Objective To investigate the expression and its clinical significance of serum urotensin Ⅱ(UⅡ)and nitric oxide(NO)in chronic cor pulmonale patients in acute exacerbation.Methods Totally 55 cases of chronic cor pulmonale in acute exacerbation(chronic cor pulmonale group)from Respiratory Department of First Affiliated Hospital of Jiamusi University during Nov.2006 to Mar.2007 and 40 healthy volunteers(normal control group)were collected.The levels of serum UⅡ and NO were detected in 55 cases of chronic cor pulmonale in acute exacerbation and 40 normal controls respectively.The condition of everyone's smoking history was collected.Arterial blood gas analysis was made in 37 cases of chronic cor pulmonale in acute exacerbation.The relationship among the levels of serum UⅡ,NO and PaO2,PaCO2,smoking history was observed.Results The levels of serum UⅡ and NO in chronic cor pulmonale in acute exacerbation were significantly higher than those of normal control group(all P

AIM: To investigate the effect of Thymosin ? 1 on the development and matutation of thymocytes. METHODS: The proportion of CD4+CD8+ thymocytes and the expression of smoothened (Smo) of the hedgehog (Hh)-signaling in CD4-CD8-thymocytes were examined to observe the effect of thymosin ? 1 on thymocyte development and matutation. RESULTS: Flowcytometric analysis showed that thymosin ? 1 showed activity at a low dose of 30 ?g/kg, and 30 ?g/kg thymosin ? 1 accelerated the replenishment and maturation of thymocytes according to the expression of Smo of the Hh-signaling in CD4-CD8-thymocytes, the potent negative regulator of proliferative responses. CONCLUSION: Thymosin ? 1 can accelerate thymocyte development from CD4-CD8- to CD4+CD8+.
3.Therapeutic evaluation of retrievable esophageal covered stent in treating achalasia

Objective To evaluate the clinical effect of retrievable esophageal covered metal internal stent in treating patients with achalasia.Methods Under DSA guidance,peroral"Z-type"double horn covered metal internal stent implantation was performed in 16 patients with achalasia.Esophagography was carried out about 28 days after the procedure and the stent was retrieved.Results Of 16 cases,the stent fell off into the stomach two weeks after the operation in one.And the stent was successfully replaced after it was taken out.The placed stent was successfully retrieved in all cases 28 days after the treatment.No serious complications occurred.All the patients were followed up for 3 months to 3 years.During the follow-up period restenosis of the esophagus developed in two cases(at one and 1.5 years respectively),and the restenosis degree was relived after balloon dilation.Clinically,no esophageal symptoms,such as dysphagia,occurred in all patients.Conclusion As a simple and safe technique,the retrievable esophageal covered metal internal stent implantation is very effective with fewer complications for the treatment of achalasia.Moreover,the technique carries lower restenosis occurrence.
4.Therapeutic Observation of Auricular Point Sticking with Magnetic Bead for Insomnia in Hepatocirrhosis

Objective To observe the clinical efficacy of auricular point sticking with magnetic bead in treating insomnia in hepatocirrhosis patients. Method Ninety patients with hepatocirrhosis complicated with insomnia were randomized into a treatment group and a control group, 45 in each group. The treatment group was intervened by auricular point sticking with magnetic bead, while the control group was by auricular point sticking with medical adhesive tape. After successive 2-week treatments, the Insomnia Severity Index (ISI) and the Evaluation Criteria of Therapeutic Efficacy for Mental Disorders were observed for evaluating the treatment result. Result The ISI was significantly improved in the treatment group after intervention (P<0.05), and the improvement was significantly different from that in the control group (P<0.05). The total effective rate was 86.7% in the treatment group versus 60.0% in the control group, and the difference was statistically significant (P<0.01). Conclusion Auricular point sticking with magnetic bead is easy-to-operate and can produce a satisfactory efficacy in treating insomnia of hepatocirrhosis patients.
5.Effect of lead acetate on the apoptosis and the expression of bcl-2 and bax genes in rat brain cells.

OBJECTIVESTo explore the effect of lead acetate on the apoptosis of rat brain neural cells and the relationship between the apoptosis and the bcl-2 as well as bax gene expression.
METHODSLead acetate was given to SD rats by intraperitoneal injection for 5 days at the dosage of 25, 50 and l00 mg/kg body weight respectively. The rates of apoptosis and the expression of bcl-2 (Bcl-2) and bax (Bax) in neural cells from cerebral cortex, hippocampus and carebellum were measured respectively by flow cytometry (FCM).
RESULTSThe rates of apoptosis in neural cells from cerebral cortex, hippocampus and cerebellum in every treatment group were significantly higher than that of control (P < 0.01), and there was a significant dose-response relationship (r = 0.998, 0.989 and 0.997 respectively). The expression of bcl-2 was significantly decreased, whereas bax was significantly increased, in neural cells from cerebral cortex, hippocampus and cerebellum in every lead acetate treatment group (FI) compared with the control group, and there was a significant dose-response relationship (r = -0.886, -0.787 and -0.832 respectively for bcl-2, r = 0.971, 0.988 and 0.991 respectively for bax). The value of Bcl-2/Bax in every treatment group decreased significantly compared with control, and there was a nice dose-response relationship (r = -0.863, -0.829 and -0.999, respectively). Correlation analysis showed that rates of apoptosis were inversely correlated with the expression of bcl-2 (r = -0.750, -0.509, and -0.667, respectively), whereas positively correlated with the expression of bax (r = 0.748, 0.56l, and 0.668, respectively). And there were inverse correlations between the rates of apoptosis and Bcl-2/Bax expression.
CONCLUSIONLead may induce apoptosis in rat brain neural cells through the down regulation of bcl-2 and the up regulation of bax gene expression.

6.Relationship between methylenetetrahydrofolate reductase gene 677C / T, 1298A / C SNP and lung cancer

Objective To explore the relationship between MTHFR gene polymorphism and lung cancer in Han population of Heilongjiang Province.Methods Two hundred and twenty-five lung cancer patients were selected as the experimental group and the healthy subjects in the outpatient physical examination as the control group,A case-control study was used to analyze the association of MTHFR gene 677C/T,1298A/C SNP and lung cancer,and the gene typing was detected by Sanger double deoxidization chain termination method.Results In the control group,the frequencies of wild-type CC,mutant heterozygote CT,and homozygous TT genotypes of the MTHFR gene C677T were 34.2%,55.1%,and 10.7%,respectively.The frequencies of the three genotypes in the experimental group were 26.7%,50.2%,23.1%,respectively.The difference in the distribution of C677T SNP genotype frequencies of the experimental group and the control group was statistically significant (P =0.002),in which the mutation homozygous TT carriers were 2.78 times more likely to develop lung cancer than wild-type CC (OR(95%CI):2.78 (1.54 ~ 5.02),P =0.001);AA,AC and CC genotype frequencies ofthe A1298C locus of the MTHFR gene were 34.2%,55.1%,and 10.7%,respectively,and the control group was 64.0%.32.0%,4.0%,there was no significant difference between the two groups (P =0.247).The frequencies of AA,AC and CC genotypes in the A1298C locus of the MTHFR gene were 34.2%,55.1%,and 10.7%,respectively,and 64.0.%,32.0%,and 4.0% in the control group,respectively.There was no significant difference between the two groups (P).=0.247).The haploid analysis showed that the distribution frequency of TA haplotype in the experimental group was significantly higher than that in the control group (43.1% vs.35.3%).There was a statistically significant difference between the two groups (OR(95%CI):1.39 (1.06-1.81),P=0.016);while the frequencies of CC haplotypes in the experimental group and the control group were 10.6% and 17.1%,respectively.The difference was statistically significant (OR(95%CI):0.58 (0.39-0.85).P=0.005).There was a linkage disequilibrium between the two points of MTHFR gene 677 and 1298 (D'=0.48,P=0.003).The gene-environment interaction analysis of the MTHFR gene C677T polymorphism showed that based on the comparison between TT and CC genotype,age over 60 (OR(95%CI):4.0(1.78-9.32),P =0.001),male (OR (95%CI):5.55 (2.10-14.67),P=0.000),smoking (OR(95%CI):8.13 (2.29-28.85),P=0.000) and small cell lung cancer (OR (95%CI):1.28 (1.10-1.44),P=0.000) can increase the risk of lung cancer;based on the comparison between CT and CC genotype,women (OR(95%CI):2.09 (1.05-4.16),P=0.030),non-smoking population (OR(95%CI):2.43 (1.25-4.74),P=0.008) and small cell lung cancer (OR (95% CI):0.31 (1.16-1.59),P =0.000) can increase the risk of lung cancer.Conclusion MTHFR gene C677T is a genetic susceptibility gene for lung cancer and is associated with the risk of lung cancer.
7.Research progress of PD-1/PD-L1 inhibitors in renal cell carcinoma

Renal cell carcinoma is a malignant tumor originating from the renal parenchyma. Different subtypes of renal cell carcinoma have different pathological features and molecular mechanisms. Because radiotherapy and chemotherapy are difficult to achieve ideal therapeutic effect on renal cell carcinoma, targeted therapy based on vascular endothelial growth factor and mammalian target of rapamycin will eventually in the case of drug resistance, it is currently necessary to explore a new way to improve the treatment status of metastatic renal cell carcinoma. With the continuous research and exploration of programmed death-1 (PD-1)/programmed death-ligand-1 (PD-L1) inhibitors, immunotherapy certain effects have been shown in metastatic renal cell carcinoma. PD-1/PD-L1 inhibitors have considerable prospects for single-agent or other drug treatment in renal cancer, and the long-term adverse reaction rate of this drug is low. At present, there are studies that treat PD-1 or PD-L1 inhibitors in combination with radiochemotherapy, anti-tumor drugs and surgery, bringing new hope to patients who are not sensitive to radiochemotherapy, are resistant to drugs, and have lost surgery. While PD-1 or PD-L1 also significantly improves overall survival in monotherapy. This article reviews and summarizes the application and research progress of PD-1/PD-L1 inhibitors in renal cell carcinoma in recent years, and reviews them.
8.Application of absorbable barbed suture in closure of epicranial aponeuroisis

Objective To clarify the value of absorbable barbed suture in closure of galeal. Methods A total of 101 patients had craniotomy treated in Shengjing Hospital of China Medical University from October 2018 to February 2019 were divided into two groups according to the admission date. In the barbed suture group, 45 patients were sutured with QUILL by continuous stitching. In the control group, 56 patients were sutured with traditional stitchingby intermittent suture. Compare the differences in suture speed, average postoperative hospital stay, incision complication rate, and the average hospital costs of the two groups. Results The suture speed in barbed suture group was (0.330 ± 0.012) cm/min , and was significantly faster than that in control group (0.540 ± 0.016) cm/min;the postoperative average hospitalization days in barbed suture group was (10.91 ± 0.62) d, and was significantly shorter than that in control group (12.73 ± 0.41) d, there were significant differences (P<0.05) . However, the complications and hospital costs in two groups had no significant differences (P>0.05). Conclusions The use of absorbable barbed close epicranial aponeuroisiscan improve suture speed, shorten the postoperative average hospitalization days, which is worthy of promotion.
9.Inhibitory effects of Beclin 1 gene expression by RNA interference on the proliferation and apoptosis in fibroblasts of naked mole rat

Objective To investigate the effects of down-regulation of Beclin 1, which is an autophagy regulatory molecule, expression induced by RNA interference on the proliferation and apoptosis in skin fibroblasts of naked mole rat. Methods The expression levels of Beclin 1 were detected after starvation or H2 O2 treatment.The fibroblasts were transi-ently transfected with specific siRNA targeting Beclin 1 and then screened by real-time PCR and Western blot.Cell prolifer-ation and apoptosis were determined using CCK-8 detection kit and flow cytometry ( FCM ) .The expressions of related genes were detected by Western blot.Results The expression of Beclin 1 gene at mRNA and protein levels was signifi-cantly lower in fibroblasts of the naked mole rat.Starvation and H2 O2 treatment induced changes of the Beclin 1 expression. Inhibition of Beclin 1 gene expression can inhibit cell proliferation and induce early and late apoptosis.The protein levels of p53, BAX, Bcl2, LC3B, p-AKT and mTOR were reduced after transient transfection with Beclin 1-siRNA.Conclusions The expression of Beclin 1 in fibroblasts of naked mole rat are changed in response to starvation or H2 O2 stimulation.Inhi-bition of Beclin 1 gene expression can inhibit cell proliferation and induce apoptosis.Therefore, Beclin1 gene may play a regulatory role in autophagy, proliferation and apoptosis in the skin fibroblasts of naked mole rat.
10.The expression and significance of serum IL-27 and TGF-β1 in patients with pulmonary fibrosis

Objective To investigate the expression and clinical significance of IL-27 and TGF-β1 in patients with pulmonary fibrosis. Methods The expression levels of IL-27 and LTB4 were determined by ELISA method in 20 cases of pulmonary fibrosis patients and 15 cases of healthy control group. Results(1)The levels of IL-27 and TGF-β1 in pul-monary fibrosis patients were higher than the control group;IL-27 and TGF-β1 levels were gradually increased from the control group, pulmonary fibrosis mild hypoxia group, moderate hypoxia group, severe hypoxia group, the differences were statistically significant (F=21.58, P<0.001; F=18.75, P<0.001);(2)The serum IL-27 and TGF-β1 levels were significantly reduced with the increase of PaO2, the linear correlation was found between the two groups and PaO2 were negatively correlated(r=-0.754,P<0.05;r=-0.793,P<0.05);It was positively correlated between IL-27 and TGF-β1(r=0.854,P<0.05). Conclusion IL-27 and TGF-β1 were being involved in the pathogenesis of pulmonary fibrosis, detec-tion simultaneously, there were important significances for pulmonary fibrosis clinic diagnosis and therapy.
