1.Effect of recipe for kidney tonification on pituitary-thyroid axis and bone density in rat with experimental osteoporosis

BACKGROUND: In the research, the recipe for kidney tonification is prepared into "topical form" and "oral form". According to the interrelationship of "acupoints-meridans and collaterals-internal organs-target organs", osteoporosis is treated by the approaches of point compress and oral application respectively. According to the traditional theory of Chinese medicine, it is to infer that the herbs bring the adjustment into play by"being attributive to kidney meridian", but,whether does the specialty of herbs on "targeting medication" happenpractically or not? Hormones in "hypothalamus-pituitary- target gland" system is assayed to further analyze estrogen receptor and androgen receptor of target organs,such as kidney,bone, uterus, thyroid gland and testis so as to observe the relativity between "tropism and receptor".DESIGN: Complete random design with the objects of osteoporosis rats, and the control experimental study with kidney tonification recipe administrate with different approaches.OBJECTIVE: To probe into the effect of kidney tonification recipe on pituitary-thyroid axis in experimental osteoporosis.SETTING: College of Traditional Chinese Medicine of Hebei Medical University.MATERIALS: The experiment was performed in Chinese-Western integrated Basic Experiment Room of Hebei Medical University from January 2000 to December 2004. Sixty SD healthy female rats of 3-month old were employed,weighted(300 ± 20) g. The experimental animals were provided by Hebei Experimental Animal Center. After bred routinely for 1 week in Experimental Room, the rats were randomized into 6 groups,namely normal control(10rats), pathological model group(10 rats), infusing group(10 rats) in which kidney tonification recipe was applied based on 0. 8 g/100 g body mass,acupoint paste on bladder meridian group (10 rats), acupoint paste on kidney meridian group(10 rats) and paste on non-meridian group(10 rats). METHODS: Osteoporosis model was prepared with injection of dexamethasonc. Antagonism osteoporosis acupoint paste(AOAP)on acupoints and non-neridian places was administrated to compare with oral recipe for kidney tonification in the treatment of osteoporosis. It took the changes in serum estrogen, bone density, calcitonin/parathyroid hormone(CT/PTH), thyrotropic hormone(TSH), freeT3andfreeT4asthemeasuringindexes. Immunocyto-chemistry stain and morphological measurement of TSH cell,follicular adenoma of thyroid gland(C cell) and thyroid globulin(Tg) were applied in pituitary gland and thyroid gland to observe the therapeutic results.MAIN OUTCOME MEASURES: ① Adjustment of the recipe for kidney tonification on thyroid function. ② Adjustment of the recipe for kidney tonification on bone density.RESULTS: AOAP improved the levels of estrogen, CT/PTH, TSH, free T3and free T4, increased bone density. The counts of TSH cell, C cell and Tg positive reaction substance were increased and they were colored deeply.CONCLUSION: AOPA, by "multiple relevant convergence reaction", adjusts and affects systematically the endocrinal function of bone metabolism,improves the levels of estrogen and CT/PTH, reverses the declining tendency of pituitary-thyroid axis function and improves thyroid function so as to weaken osteocyte in the bony absorption, enhance osteoblast in osteogenesis and increase bone density.
2.Effect of Simiaoyongan Decoction with Shengmai Decoction on Apoptosis of Hypoxia/Reoxygenation Neonatal Rat Cardiomyocytes

Objective To observe the effect of Simiaoyongan decoction with Shengmai decoction on apoptosis of hypoxia/reoxygenation model of primary cultured neonatal rat cardiomyocytes. Methods The combination of trypsin and collagenase was used to isolate neonatal rat myocardial cells. Hypoxia/reoxygenation model of cardiomyocytes was made by high purity nitrogen gas. Primary cultured neonatal rat cardiomyocytes were randomly divided into control group, model group and treatment group (treated with Simiaoyongan decoction and Shengmai decoction). The myocardial apoptotic rates were detected by Annexin V-FITC/PI double-staining method. Results Compared with the control group, the apoptotic rate of model group and treatment group was high (P<0.05). Drug intervention lowered the apoptotic rate compared with the model group, the difference was significant (P <0.05). Conclusion The alcohol sedimentation solution of Simiaoyongan decoction with Shengmai decoction played a protective role on hypoxia/reoxygenation cardiomyocytes.
3.Situation of school toilets in rural areas of Laibin City in 2018

Objective:
To understand the sanitary condition of rural school toilets in Laibin City, and to provide scientific basis for the government and education departments to improve the sanitary condition of rural school toilets and to rebuild and build new harmless sanitary toilets.
Methods:
The sanitary condition of toilets and feces treatment of the rural areas of 6 counties of Laibin City were thoroughly investigated through file review, on-site inspection and interviews.
Results:
There were 36 schools without toilets, all of which were primary schools. Those schools distributed in Jinxiu County (31), accounting for 35.63% in its primary schools(31/87). Among the 651 schools with toilets, there were 1 559 sanitary toilets and 90 unsanitary toilets (all distributed in primary schools). and the percentage of unsanitary toilets in primary schools was that Jinxiu County 44.64% and Heshan City 31.25%. The proportion of sanitary toilets in teaching buildings, offices or dormitory buildings in primary schools, middle schools and nine-year schools was 77.36%, 94.20% and 30.95%, respectively. Hierarchical analysis found that outdoor toilets accounted for the largest proportion of middle schools in Xiangzhou County (72.46%), followed by nine-year schools in Xingbin District (71.79%). Among sanitary toilets, the proportion of harmless sanitary toilets was 95.51%, which was 100% in middle schools and nine-year schools, and 94.30% in primary schools. Four of the six counties (cities and districts) accounted for 100%, and the proportion of primary schools of Xingbin District was lowest, which accounted for 89.79%.
Conclusion
At present, the sanitary condition of toilets in rural schools in Laibin City has been obviously improved, but there are still many schools lack of toilets or sanitary toilets, in some counties (cities, districts), especially in primary schools. Quality of toilet management should be improved while strengthening the construction of harmless sanitary toilets in schools.
4. Establishment and application of whole genome high-throughput sequencing of hepatitis B virus with different concentrations

Objective:
To establish a high-throughput sequencing method for a whole genome in different hepatitis B virus (HBV) concentrations.
Methods:
Two method of amplicon-sequencing and direct sequencing without PCR amplification were used for library construction in the three plasmids, including the low HBV load sample, the moderate HBV load sample and the high HBV load sample. Whole genome sequencing was performed on Illumina MiSeq platform.
Results:
There are significant differences in data yield between the two different library construction method. Only a few reads could be mapped to the HBV genome for direct sequencing. However, three samples were successfully amplified by the nested PCR amplification and amplicon-sequencing showed that all HBV samples had a good coverage and depth, which was not affected by HBV concentration. The alignment rate of HBV genome approached 80%. A total of 27 intra-host single nucleotide variations (iSNVs) were identified and 13 iSNVs were low-frequency mutation in three samples. Compared with the high HBV load sample, mutations in the reverse transcription (RT) region was more easily appeared in the low HBV sample and the moderate HBV load sample.
Conclusions
Integrating nested PCR with high-throughput sequencing to the HBV whole genome sequencing is not only a practical method to detect the infection of HBV with a high-sensitivity and accuracy, but also enables to detect the mutation in the infected patient with low HBV copy number.
5.Analysis of influential factors and clinical value of HIV-1 DNA

Objective:To explore the influencing factors of HIV-1 DNA and its clinical value.Methods:The relationship between HIV-1 DNA and CD4 + T cell count, CD4/CD8, HIV viral load and subtype was analyzed in 304 patients with HIV/AIDS in order to explore the factors affecting HIV-1 DNA and the value of clinical application. Results:There was no statistically significant difference in HIV-1 DNA levels between the different CD4 + T cell level groups (Z=1.194, P>0.05). HIV-1 DNA levels were higher in the CD4/CD8≤0.5 group than in the CD4/CD8>0.5 group (Z=-2.788, P<0.01). HIV-1 DNA levels were higher in the HIV viral load >100 copies/ml group than in the ≤100 copies/ml group (Z=-2.953, P<0.01). HIV-1 DNA levels were higher in those with CD4 + T cell counts ≤200 at diagnosis than in those with CD4 + T cell counts >200 at diagnosis (Z=-2.175, P<0.05). HIV-1 DNA levels were higher in patients with CD4/CD8 ≤0.2 at diagnosis than in those with CD4/CD8 between 0.2 and 0.5, but there was no significant difference between the two groups (Z=-0.893, P>0.05). HIV-1 DNA levels were higher in both groups than in the CD4/CD8≥0.5 group (Z=-2.568, Z=-1.960, P<0.05). Higher HIV-1 DNA levels were found in people with an HIV viral load >100, 000 copies/ml at diagnosis than in people with an HIV viral load ≤100, 000 copies/ml at diagnosis (Z=-3.520, P<0.001). The level of HIV-1 DNA was higher in the CRF01_AE group than in the non-CRF01_AE group, and the difference was statistically significant (Z=-2.848, P<0.01). CD4/CD8 seemed to be a protective factor for HIV-1 DNA>500 copies/ml. (OR=0.214(95%CI: 0.056~0.822, P<0.05) Conclusion:CD4 + T lymphocyte count, CD4/CD8, viral load and subtype are factors that influence HIV-1 DNA levels, while HIV-1 DNA may be informative for immune status assessment and disease progression determination.
6.Progress of Clinical and Pharmacological Mechanisms of Kidney-tonifying Formula for the Treatment of Alzheimer's Disease

Alzheimer's disease(AD) is a common neurodegenerative disease with increasing incidence rate. Up to now,there is no ideal treatment for AD. It has become a public health problem worldwide. Traditional Chinese medicine (TCM) believes that kidney deficiency is the key symptomatic element of deterioration and temporal progression symptoms,accompanied by the AD process. The treatment of tonifying kidneys,supplementing essence and replenishing marrow is the fundamental method for AD in TCM. Clinical studies have shown that kidney-tonifying formula can significantly improve the cognitive function and daily ability of patients with mild and moderate AD and have no obvious adverse reactions. Its mechanism of action may be related to the protection of nerves,reduction of β-amyloid (Aβ) level in the brain,inhibition of inflammatory factors activation and anti-oxidative stress. Besides reviewing the clinical and pharmacological research progress of kidney-tonifying formula for AD,this article also discusses the advantages and shortcomings of kidney-tonifying formula in the prevention and treatment of AD based on TCM theory and modern medical research. The aim of this study is to provide references of kidney nourishing therapy in TCM for the prevention and treatment of neurodegenerative diseases.
