cholesterol (HDL-C).After 12 weeks,absolute diet 12 h,get blood from heart,serum,centrifugal separation measure serum SOD,MDA content,test the content of T-AOC.Results Mappianthus iodoies flavonoids can decrease the level of TG,TC,LDL-C,MDA and improve HDL-C,SOD and T-AOC in lipid of lipidemia rats.Especially compared with the NC group,the level of TG,TC,LDL-C,MDA in MC group were in creased,and HDL-C,SOD,T-AOC level were decreased in rats fed high fat diet(P<0.05),meanwhile the levels of TC,TG and LDL C in the treatment groups were decreased,and HDL-C levels were increased(P<0.05);MIF group,which are compared with MC,the level of TC,TG,LDL-C and MDA weresignificantly decreased and the level of HDL-C,SOD,T-AOCwere increased significantly(P<0.05).Conclusion Mappianthus iodoies flavone may improve the level of SOD,T-AOC and decrease the level of MDA to decrease lipid.
8.Differential effects of Ach and ATP on [Ca2+]c changes in mouse pancreatic β-cells

ObjectiveTo identify the differences between acetylcholine(Ach)-induced increase and adenosine triphosphate(ATP)-induced increase in cytosolic free Ca2+ concentration ([Ca2+]c) in mouse pancreatic β-cells. MethodsMouse pancreatic β-cells were primarily cultured and divided into two groups,one group was stimulated by Ach and another by ATP.[Ca2+]c was recorded with Fura-2 in normal condition, chelation of extracellular Ca2+ by EGTA and depletion of intracellular Ca2+ stores by Thapsigargin.ResultsAch induced a transient peak increase and sustained increase in [Ca2+]c. ATP induced a transient peak increase and no sustained increase. Chelation of extracellular Ca2+ by EGTA eliminated the sustained increase induced by Ach, and did not eliminate the transient peak increase induced by Ach and ATP. Depletion of intracellular Ca2+ stores by Thapsigargin eliminated the transient peak increase induced by Ach and ATP and the sustained increase induced by Ach. ConclusionsAch induces intracellular Ca2+ release and the following Ca2+ release-activated Ca2+ influx, and ATP induces intracellular Ca2+ release, but blocks the following Ca2+ release-activated Ca2+ influx.
9.The inhibitory effect of tamoxifen on the proliferation of the anterior pituitary cell of rats

AIM To investigate the effect of tamoxifen on the proliferation of the anterior pituitary cell of rats and its mechanism. METHODS Primary culture of the anterior pituitary cell of rats and 3H TdR incorporation method were applied. The changes of cell morphology were observed directly by electric microscope. RESULTS Tamoxifen could inhibit the proliferation of the anterior pituitary cell of rats. The inhibitory effect of tamoxifen (0 1 ?mol?L -1 ) could be reversed by estrogen.The classical apoptotic changes appeared in the cells after tamoxifen incubation for 48 h. CONCLUSION Tamoxifen can inhibit the proliferation of the anterior pituitary cell of rats and resultin the cell apoptosis.
10.Damage of viability of insulin-secreting cells by cholesterol in vitro

ObjectiveTo identify the influence of cholesterol on insulin-secreting cells.Methods MIN6 mouse insulin cells were cultured in vitro. After confluence of cells, on one hand, free cholesterol in series concentrations was added to cultu re medium to act for 24 hours, and on the other hand, free cholesterol in same c oncentration was added to culture medium to act for different periods. MTT test was used to evaluate the viability of MIN6 cells.Results25μmol/L free cholesterol significantly decreased the viability of MIN6 c ells after 24 hours and 100μmol/L free cholesterol significantly decreased the viability of MIN6 cells after 12 hours and induced cells death after 24 hours.Conclusions Free cholesterol decreases viability of MIN6 cells in a dose-dependent and time-dependent manner, and it is indicated that elevated free cholesterol concentration in blood may be one factor involved in the dysfunction of pancreatic β-cells.
